Summary The possibility of producing agronomically-useful somaclones via organogenesis and somatic embryogenesis from callus cultures of pea (Pisum sativum L.) was studied. Organogenic calli were induced from immature leaflets on MSB medium with NAA and BAP. Embryogenic calli were derived either from immature zygotic embryos (using 2,4-D) or from shoot apices (using picloram) of aseptically-germinated seedlings.The seed progenies (T1 to T3-generation) of primary regenerants were grown in field conditions and their phenotypic variation was evaluated and compared with control, non-tissue culture-derived plant material. In addition, electrophoretic analyses of selected isoenzyme systems and total proteins have been done. The results do not show dramatic changes in qualitative and quantitative traits. The aim of this study was to investigate temporal and spatial patterns of denitrification enzyme activity (DEA) and nitrous oxide (N2O) fluxes in three adjacent riparian sites (mixed vegetation, forest and grass). The highest DEA was found in the surface (0–30 cm) soil and varied between 0.7±0.1 mg N kg–1 day–1 at 5°C and 5.9±0.4 mg N kg–1 day–1 at 15°C. There was no significant difference (P >0.05) between the DEA in the uppermost (0–30 cm and 60–90 cm) soil depths under different vegetation covers. In the two deepest (120–150 cm and 180–210 cm) soil depths the DEA varied between 0.0±0.0 mg N kg–1 day–1 at 5°C and 4.4±0.9 mg N kg–1 day–1 at 15°C and was clearly associated with the accumulation of buried organic carbon (OC). Two threshold values of OC were observed before DEA started to increase significantly, namely 5 and 25 g OC kg–1 soil at 10–15°C and 5°C, respectively. In the three riparian sites N2O fluxes varied between a net N2O uptake of –0.6±0.4 mg N2O-N m–2 day–1 and a net N2O emission of 2.5±0.3 mg N2O-N m–2 day–1. The observed N2O emission did not lead to an important pollution swapping (from water pollution to greenhouse gas emission). Especially in the mixed vegetation and forest riparian site highest N2O fluxes were observed upslope of the riparian site. The N2O fluxes showed no clear temporal trend.  相似文献

Two species of Cryptosporidium are commonly identified in pigs: Cryptosporidium suis and Cryptosporidium pig genotype II. Detection of Cryptosporidium spp. is routinely based on molecular methods such as polymerase chain reaction (PCR) and subsequent restriction fragment length polymorphism (RFLP) or gene sequencing. However, most of these methods are hampered by low sensitivity to mixed infections. As a solution of this problem, novel species-specific primers were designed and tested in the present study. Sensitivity of our primers was identical to genus-specific primers, but more (1:48) mixed infections were detected using these species-specific primers on 477 DNA samples originating from naturally infected pigs of different age categories. Our results show differences in age-dependent susceptibility of pigs to the infection. Whereas C. suis was found in all tested categories of pigs (1-12 week of age and sows), Cryptosporidium pig genotype II was recorded only in animals older than 6 week of age. Usage of species-specific primers could help to better the understanding of epidemiology of pig specific Cryptosporidium spp. and its occurrence, which, on the basis of our results, is underestimated.  相似文献

The purpose of this study was to gain information concerning chronology of postnatal dental eruption in degus (Octodon degu). Specific findings pertain to the estimation of postnatal age at tooth emergence through the gingiva and to the age when a particular tooth is in functional occlusion. Fifty newborn degu pups were included in this study. The oral cavity endoscopy was carried out under isoflurane anesthesia in all animals 3 to 4 and 24-hours after birth, then at 1-day intervals until the age of 40-days, and subsequently at 2-day intervals until 75-days of age. Tooth emergence was considered to have occurred when any portion of the crown had penetrated the gingiva. The stage when the teeth come into occlusion was defined as the functional occlusion. The degus were born with completely erupted permanent incisors that were already in functional occlusion. All the premolar teeth and all the first molar teeth erupted on days 2 - 3 and 4 - 5 of life, respectively. The first signs offunctional occlusion ofall the first 2 cheek teeth were observed on days 9 - 11. Maxillary and mandibular second molar teeth emerged between days 17 - 19 and 18 - 20, respectively. First signs of functional occlusion were observed on days 30 - 31 and 31 - 33for maxillary and mandibular second molar teeth, respectively. Maxillary third molar teeth emerged on days 38 - 44 and erupted into occlusion on days 48 - 58. Mandibular third molar teeth emerged on days 39 - 46 and erupted into occlusion on days 58 - 72.  相似文献

