Soybeans contain oil bodies that are coated by a layer of oleosin proteins. In nature, this protein coating protects the oil bodies from environmental stresses and may be utilized by food manufacturers for the same purpose. In this study, oil bodies were extracted from soybean using an aqueous extraction method that involved blending, dispersion (pH 8.6), filtration, and centrifugation steps. The influence of NaCl (0-250 mM), thermal processing (30-90 degrees C, 20 min) and pH (2-8) on the properties and stability of the oil bodies was analyzed using zeta-potential, particle size, and creaming stability measurements. The extracted oil bodies were relatively small ( d 32 approximately 250 nm), and their zeta-potential went from around +12 mV to -20 mV as the pH was increased from 2 to 8, with an isoelectric point around pH 4. Susceptibilities of UK and mainland European samples of Trialeurodes vaporariorum (Westwood) to the neonicotinoid insecticide imidacloprid were investigated over a 7 year period. All 24 strains collected between 1997 and 2003 showed similar baseline levels of susceptibility to that of a known susceptible laboratory strain when exposed to a diagnostic concentration (128 mg L(-1)) of formulated imidacloprid. Two samples collected during 2004, one from the UK and one from The Netherlands, demonstrated reduced susceptibility at this concentration. Using dose-response assays, the presence of resistant individuals was disclosed in both these strains; some individuals were unaffected at doses high enough to induce phytotoxic effects. This report represents the first confirmed cases of neonicotinoid resistance inducing control failures in T. vaporariorum, and highlights a need for careful vigilance to sustain the effectiveness of imidacloprid and related neonicotinoid insecticides.  相似文献

It has been reported previously that the major resistance mechanism to pyrethroid insecticides by the cotton bollworm Helicoverpa armigera (Hübner) in Australia is a consequence of overproduction of esterase isoenzymes. This paper reports structure-activity relationships that support such a view, based on in vivo bioassays conducted with a range of pyrethroid structures containing a variety of acid and alcohol moieties and the correlation with in vitro esterase inhibition assays against the same structures, and identifies the critical regions of the molecule with regard to esterase inhibition, and hence resistance. The implications of this work in terms of possible resistance management are evaluated and discussed.  相似文献

Acute bovine liver disease (ABLD) is a sporadic hepatic disease affecting cattle in southern Australia, characterized histologically by striking periportal hepatocellular necrosis. The cause of ABLD is unknown; however, the seasonality and acute presentation of outbreaks suggest mycotoxin involvement. We describe here the clinical and pathologic findings of ABLD in 45 naturally affected cattle from 13 outbreaks occurring from 2010 to 2019 in Victoria, Australia. Outbreaks occurred in herds located along the southern coastal plain of Victoria and were observed most frequently in lactating dairy cattle. Clinical signs commonly included a combination of mild photosensitization, progressive neurologic signs, and hypogalactia, which preceded death by ≤ 48 h. All affected animals had marked elevations in activities of glutamate dehydrogenase, aspartate aminotransferase, and gamma-glutamyl transferase. At autopsy, the most common lesions were serosal petechiae and/or gastrointestinal hemorrhage, and hepatomegaly with a pronounced hepatic reticular pattern. The principal histologic lesion was widespread—severe periportal hepatocellular coagulative necrosis and erythrocyte pooling—which often extended to massive necrosis. Lesions in other organs were uncommon. Our study of ABLD suggests involvement of a potent hepatotoxin that is either directly cytopathic or requires bioactivation by periportal-specific enzymes.  相似文献

Waste milk samples from 103 farms in England and Wales were examined for the presence of β-lactam antibiotics and ESBL-producing Enterobacteriaceae. Approximately 10 months after the initial sampling, further waste milk, environmental and faecal samples from farms shown to be positive for CTX-M Escherichia coli were investigated further. Isolates with an ESBL phenotype were tested by PCR for the presence of blaCTX-M, blaOXA, blaSHV and blaTEM genes. Isolates positive for blaCTX-M were sequenced to determine CTX-M type. Representative isolates were further examined by PFGE, plasmid replicon typing and serotyping. Of particular interest, 21.4% of waste milk samples contained residues of the cephalosporin cefquinome, which was significantly associated with CTX-M bacteria. Such bacteria occurred in 5.8% of the waste milk samples (including 3.9% CTX-M E. coli). CTX-M types identified were 1, 14, 14b and 15, but none of the E. coli were serotype O25, the serotype of the human pandemic strain.  相似文献

  • 1. The coastal waters surrounding Britain and Ireland became warmer during the 20th century and, according to the UK Climate Impact Programme 2002 scenarios of change and other sources, average annual seawater temperatures may rise a further 2°C or more by the 2050s. This warming is part of a global rise in sea‐ and air‐surface temperatures that will cause changes in the distribution and abundance of species.
  • 2. Initially, there will not be a wholesale movement northwards of southern species or retreat northwards of northern species, because many additional factors will influence the responses of the different organisms. Such factors include the hydrodynamic characteristics of water masses, the presence of hydrographical and geographical barriers to spread and the life history characteristics (reproductive mode, dispersal capability and longevity) of species. Survey data over the past century show how organisms react to changes of the order of 0.5°C, and in the last two decades, when sea temperatures have risen by as much as 1°C, there have been significant local changes in the distribution of intertidal organisms. These past changes provide a clue to more extensive changes expected in the future if global warming develops as predicted.
  • 3. Where species affected by climate change are dominant or key structural or functional species in biotopes, there may be a change in the extent and distribution of those biotopes. Some, dominated by predominantly northern species such as the horse mussel Modiolus modiolus, may decline and reduce their value as rich habitats for marine life. Others, characterized by southern species, for example the sea fan Eunicella verrucosa and the alcyonacean Alcyonium glomeratum, may increase in extent.
  • 4. Using information on the life history characteristics of species, their present distribution and other factors, a key supported by a decision tree has been constructed to identify ‘types’ of organism according to their likely response to temperature rise. Conspicuous and easily identified rocky substratum species are good candidates to track change. Using the key, many species are shown as likely to increase their range northwards significantly. In contrast, fewer will decline in abundance and extent in the north. If, as anticipated, global warming continues, then species with distributions already accurately mapped, or being mapped at present, will provide baseline data to test forecasts.
