Two novel metabolites with potential antifeedant activity were isolated from cultures of the fungus Trichoderma citrinoviride strain ITEM 4484 grown in solid-state fermentation on sterile rice kernels. The producing strain was identified at species level by sequence analysis of the internal transcribed spacer regions ITS-1 and ITS-2 of the nuclear rDNA and a fragment of the translation elongation factor gene TEF-1alpha. Fractionation by column chromatography and TLC of the culture organic extract, followed by feeding preference tests on the aphid pest Schizaphis graminum (Rondani), allowed the purification of 5.8 and 8.9 mg/kg of culture of two bioactive metabolites, which were named citrantifidiene and citrantifidiol ( 1 and 2). Citrantifidiene and citrantifidiol, whose structures were determined by spectroscopic methods (NMR and MS) are a symmetrical disubstituted hexa-1,3-dienyl ester of acetic acid and a tetrasubstituted cyclohexane-1,3-diol, respectively. The pure metabolites influenced the feeding preference of S. graminum restraining individuals from feeding on wheat leaves dipped in 5% aqueous methanol solution containing 0.57 mg/mL of citrantifidiene or 0.91 mg/mL of citrantifidiol.  相似文献

Coke Wastewater Treatment by a Three-Step Activated Sludge System   总被引:1,自引:0,他引:1  
The treatment of industrial coke wastewater was studied in a laboratory-scale activated sludge system. The concentrations of the main pollutants in the wastewater ranged between 800 and 1870 mg COD/l, 100–221 mg phenols/l, 198–427 mg SCN/l, 133–348 mg ${\text{NH}}_4^ + - {{\text{N}} \mathord{\left/ {\vphantom {{\text{N}} {\text{l}}}} \right. \kern-0em} {\text{l}}}$ and 11–41 mg CN?/l. To avoid inhibition phenomena resulting from the high concentrations of thiocyanate, ammonium nitrogen and cyanide a three-step process was implemented. The first step was anoxic for the removal of nitrates, followed by an oxic step during which biodegradation of phenols and thiocyanates took place, and by a second oxic step to oxidize ammonium nitrogen to nitrate. The dilution effect due to the recirculation of the final effluent to the head of the process and also, the separation of the nitrification step from the biodegradation of thiocyanate led to much higher efficiencies than when the process was carried out simultaneously. Very high removals were obtained (99% phenols, 97% SCN?, 63% COD, 98% ${\text{NH}}_4^ + - {\text{N}}$ , 90% total-N and 99% cyanide) employing hydraulic residence times of 15.4 h for denitrification, 98 h for phenol and thiocyanate biodegradation and 86 h for nitrification.  相似文献

Soil CO2 emission (FCO2) in agricultural areas results from the interaction of different factors such as climate and soil conditions. Our objective was to investigate the spatiotemporal variation of FCO2, temperature (Tsoil), moisture (Msoil) and air-filled pore space (AFPS), as well as their interactions, during the sugarcane field reform. The study was conducted on a 90 × 90 m sampling grid with 100 points at 10 m spacings. Ten assessments of FCO2, Tsoil and Msoil were carried out at each point over a 28-day period. The greatest mean values of FCO2 (0.74 g m−2 hr−1) and Msoil (31.7%) were obtained on Julian day 276, 2013, being associated with precipitation events at the study site. Also, the smallest values of AFPS (19.17%) and Tsoil (20.90°C) were observed on the same day. The spatial variability of FCO2, Tsoil, Msoil and AFPS was best described by an adjusted spherical model, although an exponential model better fitted some results. The spatial pattern of all soil attributes showed little temporal persistency, indicating a high complexity for FCO2 during precipitation. Correlation maps assisted in identifying regions where Msoil and AFPS better controlled the emission process and where Tsoil was important. A major challenge for world agriculture is to increase the efficiency of conventional soil management practices. We highlight the importance of the spatial pattern of soil properties that directly influence the CO2 emission dynamics. Future mitigation actions should involve less intense tillage and ensure homogeneous applications of soil inputs, thereby reducing production costs and the contribution of these activities to CO2 emissions during the sugarcane field reform.  相似文献

The “gold standard” for verification of patient positioning before linear accelerator‐based stereotactic radiation therapy is kilovoltage cone‐beam computed tomography (kV‐CBCT), which is not uniformly available or utilized; planar imaging is sometimes used instead. The primary aim of this study was to determine if the position of the bony skull, when used as a surrogate for isocenter verification, is different when orthogonal megavoltage (MV) portal or kilovoltage (kV/kV) radiographs are used for image guidance, rather than kV‐CBCT. A secondary aim was to determine the influence of intra‐observer variability, body size and skull conformation on positioning, as determined using these three imaging modalities. Dogs and cats receiving radiotherapy of the head were recruited for this prospective analytical study. Planar (MV portal and kV/kV images) and volumetric (kV‐CBCT) images were acquired before treatment, and manually coregistered with reference images. Differences in skull position when matched based on MV portal, kV/kV images and kV‐CBCT were compared. A total of 65 subjects and 148 unique datasets were evaluated. The Wilcoxon rank‐sum test was used to evaluate effects of transitioning between imaging modalities. When comparing magnitude of shifts in MV to kV‐CBCT, MV to kV/kV and kV/kV to kV‐CBCT, there were statistically significant differences. Results were not measurably impacted by body size, skull conformation or interobserver differences. Based on shift magnitude and direction, an isotropic setup margin of at least 1 mm should be incorporated within the planning target volume when MV or kV planar imaging is used for position verification.  相似文献