ObjectiveTo develop a safe and effective immobilization protocol in rhesus monkeys, which is not based on dissociative anaesthetic agent.Study designProspective, randomised, experimental trial.AnimalsTwenty rhesus monkeys, weighing 2.6–8.0 kg, 1–3 years of age, of both sexes.MethodsThe monkeys received 50 μg kg?1 medetomidine, 0.25 mg kg?1 midazolam and 5 μg kg?1 fentanyl with 150 IU hyaluronidase intramuscularly (IM). The animals were closely observed for behavioural changes and reaction to sound stimulus. Pulse rate and oxygen saturation of haemoglobin (SpO2) were monitored every 5 minutes, for 20 minutes. After this period, 250 μg kg?1 atipamezole or a placebo was administered IM and behavioural changes were closely observed.ResultsFull immobilization was observed after mean 269 ± SD 116 seconds. Ten minutes after injection mean arterial oxygen saturation of haemoglobin was 94 ± 4%, but did not fall significantly further. The median pulse rate was 116 beats minute?1 5 minutes after the administration of the drug. This level further decreased to a median level of 108 beats minute?1 20 minutes after the drug's administration. The median time to recover from immobilization was significantly shorter after atipamezole administration when compared to placebo (2.7 versus 55 minutes). All animals awoke smoothly and no side effects such as vomiting or agitation were observed.ConclusionsShort term and reversible pharmacological immobilization was achieved using combination of midazolam, medetomidine, and fentanyl.Clinical relevanceThe present study demonstrates that 20-minute pharmacological immobilization with a combination of midazolam, medetomidine, and fentanyl is feasible in rhesus monkeys with minimal effect on heart rate.  相似文献

In this case report from Italy we describe a fatal infection caused by A. hydrophila in a 39 yr old cirrhotic patient. This pathogen was isolated as a pure single culture from the patient's blood sample. The patient died on the second day of hospitalization from overwhelming sepsis. The A. hydrophila isolate was tested for different potential virulence properties, such as invasiveness, adherence, exotoxins production, presence of fimbriae and for the patterns of resistance to a variety of antimicrobial agents. Although, the Aeromonas species are infrequently reported as a cause of human infections, the present case study confirms the capability of these pathogens to induce serious human infections.  相似文献

Flow cytometry was used to screen a panel of 320 mAbs, submitted to the Animal Homologues Section of the HLDA8, for mAbs that recognize epitopes conserved on orthologous leukocyte differentiation antigens (LDA) in goats, lamas, and rabbits. Nineteen mAbs specific for CD11a (1), CD14 (3), CD18 (1), CD21 (1), CD29 (2), CD44 (2), CD47 (3), CD49d (1), CD172a (1), CD45RB (1), CD61 (1), RACT48A, and GBSP71A reacted with goat LDA. Twenty three mAbs specific for CD7 (1), CD9 (2), CD11a (1), CD14 (3), CD18 (4), CD29 (1), CD32 (1), CD44 (1), CD47 (4), CD49d (2), CD50 (1), CD80 (1), CD172a (1), and GBSP71A reacted with llama LDA. Eighteen mAbs specific for CD9 (2), CD11a (1), CD14 (2), CD18 (4), CD21 (1), CD44 (2), CD45RB (1), CD49d (1), CD209 (1), RACT48A, and GBSP71A reacted with rabbit LDA. The specificities of two cross reactive mAbs that recognize different conserved epitopes on all leukocytes in two species (RACT48A) and all three species (GBSP71A) have not been determined. The patterns of reactivity of most of the mAbs were consistent with patterns of reactivity noted on human leukocytes. The specificity of some cross reactive mAbs generated in non-human species were validated on human leukocytes. Further studies are needed to verify that CD7, CD32, CD45RB, CD50, and CD209 recognize orthologous molecules in the indicated species.  相似文献

Vertebral column pathologies requiring surgical intervention have been described in pet ferrets, however little information is available on the normal vertebral formula and congenital variants in this species. The purpose of this retrospective study was to describe vertebral formulas and prevalence of congenital vertebral anomalies in a sample of pet ferrets. Radiographs of 172 pet ferrets (96 males and 76 females) were included in this retrospective study. In 143 ferrets (83.14%), five different formulas of the vertebral column were recorded with normal morphology of vertebrae (rib attachment included) but with a variable number of thoracic (Th), lumbar (L), and sacral (S) vertebrae. The number of cervical (C) vertebrae was constant in all examined animals. Observed vertebral formulas were C7/Th14/L6/S3 (51.74%), C7/Th14/L6/S4 (22.10%), C7/Th14/L7/S3 (6.98%), C7/Th15/L6/S3 (1.74%), and C7/Th15/L6/S4 (0.58%). Formula C7/Th14/L6/S4 was significantly more common in males than in females (P < 0.05). Congenital spinal abnormalities were found in 29 ferrets (16.86%), mostly localized in the thoracolumbar and lumbosacral regions. The cervical region was affected in only one case. Transitional vertebrae represented the most common congenital abnormalities (26 ferrets) in the thoracolumbar (13 ferrets) and lumbosacral regions (10 ferrets) or simultaneously in both regions (three ferrets). Other vertebral anomalies included block (two ferrets) and wedge vertebra (one ferret). Spina bifida was not detected. Findings from the current study indicated that vertebral formulas may vary in ferrets and congenital abnormalities are common. This should be taken into consideration for surgical planning.  相似文献
