酚类物质与蝴蝶兰褐变关系初探   总被引:1,自引:0,他引:1  
 利用高效液相色谱法对3个褐化程度不同的蝴蝶兰品种A1 (大白花红心) 、B3 (迷你型白花黄心) 和R4 (深红花红心) 进行了9种酚酸定性定量分析, 并研究了以叶片为外植体的初代培养过程中总酚含量的动态变化。结果表明: 绿原酸、邻苯二酚、儿茶酚、咖啡酸及没食子酸、对羟基苯甲酸、香豆酸可能与蝴蝶兰褐变相关, 苯甲酸对蝴蝶兰褐变影响很小; 在褐变过程中, 总酚含量越高, 褐变程度越严重。  相似文献

蝴蝶兰组培中pH和温度对外植体褐化的影响   总被引:14,自引:0,他引:14  
 以蝴蝶兰R4品种叶片为外植体, 研究不同pH 培养基、培养温度对褐化率、多酚氧化酶( PPO) 活性和总酚含量的影响以及褐化率与PPO、总酚之间的相关性。结果表明: 培养基pH 6.5或培养温度20℃时外植体褐化率最低。在相同温度(25℃) 下, PPO活性、总酚含量高低并不与pH值大小成正比, 且褐化率与PPO无显著相关性, pH 5.0时, 褐化率与总酚含量显著相关; 在相同pH (6.0) 下, 温度越高, 褐化率越高, PPO活性越强, 总酚含量越高, 且20℃下, 褐化率与PPO、总酚含量分别达到极显著、显著相关, 30℃下, 褐化率与总酚含量显著相关。  相似文献

为观察鉴定茶树新品系福选9号在重庆市巴南区的适应性及表现,以福鼎大白茶为对照品种,于2002-2004年进行了品比试验。结果表明,福选9号表现出如下特性:(1)春芽萌动特早,较对照提早11~13d,春茶产量较高。(2)育芽能力强,生长势旺盛,秋梢休止期较对照晚5~7d,全年生育期较对照长16~20d,平均单产高20.5%。(3)抗逆性强,耐寒性和抗旱性与对照相当,抗病虫性稍强。(4)发芽整齐度高,适宜机械化作业。(5)制绿茶品质优良,适制性强。  相似文献

AIM: To investigate the roles of nitric oxide/L-arginine (NO/L-Arg) pathway and urotensin-Ⅱ (UⅡ) in the development of pulmonary hypertension induced by chronic hypoxia-hypercapnia in rats.METHODS: Forty male Sprague-Dawley rats were randomly divided into four groups (n=10): normal control group (A), hypoxia-hypercapnia+saline group (B), hypoxia-hypercapnia+L-Arg liposome group (C) and hypoxia-hypercapnia+N-nitro-L-arginine methyl ester (L-NAME) group (D).Contents of UⅡ, UⅡ mRNA and receptor of UⅡ (UT) mRNA in pulmonary arterioles were measured with immunohistochemistry analysis and in situ hybridization, respectively.Change of small pulmonary vascular microstructure was also investigated.RESULTS: (1) The mean pulmonary artery pressure (mPAP) and the weight ratio of right ventricle to left ventricle plus septum [RV/(LV+S)] in B and D groups were all higher than those in A group (respectively, P<0.05), with C group significantly lower than those in B group (respectively, P<0.01).(2) Light microscopy showed that the ratio of vessel wall area to total area (WA/TA) and the media thickness of pulmonary arterioles (PAMT) in B group were higher than those in A group (P<0.05), with C group significantly lower than those in B group.(3) The contents of UⅡ, UⅡ mRNA and UT mRNA in pulmonary arterioles in B and D groups were all higher than those in A group (respectively, P<0.01), while the expression of UⅡ and UⅡ mRNA in C group were lower than those in B group (P<0.01).CONCLUSION: The pathological process of pulmonary hypertension induced by chronic hypoxia-hypercapnia might be related to upregulation of UⅡ located in pulmonary arterioles, which might be partially inhibited by exogenous NO in rats.  相似文献

