The effects of night temperature on biomass accumulation and plant morphology were examined in rice ( Oryza sativa L.) during vegetative growth. Plants were grown under three different night temperatures (17, 22 and 27°C) for 63 days. The day temperature was maintained at 27°C in all treatments. The final biomass of the plants was greatest in the plants grown at the highest night temperature. Total leaf area and tiller number were also the greatest in this treatment. Growth analysis indicated that the relative growth rate in the 27°C night-temperature treatment was maximal between days 21–42 and this was caused by increases in leaf area ratio, leaf weight ratio and specific leaf area. Plant total nitrogen contents did not differ among treatments. However, nitrogen allocation to the leaf blades was highest and the accumulation of sucrose and starch in the leaf blades and sheaths was the lowest in the 27°C night-temperature treatment by day 42. Despite this, dark respiration was also highest, and both the gross and net rates of CO2 uptake at the level of the whole plant at day 63 were the highest in the 27°C night-temperature treatment. Thus, high night temperature strongly stimulated the growth of leaf blades during the early stage of rice plant growth, leading to increased biomass during the vegetative stage of the rice plants. As the CO2 uptake rate per total leaf area was higher, photosynthesis at the level of the whole plant was also stimulated by a high night temperature.  相似文献

The aim of the present study was to detect quantitative trait loci affecting fatty acid composition in back fat and intramuscular fat in a Duroc pig population comprising seventh‐generation pedigrees using genome‐wide association studies (GWAS). In total, 305 animals were genotyped using single nucleotide polymorphisms (SNPs) array and five selected SNPs from regions containing known candidate genes related to fatty acid synthesis or metabolism. In total, 24 genome‐wide significant SNP regions were detected in 12 traits, and 76 genome‐wide suggestive SNP regions were detected in 33 traits. The Sus scrofa chromosome (SSC) 7 at 10.3 Mb was significantly associated with C17:0 in intramuscular fat, while the SSC9 at 13.6 Mb was significantly associated with C14:0 in intramuscular fat. The SSC12 at 1.0 Mb was significantly associated with C14:0 in back fat and the SSC14 at 121.0 Mb was significantly associated with C18:0 in intramuscular fat. These regions not only replicated previously reported loci containing some candidate genes involved in fatty acid composition (fatty acid synthase and stearoyl‐CoA desaturase) but also included several additional related loci.  相似文献

A rigid-type of polyethylene T-cannula was fitted into the anterior ileum of six horses in order to improve the cannulation techniques. A piece of polyethylene net was fastened onto the intestinal wall around the cannula to prevent dislodgment of the cannula by promoting a secure adhesion between the ileum and the abdominal wall. The cannula barrel sheathed with silicone tubing was exteriorized through a stab incision at the lateral ventral wall on the transverse line of the second lumber vertebra, and a flange was screwed onto the barrel. The feeding regime gradually increased concentrate without roughage prevented any colic signs. The use of these techniques succeeded in the ileal cannulation with no leakage of digesta.  相似文献

OBJECTIVE: To determine effects of carprofen and dexamethasone on chondrocytes in a culture model of osteoarthritis (OA). SAMPLE POPULATION: Chondrocytes isolated from articular cartilage of the humeral head of 5 adult dogs. PROCEDURE: Chondrocytes were harvested, cultured and subcultured in monolayer, and then cultured in a 3-dimensional (3-D) medium. Cells from each dog were distributed into 6 groups with differing content of liquid medium for each 3-D construct (agarose [AG], AG plus interleukin [IL]-1beta, AG plus carprofen [4 microg/mL], AG plus dexamethasone [1 mg/mL], AG plus IL-1beta [20 ng/mL] plus carprofen [4 microg/mL], and AG plus IL-1beta (20 ng/mL) plus dexamethasone (1 mg/mL). On days 3, 6, 12, and 20 of culture, samples from all groups were collected. Liquid media were assayed for glycosaminoglycan, prostaglandin (PG)E2, matrix metalloprotease (MMP)-3, and MMP-13 concentrations. All 3-D constructs were evaluated for viability, cell morphology, proteoglycan staining, and collagen type-II concentration. Total glycosaminoglycan content in each 3-D construct was quantitated by spectrophotometric assay. RESULTS: Addition of IL-1beta caused a significant loss of cell viability and matrix production. Addition of carprofen or dexamethasone caused significant decreases in PGE2 in the liquid media, and each was minimally effective in protecting chondrocytes against negative effects of IL-1beta. CONCLUSIONS AND CLINICAL RELEVANCE: Human recombinant IL-1beta resulted in loss of cell viability, alterations in extracellular matrix components, and production of PG and MMP Carprofen and dexamethasone had little effect on cell and matrix variables but did decrease PGE2 concentrations and primarily affected the inflammatory pathway of osteoarthritis.  相似文献

OBJECTIVE: To determine biomechanical and biochemical properties of the medial meniscus in a semi-stable stifle model and in clinical patients and to determine the effect of canine recombinant somatotropin hormone (STH) on those properties. ANIMALS: 22 healthy adult dogs and 12 dogs with meniscal damage secondary to cranial cruciate ligament (CCL) rupture. PROCEDURE: The CCL was transected in 15 dogs, and stifles were immediately stabilized. Implants releasing 4 mg of STH/d were placed in 7 dogs, and 8 received sham implants. Seven dogs were used as untreated controls. Force plate analysis was performed before surgery and 2, 5, and 10 weeks after surgery. After 10 weeks, dogs were euthanatized, and menisci from surgical and contralateral stifles were harvested. The torn caudal horn of the medial meniscus in dogs with CCL rupture comprised the clinical group. Creep indentation determined aggregate modulus (HA), Poisson's ratio (v), permeability (k), and percentage recovery (%R). Water content (%W), collagen content (C), sulfated glycosaminoglycan (sGAG) content, and collagen type-I (cI) and -II (cII) immunoreactivity were also determined. RESULTS: Surgical and clinical groups had lower HA, k, %R, C, sGAG, cI, and clI and higher %W than the non-surgical group. Surgical stifles with greater weight bearing had stiffer menisci than those bearing less weight. Collagen content was higher in the surgical group receiving STH than the surgical group without STH. CONCLUSIONS AND CLINICAL RELEVANCE: Acute stabilization and moderate weight bearing of the CCLdeficient stifle appear to protect stiffness of the medial meniscus. Normal appearing menisci from CCL-deficient stifles can have alterations in biomechanical and biochemical properties, which may contribute to meniscal failure.  相似文献