Swine are known reservoirs for Clostridioides difficile, formerly known as Clostridium difficile, and transmission from swine to human farm workers is strongly suggested by previous studies. This cross‐sectional study evaluated the potential role of farm environmental surfaces, including those in worker breakrooms and swine housing areas, in the possible transmission of C. difficile from swine to farm workers. Environmental surfaces and piglet faeces at 13 Ohio swine farms were sampled in 2015. Typical culturing techniques were performed to isolate C. difficile from samples, and amplification of toxin genes (tcdA, tcdB and cdtB) and PCR‐ribotyping were used to genetically characterize recovered isolates. In addition, sequencing of toxin regulatory gene, tcdC, was done to identify the length of identified deletions in some isolates. A survey collected farm‐level management risk factor information. Clostridioides difficile was recovered from all farms, with 42% (188/445) of samples testing positive for C. difficile. Samples collected from all on‐farm locations recovered C. difficile, including farrowing rooms (60%, 107/178), breakrooms (50%, 69/138) and nursery rooms (9%, 12/129). Three ribotypes recovered from both swine and human environments (078, 412 and 005) have been previously implicated in human disease. Samples taken from farrowing rooms and breakrooms were found to have greater odds of C. difficile recovery than those taken from nursery rooms (OR = 40.5, OR = 35.6, p < .001 respectively). Farms that weaned ≥23,500 pigs per year had lower odds of C. difficile recovery as compared to farms that weaned fewer pigs (OR = 0.4, p = .01) and weekly or more frequent cleaning of breakroom counters was associated with higher odds of C. difficile recovery (OR = 11.7, p < .001). This study provides important insights into the presence and characterization of C. difficile found in human environments on swine farms and highlights how these areas may be involved in transmission of C. difficile to swine farm workers and throughout the facility.  相似文献

The RAPD-PCR technique has been used to generate DNA amplification profiles from a set of advanced generation inbred lines of white clover ( Trifolium repens L.). The genetic distances between lines have been estimated and have been compared with the extent of positive heterosis for dry matter yield in hybrid crosses. The analysis clearly separated the four major groups (H, J, R and S), with little within group variation. Heterosis, previously shown to be positively correlated with the extent of phenotypic variation between parental lines, was greatest between lines from groups showing the least genetic distance. The lack of correlation between heterosis and genetic divergence is discussed in the context of results from other systems. Additionally, the data allow a comparison of the extent of phenotypic variation for a trait under selective pressure with the degree of variation at the DNA level detected with the RAPD technique. The pecan nut [Carya illinoinensis (Wangenh) C. Koch] is a natural source of polyphenols with antioxidant properties. In this study, the encapsulation of aqueous and hydroalcoholic extracts of pecan nut shell were evaluated for the release of bioactive compounds and antioxidant potential in order to explore food applications using zein as encapsulating agent. The extracts showed high contents of total phenolics, condensed tannins and high antioxidant activity. Concentrations of proanthocyanidins were 9-fold higher in hydroalcoholic extracts. The LC-DAD analysis showed that catechins were the major phenolic compounds in samples, with epigallocatechin levels up to 138.62 mg mL?1. Zein microparticles loaded with aqueous extract released 2.3 times more phenolic compounds than the hydroalcoholic extracts and the DSC thermograms showed that extracts of pecan nut shell remained thermally stable up to 240 °C. The zein microcapsules obtained in this study were efficiently encapsulated and represent an interesting additive due its high antioxidant capacity, physicochemical characteristics and morphology. The use of zein microparticles combined with natural extracts constitute a step forward in the improvement of current technology for delivering phenolic compounds with applications in functional foods and nutraceuticals.  相似文献

Twig cankers and diebacks were observed on cherry laurel (Prunus laurocerasus L.) hedges located in a private garden of Perugia (Central Italy). Four fungal isolates, forming gray colonies and aerial mycelium, were obtained from cankers. For each isolate, Koch’s postulates were fulfilled by inoculating cherry laurel plants. Based on cultural characteristics, rep-PCR and phylogenetic analyses on ITS and EF1-α nucleotides sequences, the isolates were identified as Diplodia seriata De Not. To the best of our knowledge, this is the first report of D. seriata as a new pathogen of P. laurocerasus. The severe symptoms and the wide distribution of the host plant make the pathogen potentially dangerous for this ornamental species.  相似文献
