1. The effect of preincubation storage of chicken eggs on hatchability was studied in an Australorp subline (ASS) that had shown a steady decrease of hatchability compared with the original selection line (AS) since its derivation 10 generations ago. The AS line was also included in the study.

2. Eggs were collected over 21 d and the collected eggs were stored at 10° to 12°C and relative humidity of 70 to 80% until they were set together on d 21. Fertility and hatchability of fertile eggs were calculated for each d of collection to study their relationships with age of eggs (length of storage).

3. The hatchability of fertile eggs was considerably different between the two lines in the average over 21 d and, more importantly, in the pattern of decline with length of storage.

4. In the first 3 to 4 d of storage, hatchability was very similar between the lines. However, it decreased linearly at a rate of 1.9% per day of storage over the 21‐d period in the ASS line (average 65.9%), whereas in the AS line (average 78.6%), it stayed at around 84% up to 12 d of storage, when it started to decline at a similar rate as in the ASS line.

5. Fertility was 2.6% higher in the AS line than in the ASS line, but in each line showed no significant trend with age of eggs.  相似文献

This study was carried out to investigate the motor neurone degeneration in the ventral horn following transient spinal cord ischaemia at normothermic conditions in rabbits. Transient spinal cord ischaemia was induced by occlusion of the abdominal aorta underneath the left renal artery for 15 min at normothermia (38.7 degrees C). Sections at the level of L7 were examined using histochemical and electron microscopic methods. Cresyl violet-positive motor neurones began to reduce in number at 3 h after ischaemia reperfusion, and were not detectable at 48 h after ischaemia reperfusion. Acid fuchsin-positive motor neurones were detected at 1 h after ischaemia reperfusion, significantly increased up to 6 h after the ischaemia reperfusion, and eventually disappeared by 48 h after ischaemia reperfusion. In electron microscopic findings, the disintegration of cytoplasmic membranes, and the disruption of mitochondria and endoplasmic reticulum were observed in motor neurones at 30 min after ischaemia reperfusion. Motor neurones showed necrotic findings with pyknotic degeneration at 1 h after ischaemia reperfusion. The necrotic degeneration became severer time dependently after ischaemia reperfusion. At 48 h after ischaemia reperfusion, cellular components were not detectable in motor neurones. In conclusion, we suggest that the degeneration pattern of motor neurones of the ischaemic spinal cord was necrotic after ischaemia reperfusion under normothermic conditions.  相似文献

Chicken interleukin-17 (chIL-17) gene was previously characterized through cloning from a chicken intestinal expressed sequence tag (EST) cDNA library. To further investigate the biological properties of chIL-17, six monoclonal antibodies (mAbs) against a bacterially expressed chIL-17 recombinant protein were produced and their binding specificities characterized. Antibodies which were initially selected on the basis of their specific binding reactivity with recombinant chIL-17 in ELISA were further characterized by Western blot analysis. Monoclonal antibodies specific for chIL-17 identified 20 and 21kDa protein bands in the culture supernatant and cell lysate of CU205 cells. These mAbs also recognized specific bands for chIL-17 in the cell lysate from conconavalin A (Con A)-activated, but not from normal splenic lymphocytes. Furthermore, these mAbs detected a 16kDa protein in the lysate of CU205 cells treated with tunicamycin and stained an intracellular protein in CU205 cells in flow cytometric analysis. Together, these results indicate that these new mAbs are specific for chIL-17 and will be a useful tool for structural and immunological studies of IL-17 in poultry.  相似文献

This study investigated the effects of dietary δ-aminolevulinic acid (ALA) on growth performance, nutrient digestibility, blood parameters and whether ALA improved the immune response of weanling pigs challenged with Escherichia coli lipopolysaccharide (LPS). Eighty pigs (body weight = 7.21 ± 0.51 kg) were allotted to four dietary treatments, with four pens per treatment and five pigs per pen. Basal diets were supplemented with 0, 5, 10, and 15 mg/kg ALA (as-fed basis) and fed for 35 days. At the end of the feeding period, 10 pigs were selected from both the 0- and 10-mg/kg ALA treatment groups; five were injected i.p. with LPS (50 μg/kg BW) and the other five pigs with an equivalent amount of sterile saline, resulting a 2 × 2 factorial arrangement. Blood sample and rectal temperature data were collected at 0, 2, 4 and 12 h after challenge. Growth performance was not affected by dietary treatments over the total experimental period. However, dry matter (DM) and nitrogen (N) digestibility was improved in the 15-mg/kg ALA treatment group at day 35 (P < 0.05). Serum hemoglobin (Hb) and iron levels were also increased, with the 10-mg/kg ALA treatment showing the highest concentration (P < 0.05). On day 35, red (RBC) and white blood cell (WBC) counts were elevated, with the 5- and 10-mg/kg ALA treatments having the highest counts (P < 0.05). During challenge, LPS injection elevated rectal temperature at 2 and 4 h postchallenge (P < 0.05). Plasma cortisol concentration was also increased by LPS injection at 2 and 4 h postchallenge and an ALA-alleviating effect was evident at 2 h postchallenge (P < 0.01). Concentration of plasma insulin-like growth factor-I (IGF-I) was increased in the ALA-supplemented treatments at 2 h postchallenge (P < 0.05). LPS injection increased plasma tumor necrosis factor-α (TNF-α) concentrations at 2, 4 and 12 h (P < 0.01), while an ALA-alleviating effect was observed at 2 and 4 h postchallenge (P < 0.05 and P < 0.10, respectively). Challenge with LPS decreased WBC counts at 2 and 4 h postchallenge (P < 0.01). At 12 h postchallenge, RBC, WBC and lymphocyte counts were affected by LPS challenge, while an ALA effect was only observed on WBC count (P < 0.05). In conclusion, dietary supplementation of ALA in weanling pigs can improve DM and N digestibilities, and iron status and have a beneficial effect on the immune response during inflammatory challenge.  相似文献

