A monoclonal antibody (mAb) against zearalenone (ZEN) was produced using ZEN-carboxymethoxylamine and -BSA conjugates. Antibody produced by one clone showing a very high binding ability was selected and found to have a higher affinity for ZEN compared to a commerciall ZEN antibody. We developed two direct competitive ELISA systems using the selected antibody (ZEN-coated and anti-ZEN antibody-coated ELISA). Quantitative ranges for the anti-ZEN antibody-coated ELISA and ZEN-coated ELISA were from 25 to 750 ppb and from 12.5 to 100 ppb, respectively. The detection limit of both methods as measured with standard solutions was 10 ppb. The intra-plate and inter-well variation of both ELISAs were less than 10%. The IC50 values for α-zearalenol, β-zearalenol, α-zearalanol, and β-zearalanol compared to ZEN were 108.1, 119.3, 114.1, and 130.3% for the ZEN-coated ELISA. These values were 100.7, 120.7, 121.6, and 151.6% for the anti-ZEN antibody-coated ELISA. According to the anti-ZEN antibody-coated ELISA, the average recovery rates of ZEN from spiked animal feed containing 150 to 600 ng/mL of ZEN ranged from 106.07 to 123.00% with 0.93 to 2.28% coefficients of variation. Ketamine has been reported to decrease the immune functions of phagocytes. Previously, we observed that the phagocytic capacity and oxidative burst activity (OBA) of canine peripheral blood polymorphonuclear cells (PMNs) were inhibited by the supernatant from canine peripheral blood mononuclear cells (PBMCs) cultures treated with ketamine. In the present study, we examined whether in vitro treatment with ketamine modulates prostaglandin E2 (PGE2) production in PBMCs. Treatment with ketamine or with ketamine-treated PBMCs culture supernatant simultaneously decreased the phagocytic capacity and OBA of PMNs. Ketamine increased PGE2 production by PBMCs. Recombinant PGE2 decreased the phagocytic capacity and OBA of PMNs. AH-6809, an E-prostanoid 2 (EP2) antagonist, restored the phagocytic capacity and OBA of PMNs, decreased by either the ketamine-treated PBMCs culture supernatant or recombinant PGE2. These results suggest that ketamine inhibits the phagocytic responses of canine PMNs, and that this results from the increase in PGE2 produced by canine PBMCs. 相似文献

1. The effect of the ligation of the caeca on nitrogen utilisation and nitrogen excretion was examined in conventional chickens fed a diet containing 50 g protein/kg plus urea.
2. Ligation of the caeca significantly improved nitrogen balance and utilisation by up to more than 2 times as much as those of controls (P< 0.05).
3. The treatment significantly decreased uric acid excretion by 77 mg nitrogen/day (P< 0.01) and also total nitrogen excretion (P<0.05): the former decrease almost explained the latter.
4. No effect of the ligation of caeca on urea and ammonia excretion was observed.

The chemical investigation of the marine sponge Dysidea sp., which was collected from Bohol province in the Philippines, resulted in the identification of 15 new scalarane-type sesterterpenoids (1–14, 16), together with 15 known compounds. The chemical structures of the new compounds were elucidated based on NMR spectroscopy and HRMS. The structure of 12-epi-phyllactone D/E (15) isolated during this study was originally identified in 2007. However, careful inspection of our experimental 13C NMR spectrum revealed considerable discrepancies with the reported data at C-9, C-12, C-14, and C-23, leading to the correction of the reported compound to the C-12 epimer of 15, phyllactone D/E. The biological properties of compounds 1–16 were evaluated using the MDA-MB-231 cancer cell line. Compound 7, which bears a pentenone E-ring, exhibits significant cytotoxicity with a GI50 value of 4.21 μM. 相似文献

