Brassica napus L. is one of the most important oilseed crops in the world. Improving oil quality by increasing vitamin E content is a major target of rapeseed breeding. Vitamin E compounds, or tocopherols, are lipid‐soluble antioxidants that are essential nutrients for mammals. In this study, we report the characterization of rapeseed orthologs of the Arabidopsis tocopherol genes VTE1, VTE2 and PDS1. For each gene, at least two homologous sequences were found, and their expression was analysed in different tissues from three rapeseed genotypes. Genetic complementation experiments demonstrated that BnaX.VTE1.b and BnaA.VTE2.b homologs are capable of recovering seed γ‐tocopherol in Arabidopsis mutants. Overexpression of the genes in Arabidopsis Col‐0 shifted the seed tocopherol composition towards higher α‐tocopherol without increasing the total tocopherol content. To address the functionality of BnPDS1 sequences, we performed overexpression tests in Escherichia coli and enzymatic activity analyses. Overall, our results show that the identified sequences from rapeseed are functional orthologs of the Arabidopsis VTE genes and thus have considerable potential as molecular markers for selecting rapeseed with improved seed oil quality.  相似文献

The effects of replacement of sugarcane by spineless cactus (0, 33, 66, and 100 % on dry matter (DM) basis) were evaluated on intake, digestibility of nutrients, ingestive behavior, and performance of finishing lambs. Thirty-six non-castrated Santa Ines lambs at 4 months of age and an initial body weight of 22?±?2.3 kg were assigned in a randomized block design. A quadratic effect was found for intake of DM, organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), and digestible organic matter (DOM), with maximum values of 1.31, 1.19, 0.219, 0.358, and 0.845 g/day estimated with 42.5, 38.5, 38.8, 21.3, and 44.9 % replacement level, respectively. The digestibility of DM, OM, CP, and EE increased linearly; however, there was a quadratic effect on NDF digestibility with maximum value estimated of 500 g/kg with 64.2 % replacement level. Rumination time, total chewing, and rumination efficiency decreased linearly with the replacement levels; however, idle time increased linearly. A quadratic effect was found for final weight, body weight gain, and total weight gain with maximum values estimated of 38.6 kg, 0.232 kg/day, and 16.2 kg with 39.1, 38.2, and 40.4 % replacing levels. The replacement of 49.5 % sugarcane by spineless cactus can be recommended as optimal level, and problems related to intake, digestibility, and performance are concentrated outside that level.  相似文献

By monitoring silage temperature at different locations inside silage stacks, it is possible to detect any significant increases in temperature occurring during silage decomposition. The objectives of this study were: (1) to develop novel noninvasive wireless sensor nodes for measuring the temperature inside silage stacks; (2) to design a suitable sensor protection housing that prevents physical and chemical damage to the sensor; and (3) to mathematically model temperature variations inside a silage stack, using system identification techniques. The designed wireless nodes were used to monitor temperatures in a full-sized silage stack over 53 days. Results showed that the wireless sensor nodes accurately monitored the temperature inside the silage stack at depths of 25 and 50 cm and reliably transmitted the measured data through the network; between 98.9% and 99.4% of the packets disseminated from the sensor nodes were successfully delivered to the gateway. The reliable performance of the network confirmed the correct choice of network characteristics (i.e., frequency range of 433 MHz, a handshaking communication protocol, and 10 mW transmission power). The designed sensor housings were capable of withstanding the high loads that occurred during ensiling, storage, and feed-out. Mathematical models estimating the relations between the silage temperatures (at depths of 25 and 50 cm) and air and soil temperatures were obtained. Black-box modeling using the prediction error method (PEM) was selected as the identification method. Among different black-box models such as ARX, ARMAX, output-error (OE), and Box–Jenkins (BJ), with different model orders, a third-order Box–Jenkins model structure gave the best performance in terms of prediction accuracy. The success rate of the models proposed for silage temperature prediction ranged between 90.0% and 94.3%. Furthermore, there was no significant autocorrelation remaining in the residuals. The results of this study indicate that the designed wireless sensor nodes could potentially be used for detecting silage decomposition processes and improving the efficacy of silage conservation systems.  相似文献

