A variety of membrane-specific parameters was examined in both intact cells and isolated plasma membranes following exposure of cultured human liver cells to the insecticide 1,1-(2,2,2-trichloroethylidene)bis(4-chloro)benzene (DDT). Uptake of DDT was at equilibrium within 6 hr. In contrast, a decrease in the number of β-adrenergic hormone receptors first became significant after 48 hr of cell exposure. Whereas the uptake was largely reversible, the loss in the number of β receptors did not recover after DDT-exposed cells were cultured in fresh medium lacking the insecticide. Experiments in vitro substantiated the time lag of the biological effect. The decrease in receptor proteins was persistent in membranes with increased phospholipid unsaturation. Temperature-activity profiles (“Arrhenius plots”) of Na+K+-ATPase and 5′-nucleotidase were unchanged. Endogenous tryptophan fluorescence of membrane proteins was lower in membranes from DDT-exposed cells. These selective alterations in membrane parameters suggest a specific interaction of DDT with membrane proteins; interference with cellular protein synthesis is possible. The results indicate that membrane lipid “fluidization” does not play a physiologically important role in the mechanism of DDT action in biomembranes.  相似文献

A prior method for determination of glyphosate in water samples has been modified to accommodate samples of crops and soils. Differential pulse polarography as the determinative step enables analysis in an aqueous medium, which is important during extraction of this compound. Residues are cleaned up and concentrated by ion exchange on a strong basic resin in OH- form. The method is rapid, is applicable to a relatively broad selection of sample types, and gives recoveries consistently better than 60% with good precision. The main shortcoming of the method is that the limit of detection of 0.5-1.0 ppm may sometimes be inadequate. The metabolite, aminomethyl phosphonic acid, is not detected.  相似文献

A 3-year-old Warmblood mare was presented with spinal ataxia of 3 months’ duration. Clinically, reduced mobility of the neck and a prominent atlas were identified. Neurological evaluation revealed hypermetria, circumduction and reduced proprioception, predominantly in the hindlimbs. Occipitoatlantoaxial malformation (OAAM) was diagnosed based on radiography and computed tomography, and confirmed on post-mortem magnetic resonance imaging and pathological examination. Both the atlas and axis were hypoplastic and dysplastic, and a fusion of the atlanto-occipital joint was found. A dynamic compression of the spinal cord was present at the first and second cervical vertebrae. Histologically, the spinal cord was degenerated due to compression. Furthermore, there were muscular malformations including a fibrotic obliquus capitis cranialis muscle, aplasia of parts of the rectus capitis muscles and presence of additional abnormal muscle. Such muscular abnormalities associated with OAAM have not been reported yet and this is the first report of a case of OAAM in a Warmblood horse.  相似文献

Objectives

To investigate QT instability in dogs with myxomatous mitral valve disease (MMVD) and to determine if this is associated with arrhythmogenesis.

Animals

One hundred sixty-seven MMVD dogs that met the study criteria were included.

Methods

Echocardiographic and electrocardiographic data were gathered. Fifty consecutive QT intervals were recorded for each dog. Both heart rate–corrected and uncorrected QT intervals were used to calculate average QT (QTa), QT variance (QTv), total instability (TI), short-term instability (STI), and long-term instability (LTI). Sensitivity and specificity of QTa, QTv, TI, STI, and LTI in identifying arrhythmias and cardiac remodeling were calculated. Patient follow-ups were obtained for analyses of disease progression and survival.

Results

An increase related to progression was documented for all the studied indices. QTa and STI best identified dilated hearts and arrhythmias, respectively. Dogs with QTa >272 ms and STI >8 ms were 15% more likely to develop ventricular arrhythmias (likelihood ratios of 2.31 [P = 0.0008] and 2.09 [P = 0.0049], respectively). A QTa >258 ms discriminated American College of Veterinary Internal Medicine stage B1 from stages B2/C disease with a sensitivity of 63% and specificity of 61%. Dogs in American College of Veterinary Internal Medicine stage C of MMVD have higher STI and 3.34 times increased risk of developing arrhythmias when values more than 8 ms are reached. All indices except LTI and QTv showed prognostic value, with increases relating to all-cause mortality.

Conclusion

Analyses of QT intervals demonstrated changes in STI, LTI, and TI. Increased QT prolongation and instability are significantly related to mortality and may be useful in determining prognosis of MMVD patients.  相似文献

Introduction

Left ventricular systolic function is one of the main parameters studied in echocardiography. Longitudinal systolic function, however, is less commonly evaluated in routine examinations but may provide early information on systolic dysfunction. The movement of the mitral annulus toward the apex has already been determined as a method for evaluation of longitudinal systolic function in dogs, but the study of this movement by speckle tracking with the tissue motion annular displacement (TMAD) technique has not yet been evaluated.

Animals

One hundred fifty-three client-owned healthy dogs.

Methods

Cross-sectional study. One hundred fifty-three client-owned healthy dogs underwent physical examination, electrocardiography, blood pressure measurement, and a standard and speckle tracking echocardiography. Systolic function was evaluated by global longitudinal strain (GLS) and TMAD. These parameters were compared with the standard echocardiographic data.

Results

A correlation was found between GLS, TMAD, and body weight. Tissue motion annular displacement and GLS were significantly correlated (p < 0.001) with other surrogates of systolic function, including ejection fraction and fractional shortening. There were no differences in TMAD between sexes. The coefficient of variation (CV) of the intraobserver evaluation in the global TMAD (CV 4.44) was slightly higher than that in the GLS (CV 3.74). Also, TMAD was not influenced by heart rhythm and could be acquired more rapidly than GLS.

Conclusions

Tissue motion annular displacement is a rapid and reproducible method for the assessment of left ventricle longitudinal function in healthy dogs. However, more studies are needed to validate the real clinical applicability of TMAD in animals with heart diseases.  相似文献

To gain information about the geographical origin of oil samples, measurements of delta(13)C and delta(18)O of the whole oil and some of its fractions have been performed on samples coming from fruits of Olea europaea L. produced in Greece, Morocco, Spain, Italy, Tunisia, and Turkey. The results obtained by applying statistical procedures have given pieces of evidence that oil samples have shown the trend to cluster according to the different climatic areas of growing environment of fruits. Some confusion has been observed for samples coming from neighboring countries having similar climates.  相似文献

A comparative study of two kiwifruit genotypes (Actinidia deliciosa (A. Chev.) C.F. Liang et A.R. Ferguson var. deliciosa) with different tolerance to iron (Fe) deficiency was conducted to identify biochemical features associated with tolerance to Fe deficiency. After 14 days of growth in hydroponic culture under Fe-deficient and Fe-sufficient conditions, leaf chlorophyll concentration, activities of ferric chelate reductase (FCR), phosphoenolpyruvate carboxylase (PEPC) and citrate synthase in root extracts, concentrations of organic acids in roots, leaves and xylem sap, and xylem sap pH were measured. In response to Fe deficiency, the tolerant genotype D1 showed: (i) higher FCR activity associated with a longer lasting induction of FCR; (ii) higher PEPC activity; (iii) higher concentrations of citric acid in roots; and (iv) lower xylem sap pH compared with the susceptible genotype Hayward. These findings imply that induction of FCR and PEPC activities in roots in response to Fe deficiency are important physiological adaptations enabling Fe-efficient kiwifruit plants to tolerate Fe deficiency.  相似文献