In this study, we investigated changes in glutamic acid decarboxylase 67 (GAD67) immunoreactivity and its protein levels in the gerbil somatosensory cortex after ischemia/reperfusion. GAD67 immunoreactivity was significantly increased in layers III and V of the somatosensory cortex 12 hr after ischemia/reperfusion. Thereafter, GAD67 immunoreactivity was decreased with time after ischemia/reperfusion. GAD67 immunoreactivity in the somatosensory cortex 4 days after ischemia/reperfusion was similar to that in the sham-operated group. In addition, GAD67 protein levels were also significantly increased 12 hr after transient forebrain ischemia. These results suggest that the transient increase of GAD67 immunoreactivity in layers III and V may be associated with responses to transient ischemia-induced neuronal damage.  相似文献

Aluminum (Al) accumulation increases with aging, and long-term exposure to Al is regarded as a risk factor for Alzheimer''s disease. In this study, we investigated the effects of Al and/or D-galactose on neural stem cells, proliferating cells, differentiating neuroblasts, and mature neurons in the hippocampal dentate gyrus. AlCl3 (40 mg/kg/day) was intraperitoneally administered to C57BL/6J mice for 4 weeks. In addition, vehicle (physiological saline) or D-galactose (100 mg/kg) was subcutaneously injected to these mice immediately after AlCl3 treatment. Neural stem cells, proliferating cells, differentiating neuroblasts, and mature neurons were detected using the relevant marker for each cell type, including nestin, Ki67, doublecortin, and NeuN, respectively, via immunohistochemistry. Subchronic (4 weeks) exposure to Al in mice reduced neural stem cells, proliferating cells, and differentiating neuroblasts without causing any changes to mature neurons. This Al-induced reduction effect was exacerbated in D-galactose-treated mice compared to vehicle-treated adult mice. Moreover, exposure to Al enhanced lipid peroxidation in the hippocampus and expression of antioxidants such as Cu, Zn- and Mn-superoxide dismutase in D-galactose-treated mice. These results suggest that Al accelerates the reduction of neural stem cells, proliferating cells, and differentiating neuroblasts in D-galactose-treated mice via oxidative stress, without inducing loss in mature neurons.  相似文献

Lactic acid bacteria were screened for potential probiotics for use as feed additives. We obtained 3,000 isolates from feces of: cattle, dogs, goats, and infants; milk; yogurt; cheese; fermented sausages; Kimchi; and Cheonggukjang and tested their antibacterial activity toward indicator pathogens, including Bacillus cereus, Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella enterica Enteritidis. We further tested their tolerance to artificial gastric juice (1% [w/v] pepsin, pH 2.5) and bile acid (0.1% [w/v] oxgall, pH 6.8). Six isolates exhibited strong antibacterial activity against indicator pathogens. The PA40 isolate from Kimchi exhibited marked resistance to artificial gastric juice and bile acid. The antibacterial substances produced by PA40 were stable to heat, pH, and enzymes. Strain PA40 was identified as a Lactobacillus curvatus strain using chemical tests and 16S rDNA sequencing and produced 248.4 mmol/L lactic acid after 48 hr of fermentative growth. The L. curvatus PA40 strain was also highly tolerant of the artificial gastrointestinal model system. Our results indicate that L. curvatus PA40 could be used as a potential probiotic feed additive.  相似文献

Nanocomposites of polyurethane (PU) and multi-walled carbon nanotubes (MWNTs) were prepared via in-situ polymerization of poly(ɛ-caprolactone)diol (PCL)-grafted-MWNTs, 4,4′-methylene bis(phenyl isocyanate), and 1,4-butanediol. The grafting of PCL onto MWNTs was confirmed by Fourier transform infrared (FT-IR) spectroscopy and transmission electron microscopy (TEM). The nanocomposites showed more improved mechanical properties compared to conventional nanocomposites with the same MWNT loading. The thermo-responsive shape recovery as measured in a cyclic tensile test was observed to be approximately 80 % for in-situ nanocomposites, though it showed a reduced trend as the wt% of MWNTs increased. X-ray diffraction investigation also showed that the addition of MWNTs into the polyurethane increased the crystallinity. Scanning electron microscopy and TEM measurements showed better dispersion of MWNTs in the nanocomposites synthesized using in-situ method. Consequently, the presence of PCL-g-MWNTs made an important contribution to the enhancement of the mechanical and shape memory properties of polyurethane.  相似文献

Five different types of polyester geogrids whose nominal tensile strengths vary from 100 to 400 kN/m were investigated. Series of conventional long-term (ASTM D5262) and stepped isothermal method (SIM, ASTM D6992) tests were performed to evaluate the long-term creep strain of these geogrids. The conventional long-term creep strains for 10,000 hours varied from 1.32 % and 0.53 %. Knitted geogrids showed higher creep strains than those associated with woven geogrids. The results of SIM tests showed that a minimum of 6 to 8 steps should be used in order to predict the creep behavior beyond 75-year design life. SIM tests showed good match with the results obtained from long-term creep tests for woven geogrids. Based upon SIM test results obtained from the test geogrids, the creep reduction factor of 1.50 is a reasonable value for 75-year design life.  相似文献
