The technique of fine-needle biopsy (fine-needle aspiration or fine-needle fenestration) for cytologic evaluation can be extended to many sites beyond the traditional lymph node and skin. Intra-abdominal, intrathoracic, and bone lesions can be easily and rapidly evaluated cytologically. Percutaneous fine-needle aspiration and fine-needle fenestration are useful, accurate, and inexpensive techniques with a rapid turnaround time, and outpatient applicability. For most pets, these minimally invasive techniques do not require anesthesia or analgesia. Although risks are inherent with any invasive procedure, complications are uncommon even with visceral and intrathoracic fine-needle biopsy. Attention to appropriate technique and close patient monitoring minimize the morbidity and improve the diagnostic utility. The low cost, low risk, minimal invasiveness, and high diagnostic yield make fine-needle biopsy particularly attractive to clients. In combination with ultrasound guidance and newer staining techniques, these diagnostic procedures are invaluable to the veterinary clinician.  相似文献

Field studies were conducted at Bushland, Texas, USA, in 2004 to examine usefulness of canopy temperature depression (CTD), the difference of air-canopy temperature, in screening wheat (Triticum aestivum L.) genotypes for yield under dryland and irrigated. Forty winter wheat genotypes were grown under irrigation and dryland. CTDs were recorded after heading between 1 330 and 1 530 h on 6 clear days for dryland and 9 days for irrigation. Drought susceptible index (DSI) for each genotype was calculated using mean yield under dryland and irrigated conditions. Genotypes exhibited great differences in CTD under each environment. The dryland CTDs averaged 1.33℃ ranging from -0.67 to 2.57℃, and the average irrigation CTD were 4.59℃ ranging from 3.21 to 5.62℃. A low yield reduction was observed under dryland conditions relative to irrigated conditions for high-CTD genotypes. CTD values were highly negatively correlated with DSI under dryland, and genotypes of CTDs = 1.3℃ in dryland condition were identified as drought resistant. For 21 genotypes classified as drought resistant by DSI, their CTDs were 1.68℃ for dryland and 4.35℃ for irrigation on average; for 19 genotypes classified as drought susceptible by DSI, average CTD was 0.94℃ in dryland and 4.85℃ in irrigation. The high-yield genotypes consistently had high CTD values, and the low-yield ones had low CTD values for all measurements in dryland. After heading, genotypes maintained consistent ranking for CTD. Regression results for CTD and yield suggested that the best time for taking CTD measurement was 3-4 weeks after heading in irrigation but any time before senescence in dryland. Crop water stress index (CWSI) calculated from CTD data was highly correlated with CWSI calculated from yield, which suggesting traditional costly CWSI measurement may be improved by using portable infrared thermometers. Most importantly, grain yield was highly correlated with CTD under dryland (R^2 = 0.79-0.86) and irrigation (R^2 = 0.46-0.58) conditions. These results clearly indicated grain yield and water stress can be predicted by taking CTD values in field, which can be used by breeding programs as a potential selection criterion for grain yield and drought resistance in wheat, but a second study year is needed to confirm further.  相似文献

Centrosomes organize the bipolar mitotic spindle, and centrosomal defects cause chromosome instability. Protein phosphorylation modulates centrosome function, and we provide a comprehensive map of phosphorylation on intact yeast centrosomes (18 proteins). Mass spectrometry was used to identify 297 phosphorylation sites on centrosomes from different cell cycle stages. We observed different modes of phosphoregulation via specific protein kinases, phosphorylation site clustering, and conserved phosphorylated residues. Mutating all eight cyclin-dependent kinase (Cdk)-directed sites within the core component, Spc42, resulted in lethality and reduced centrosomal assembly. Alternatively, mutation of one conserved Cdk site within γ-tubulin (Tub4-S360D) caused mitotic delay and aberrant anaphase spindle elongation. Our work establishes the extent and complexity of this prominent posttranslational modification in centrosome biology and provides specific examples of phosphorylation control in centrosome function.  相似文献

Identification of quantitative trait loci (QTL) and markers associated with heat and drought tolerance is warranted for marker-assisted selection in wheat (Triticum aestivum L.) breeding programs in areas prone to these abiotic stresses. Our study used a family-based mapping approach in which 19 families consisting of 384 individuals were developed by three-way crosses involving the heat tolerant, tetraploid cultivated emmer (Triticum turgidum L. var dicoccum) genotype IG45069 and ten heat susceptible tetraploid genotypes, IG44999, IG44961, IG45413, IG83047, IG45441, IG127682, IG45448, IG110572, IG88723 and IG54073, in order to detect QTL and markers associated with heat tolerance. The 384 individuals were phenotyped for physiological traits associated with heat tolerance and genotyped by SSR markers. The QTL associated with heat stress tolerance, as measured by chlorophyll content, flag leaf temperature depression (FLTD) and individual kernel weight (IKW) were mapped on chromosomes 1B (QChlc.tamu-1B), 2B (QFlt.tamu-2B), and 5A (QIkw.tamu-5A), respectively, using linkage analysis. Alleles from IG45069 possessed the highest associations with the phenotypic data for the studied traits. This study demonstrates that a family-based mapping approach can be utilized in rapid detection of QTL associated with heat tolerance in wheat based on linkage and association analyses.  相似文献

A combination of survival and spatial analyses was applied to a dataset containing cattle mortalities on smallholder dairy farms in two separate regions (Tanga and Iringa) of Tanzania. Cattle mortality data for the year 1998 were collected retrospectively during the period January to April 1999. The objectives of the current study were, firstly, to quantify the hazard of mortality and, secondly, to investigate whether there was a spatial pattern in the variation that was unaccounted for in the hazard of mortality. A Cox regression model with farm as a frailty term was used to quantify the hazard of mortality. Geostatistical techniques were used to explore the first- and second-order spatial distribution of the farm frailty term. A total of 1,790 cattle from 400 randomly sampled smallholder dairy farms were included in the study. The overall mortality incidence rate for the study was 11.8 per 100 animal years (95% CI 10.0–13.8). The hazard of mortality for male cattle was 2.4 (95% CI 1.7–3.4) times higher than that of female cattle. There was evidence of a first-order spatial pattern, i.e., a tendency for farms with higher frailty to aggregate.  相似文献

Neospora caninum, an apicomplexan protozoan parasite, is recognized as a major cause of abortion in cattle while limited information is presently available on association between equine Neospora infections and abortions. The aim of the present study was to document prevalence of antibodies against Neospora sp. in aborted mares as a clue to the role of N. caninum in mare reproductive failure in Normandy, France. Using an agglutination test, the number of animals with elevated (>80) anti-Neospora sp. antibody titer was higher in a group of 54 aborted mares than in randomly chosen groups of 45 mares and 76 horses sampled for equine arteritis virus and Fasciola hepatica antibodies, respectively (P<0.001). N. caninum DNA was found in 3/91 fetal brains, 2/77 fetal hearts, and 1/1 placenta, and present in both brains and hearts of two fetuses. In 13 cases for which both mare serum and fetus were available, no fetal N. caninum amplification product was present while a large variation of maternal antibody titers was found. Data prompt at additional surveys of association between equine reproductive failure and Neospora sp. infection.  相似文献