We studied population changes of the field mouse,Apodemus speciosus Temminck, by live trapping in a mixed stand of cypress and broad-leaved trees at Tama Forest Science Garden during the period
from October 1991 to March 1994. We also used radiotelemetry to investigate acorn hoarding. The number of mice captured in
1992 was three to ten times higher than that in either 1991 or 1993. The home ranges of resident mice overlapped greatly,
but a path and differences in the vegetation structure may have affected the home range of individuals. From June 1992 to
April 1993, mice actively transported and hoarded acrons. Individuals hoarded 70% of the acorns in their home ranges within
one day after release. This species was a typical scatter-hoarder and hoarded a single acorn at each cache site. Mice buried
acrons in the soil at the first hoarding, but recovered and carried them into their nests later. Although more than 50% of
the acorns were buried 0–5 cm deep, where the condition for germination seemed to be good, mice recovered and ate all cached
acorns. Mice relocated acorns farther from the original food station as they repeated hoarding. Mice transported acorns an
average of about 15–18 m (range: 2.2–49.8 m) before they ate them. From May 1993 to March 1994, the hoarding behavior of mice
was not active. Most acorns were left at food stations or cache sites for more than three months. Acorn hoarding byA. speciosus probably contributes little to the dispersal and regeneration of acorn-producing trees in years with a high rodent density. 相似文献

Morphological variation of the skull was examined in the northern treeshrew (Tupaia belangeri) from various localities across Southeast Asia. Through a multivariate analysis, the treeshrews from South Vietnam exhibited distinct morphological characteristics compared to other populations from Thailand and Laos, and Malaysia. The plots of the specimens of North Vietnam are not randomly mixed with Thailand plots segregation in the scatteregrams of canonical discriminant analysis. Since the skulls of the population from North Vietnam were morphologically similar to those form central Laos and northern and northeastern Thailand, the zoogeographical barrier effect of Mekong River was not clearly confirmed. The population of the Kanchanaburi in western Thailand is clearly smaller in size compared to the other populations. The southern border of the distribution of this species is determined by the Isthmus of Kra or Kangar-Pattani Line. In the northern treeshrew, which is distributed from southern China to Bangladesh and southern Thailand, we have detected osteometrical geographical variation driven by geography. These results indicate that the skull morphology in the Tupaia glis-belangeri complex distinctively differs in South Vietnam, western Thailand, and southern Thailand. The zoogeographical barrier and factor separating these districts are expected to clarify in the future. 相似文献

Genetic transformation is an important technique for functional genomics study and genetic improvement of plants. Until now, Agrobacterium-mediated transformation methods using cotyledon as explants has been the major approach for cucumber, and its frequency has been up to 23%. For example, significantly enhancement of the transformation efficiency of this plant species was achieved from the cotyledon explants of the cultivar Poinsett 76 infected by Agrobacterium strains EHA105 with efficient positive selection system in lots of experiments. This review is to summarize some key factors influencing cucumber regeneration and genetic transformation, including target genes, selection systems and the ways of transgene introduction, and then to put forward some strategies for the increasing of cucumber transformation efficiency. In the future, it is high possible for cucumber to be potential bioreactor to produce vaccine and biomaterials for human beings. 相似文献

Non-destructive and mathematical approaches of modeling can be very convenient and useful for plant growth estimation. To predict individual leaf area, fresh weight, and dry weight of a cucumber (Cucumis sativus L.), models were developed using leaf length, leaf width, SPAD value, and different combinations of these variables. Eight regression equations, commonly used for developing growth models, were compared for accuracy and adaptability. The three nonlinear models developed were as follows: individual leaf area (LA) = −210.61 + 13.358W + 0.5356LW (R2 = 0.980***), fresh weight (FW) = −2.72 + 0.0135LW + 0.00022LWS (R2 = 0.956***), and dry weight (DW) = 0.25 − 0.00102LS + 0.000077LWS (R2 = 0.956***), where L is the leaf length, W the leaf width, S the SPAD value, and LWS = L × W × S. For validation of the model, estimated values for individual leaf area, fresh weight, and dry weight showed strong agreement with the measured values, respectively. Leaf dry weight, especially, was estimated with a higher degree of accuracy through the use of a SPAD value, as well as leaf length and width. Therefore, it is concluded that models presented herein may be useful for the estimation of the individual leaf area, fresh weight, and dry weight of a cucumber with a high degree of accuracy. 相似文献