To investigate the ecological importance of the cellulolytic bacterium Fibrobacter succinogenes in fiber digestion, ruminal distribution of F. succinogenes was determined in relation to its phylogenetic grouping. Rumen digesta from wethers and steers fed orchardgrass hay, rice straw or fresh orchardgrass were employed as the materials for the analyses. Orchardgrass hay stem incubated in the rumen was also used. By using total DNA extracted from these materials, population sizes of total F. succinogenes and of four different phylogenetic groups of this species were quantitated through competitive polymerase chain reaction (PCR), and restriction fragment length polymorphism analysis of PCR products targeted the bacterial 16S rDNA. Rumen digesta and ruminally incubated hay stems had a reasonably high population size of F. succinogenes (×107?8/g) that was composed of strains belonging to the phylogenetic groups 1 and 3. The relative abundance of each group was different among the samples; group 1 dominated on the ruminally incubated hay stem and in the rumen of wethers fed fresh orchardgrass, while group 3 was major in the rumen of wethers and steers on hay diet. These results suggest that there could be phenotypic differences among the phylogenetic groups of F. succinogenes, and group 1 dominating on hay stem might contribute to rumen fiber digestion more than the other groups.  相似文献

Data from 1170 records of fattening calves were collected on growth and carcass traits from a Japanese Black cattle herd located in Miyagi prefecture, Japan. The objective was to determine direct and maternal heritabilities, direct and maternal genetic correlations and phenotypic correlations between bodyweight at the beginning of the fattening period (BWS), bodyweight at the end of the fattening period (BWF), carcass weight (CW), average daily gain during the fattening period (ADG), rib eye area (REA), rib thickness, subcutaneous backfat thickness (SFT), yield estimate (YE) and beef marbling score (BMS). Direct heritability estimates of 0.16 (SFT) and 0.07 (BMS) were low, whereas estimates of the other traits were medium to high and ranged between 0.44 (REA) and 0.78 (CW). Direct genetic correlations were all positive, except those that were between BWS and SFT, and between BWS and YE (?0.49 and ?0.14, respectively). The lowest positive genetic correlation was between BWS and BMS (0.04) and the highest was between BWF and CW (0.99). The phenotypic correlation coefficients ranged between ?0.41 (between SFT and YE) and 0.96 (between BWF and CW). Maternal heritability estimates were generally low and ranged between 0.00 for BMS and 0.08 for BWS, CW and ADG. Selection programs comprising information on growth and carcass traits of calves and maternal traits of dams were suggested.  相似文献

The objective of this study was to evaluate the effects of grazing on the accretion of energy and chemical components in steers, using the comparative slaughter technique and taking into consideration the growth rate. Twelve Holstein steers aged 6 months and weighing 160 kg were divided into three treatment groups – an initial, grazing and control group. The initial group was slaughtered at the beginning of the experiment. The grazing group was grazed on timothy pastures for 18 weeks. The control group was kept in a stable and offered concentrate and timothy hay in a proportion of 2:1 throughout the 18 weeks. The amount of ration offered to the control group was adjusted in order to obtain a growth rate similar to that of the grazing group. The grazing and control groups were slaughtered at the end of the experiment. The grazing and the control groups had similar growth rates of approximately 0.8 kg/day throughout the experimental period and similar final live weights of approximately 260 kg. The grazing group had lower fat and energy deposition rates than the control group (P < 0.05). Deposition rates of protein, fat, ash, moisture and energy were 130, 32, 30, 374 g/day and 4.4 MJ/day, respectively, in the grazing group and 117, 85, 32, 381 g/day and 6.2 MJ/day, respectively, in the control group. The net energy for growth was lower in the grazing group (7.7 MJ/kg) than in the control group (9.9 MJ/kg, P < 0.05). The grazing group had higher plasma concentrations in growth hormone and lower plasma concentrations in insulin and insulin‐like growth factor 1 than those in the control group (P < 0.05).  相似文献

Chloroform formation, balance, and discharge were estimated in a kraft pulp mill that used an elemental chlorine-free (ECF) bleaching line with chlorine dioxide as well as an effluent treatment process. This was achieved by collecting and analyzing discharge water and air samples that contained measurable levels of chloroform. Chloroform formation in ECF bleaching of hardwood oxygen-delignified kraft pulp (LOKP) was estimated to be 2.07–5.34g/pulp air-dried ton (adt), and approximately 30% of the chloroform produced was discharged to bleaching effluents. Chloroform in the effluents was not decomposed by activated sludge, and more than 97% was emitted to the air by volatilization. It is suggested that chloroform formation in Japanese LOKP bleaching mills can be decreased to 16–42t/year with a production of 8 million adt pulp by introducing ECF bleaching into all mills. This is a considerable fall from the chloroform discharge from ECF and chlorine bleaching mills in Japan that was estimated to be approximately 1000t for the 1999 fiscal year.  相似文献