High affinity folate binding protein (HFBP) in porcine serum was purified 2,000-fold to a specific activity of 1.4 nmol of tetrahydrofolic acid (THF) bound per mg of protein, using folic acid (FA) coupled EAH-Sepharose gel affinity chromatography. Binding activity of purified HFBP to folate was examined by ultrafiltration method linked to high-performance liquid chromatography with electrochemical detection or to liquid scintillation counting. Binding affinity of HFBP to folate was characterized by dissociation constants (Kd): 13, 17, and 31 pM for tritiated FA (3HFA), THF, and N5-methyltetrahydrofolic acid (5MF), respectively. FA, THF, and 5MF significantly inhibited binding of HFBP to 3HFA, and according to the magnitude of intensity of the binding inhibition, the order of affinity of each folate was confirmed to be FA > THF > 5MF. Binding activity was rather high and stable for THF and 5MF at pH ranging from 6.0 to 10.0. The binding activity, however, rapidly decreased at pH below 6.0 and over 10.0. No binding activity was observed pH below 3.0 and over 12. Gel filtration analysis showed that the prepared HFBP solution had specific binding activity at around 77 kDa of apparent molecular weight, which was 82 kDa by SDS-PAGE. It is considered that this specific and stable binding enables THF to distribute in porcine plasma.  相似文献

Background Chlamydophila felis, formerly known as Chlamydia psittaci var. felis, is frequently associated with ocular, respiratory, and occasionally reproduction tract infections. Even though the infection is sometimes asymptomatic, it potentially results in a latent immunosuppressive infection.ObjectiveThis study aimed to identify occurrences of feline chlamydophilosis, rarely reported in cats in Indonesia.MethodsThe observation was conducted in three cats with clinical signs of Cp. felis infection, particularly relapsing conjunctivitis. The cats'' histories were recorded based on owners'' information. Conjunctival swabs were sampled for cytology examination and molecular assay detection. A phylogenetic tree was generated using MEGA-X software to reveal group clustering. A post-mortem examination was performed on the cat that died during an examination.ResultsCp. felis was detected in both cytological examination and polymerase chain reaction assay. The phylogenetic tree demonstrated that the Cp. felis isolated in this study clustered with several other isolates from the other countries. Cp. felis can be isolated from cats with different clinical manifestations and levels of severity. The chronic fatal infection demonstrated interstitial broncho-pneumonia under histopathological examination.ConclusionsMolecular assay of Cp. felis is always recommended to obtain a definitive diagnosis of feline chlamydophilosis since the disease can have various clinical manifestations. Even though it may be subclinical and is often not fatal, an infected cat may be a carrier that could spread the pathogen in the surrounding environment. Serious disease management is suggested to avoid high costs associated with regularly relapsing disease.  相似文献

Beet necrotic yellow vein virus (BNYVV) generally has a four-segmented positive-sense RNA genome (RNAs 1–4), but some European and most Asian strains have an additional segment, RNA5. This study examined the effect of RNA5 and RNA3 on different sugar beet cultivars using a Polymyxa-mediated inoculation system under field and laboratory conditions. In field tests, the degree of sugar yield served as an index for assessing the virulence of BNYVV strains. Japanese A-II type isolates without RNA5 caused mostly 15%–90% sugar yield reductions, depending on the susceptibility of sugar beet cultivars, whereas the isolates with RNA5 induced more than 90% yield losses in the seven susceptible cultivars, but small yield losses in one Rz1-resistant and Rizor cultivars. However, a laboratory-produced isolate containing RNA5 but lacking RNA3 caused higher yield losses in Rizor than in susceptible plants, and induced scab-like symptoms on the root surface of both susceptible and resistant plants. In laboratory tests, A-II type isolates without RNA5 had low viral RNA accumulation levels in roots of Rizor and Rz1-resistant plants at early stages of infection, but in the presence of RNA5, viral RNA3 accumulation levels increased remarkably. This increased RNA3 accumulation was not observed in roots of the WB42 accession with the Rz2 gene. In contrast, the presence of RNA3 did not affect RNA5 accumulation levels. Collectively, this study demonstrated that RNA5 is involved in the development of scab-like symptoms and the enhancement of RNA3 accumulation, and suggests these characteristics of RNA5 are associated with Rz1-resistance breaking.  相似文献