AIM:To evaluate the relationship between u PA and NF-κB p65 expression and its clinical significance in breast cancer.METHODS:The uPA mRNA was measured by real-time fluorescent quan t itative PCR in 46 cases of breast cancer tissues and their adjacent counterparts .NF-κB p65 were measured using immunohistochemistry.RESULTS:The expression of uPA gene was elevated in 63% of cases ,and there was a strong correlation between NF-κB p65 and uPA expression (r =0.451,P<0.01).In addition,the expression of uPA was associated with num bers of axillary lymph node involvement and tumor size.However,no significance was found in uPA expression in the age,pathological type and distant metastasi s organ classification.CONCLUSION:The uPA gene is highly expressed in most of the brea st cancer,and there is strong correlation to NF-κB p65 expression,number of l ymph nodes involvement and tumor size.  相似文献

AIM: The purpose of this study was to observe the morphological features of neuroendocrine cells (NECs), their proliferation and apoptosis in ovarian epithelial tumors, and to discuss their biological and clinical significance. METHODS: 79 specimens of ovarian epithelial tumor samples were collected, of them 20 benign, 18 boderline, 41 milignant tumors, and 22 normal ovaries were investigated immunohistochemically. Chromogranin A was used to detect NECs and their proliferation and apoptosis were examined by double-label staining of chromogranin A and Ki67 or TUNEL. RESULTS: The positive rate of CgA, distribution and staining intensity in ovarian epithelial tumors were higher than those in normal ovary. NECs showed various shapes with neuronoid protuberances stretching to the neighboring cells or basement membrane. Occasionally, they might touch together. No TUNEL positive coexpression in all NECs was observed by double-label staining, but some NECs were coexpressed with Ki67. CONCLUSION: NECs of ovarian epithelial tumors like cancer cells showed a proliferation, but no apoptosis. Their secretion might promote their neighboring non-NECs to proliferate and prevent them from apoptosis.  相似文献

AIM: To investigate the expression of FIZZ1/RELMα in lung tissue of chronic cigarette smoking rat, and to determine the relationship between airway inflammation and airway hyperresponsiveness. METHODS: Made rat model of chronic cigarette smoking was used. The expression of FIZZ1/RELMα in lung tissue was determined by immuno-histochemistry and in situ hybridization. RESULTS: In control rats, FIZZ1/RELMα protein and mRNA expressions were observed at low levels. In cigarette smoking rats, FIZZ1/RELMα expression increased in all the cells especially in bronchial smooth muscle cells, vascular wall cells and alveolar epithelial cells. CONCLUSION: FIZZ1/RELMα is a secreted peptide specifically expressed in lung. Cigarette smoking induces its upregulation, which possibly contributes to cigarette smoking-induced airway hyperresponsiveness.  相似文献

AIM: To investigate the effects of mitochondrial ATP sensitive potassium (MitoKATP) channel opening on translocation of protein kinase C epsilon (PKCε) and the relationship between the translocation of PKCε and the production of reactive oxygen species. METHODS: The expression of PKCε in cultured adult rat ventricular myocytes was investigated with immunofluorescence and Western blotting techniques. RESULTS: (1) Diazoxide, a selective MitoKATP channel opener, caused a significant translocation to myofibrillar-like structures in cultured adult rat ventricular myocytes. (2) The N-2-mercaptopropionylglycine (MPG), a free radical scavenger, partly inhibited the translocation of PKCε caused by diazoxide. (3) Chelerythrine, a selective protein kinase C (PKC) inhibitor, completely blocked the translocation of PKCε caused by diazoxide. CONCLUSION: Opening of MitoKATP channel might activate PKCε and make it translocation to myofibrillar-like structures. PKCε activation occurs downstream of MitoKATP channel, and might be caused by production of reactive oxygen species after opening of MitoKATP channel.  相似文献