Portions of northern Mexico are experiencing a re‐emergence of Rocky Mountain spotted fever (RMSF), a tickborne disease caused by Rickettsia rickettsii, a member of the spotted fever group of rickettsiae (SFGR). Infection with R. rickettsii can result in serious and life‐threatening illness in people and dogs. Canine seroprevalence has been used as a sentinel for human RMSF in previous studies. This study aims to quantify SFGR seroprevalence in canines in three northern Mexican states and identify risk factors associated with seropositivity. A total of 1,136 serum samples and 942 ticks were obtained from dogs participating in government sterilization campaigns and from animal control facilities in 14 Mexican cities in three states. SFGR antibodies were detected using indirect immunofluorescence antibody assays at titre values ≥1/64. Six per cent (69 dogs) showed antibodies to SFGR, with the highest seroprevalence reported in Baja California (12%), Coahuila (4%) and Sonora (4%). Dogs from Baja California had three times higher odds of having SFGR antibodies compared to dogs from Sonora (OR = 3.38, 95% CI, 1.81–6.37). Roughly one quarter (25%) of surveyed dogs were parasitized by ticks (Rhipicephalus sanguineus sensu lato) at the time of sample collection. A portion of collected ticks were tested for rickettsial DNA using polymerase chain reaction. Positive samples were then sequenced, showing evidence of SFGR including R. massiliae, R. parkeri and R. rickettsii. Dogs that spent the majority of time on the street, such as free‐roaming or community‐owned dogs, showed a greater risk of tick infestation, seropositivity, bearing seropositive ticks, and may play a pivotal role in the spread of SFGR among communities. Estimating the seroprevalence of SFGR in the canine population can help public health campaigns target high‐risk communities for interventions to reduce human RMSF cases.  相似文献

Degenerative sacroiliac joint disease is a cause of lumbosacral pain in dogs; however, published information on cross‐sectional imaging characteristics is limited. Objectives of this retrospective, secondary analysis, methods‐comparison study were to test hypotheses that CT lesions reported in humans with degenerative sacroiliac joint disease are also present in dogs, and that CT is comparable to gross pathology for detecting these lesions. Matched CT and gross pathology slice images of 30 sacroiliac joints were retrieved from a previous prospective, canine cadaver study. A veterinary radiologist interpreted randomized CT images for each joint based on previously published CT characteristics of lesions in humans with degenerative sacroiliac joint disease. A veterinary pathologist independently interpreted randomized gross pathology images using the same criteria. All joints had at least one CT lesion consistent with degenerative sacroiliac joint disease. A new CT lesion was also identified and termed “subarticular cleft.” The CT and gross pathology methods agreed for detecting joints with subchondral sclerosis, subchondral erosion, and intra‐articular ankylosis lesions (P > .05, McNemar's test), but disagreed for detection of joints with subchondral cyst, para‐articular ankylosis, and subarticular cleft lesions (P ≤ .05). Using gross pathology as the reference standard, CT had 100% sensitivity for detection of subarticular cleft and subchondral cyst lesions, with 56% and 22% specificity, respectively. Para‐articular ankylosis lesions were detected by CT but not by gross pathology. Findings supported the hypothesis that CT lesions reported in humans with degenerative sacroiliac joint disease are also present in dogs, and partially supported the hypothesis that CT is comparable to gross pathology for detecting joints with these lesions.  相似文献
