We genotyped strawberry cultivars by two newly selected and two previously reported SSR markers. All four markers produced interpretable electropherograms from 75 accessions consisting of 72 Fragaria × ananassa cultivars or lines and three octoploid Fragaria species accessions. These SSR markers were highly polymorphic; in particular, one of the newly developed markers, FxaHGA02P13, was capable of distinguishing all of the accessions except for a mutant strain that was derived from another accession in the set. When two markers were combined, all 48 full-sib individuals could be distinguished. Fingerprinting patterns were reproducible between multiple samples, including the leaves, sepals, and fruit flesh of the same accession. Principal-coordinate analysis of the 75 accessions detected several groups, which reflect taxon and breeding site. Together with other available markers, these SSR markers will contribute to the management of strawberry genetic resources and the protection of breeders’ rights.  相似文献

A rapid procedure for determining tuber maturity and processing quality using a blood glucose monitor is outlined. The procedure is simple to use, inexpensive and gives acceptable percent glucose and sucrose rating (SR) values for commercial applications. The procedure has been used successfully for determining the best time to sell commercial quantities of stored tubers (cvs. Norchip, Niska) for processing as chips in Alberta.  相似文献

Ascaridia dissimilis, a roundworm in turkeys, has been noted with increased frequency in commercial turkeys. Because infected turkeys can shed A. dissimilis ova in their feces, the potential exists for the external surface of turkey eggshells to be contaminated with A. dissimilis ova. The objectives of this study were to determine the presence of and recover A. dissimilis ova on the external surface of the turkey egg. In Experiment 1, turkey eggs were collected from naturally infected flocks, and eggs were processed by a sodium hydroxide procedure to recover any A. dissimilis ova on the external egg surface. In Experiment 2, the external surface of the turkey eggs was inoculated with 116 A. dissimilis ova/g feces, and eggshells were sampled every 3 days until 28 days of incubation to assess the recovery of A. dissimilis ova from the eggshell. In Experiment 1, of the 36 eggs examined from a flock naturally infected with A. dissimilis, one egg had an A. dissimilis ovum on its external eggshell surface. Experiment 2 demonstrated that A. dissimilis ova can be recovered from the external egg surface after a 28-day incubation period in the incubator. Ova recovery declined from an average of 62 A. dissimilis ova/turkey egg at day 2 of incubation to an average of 3 A. dissimilis ova/turkey egg at day 28 of incubation.  相似文献

ABSTRACT: A facile synthesis of Tyrian purple, a valuable purple dye derived from gastropod mollusks in ancient times, has been accomplished. Tyrian purple, 6,6'-dibromoindigo, was easily obtained by three steps of reactions from the commercially available 6-bromoindole, on the basis of a biosynthetic pathway of indigo and biological precursors of the purple. Iodination of 6-bromoindole, followed by acetoxylation with silver acetate in acetic acid, afforded 6-bromo-3-acetoxyindole, whose alkaline hydrolysis accompanying air oxidation produced Tyrian purple.  相似文献

The aim of the present study was to locate Ski protein, a product of cellular protooncogene c-ski, in rat ovaries in order to predict the possible involvement of Ski in follicular development and atresia. First, expression of c-ski mRNA in the ovaries of adult female rats was confirmed by RT-PCR. Then, ovaries obtained on the day of estrus were subjected to immunohistochemical analysis for Ski and proliferating cell nuclear antigen (PCNA) in combination with terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL). Ski was expressed in granulosa cells that were positive for TUNEL, but negative for PCNA, regardless of the size of follicles. Expression of Ski in TUNEL-positive granulosa cells, but not in PCNA-positive granulosa cells, was also verified in immature hypophysectomized rats having a single generation of developing and atretic follicles by treatment with equine chorionic gonadotropin. These results indicate that Ski is profoundly expressed in the granulosa cells of atretic follicles, but not in growing follicles, and suggests that Ski plays a role in apoptosis of granulosa cells during follicular atresia.  相似文献

The possible relationship between myofiber type composition and adipose tissue development in skeletal muscle in vivo has been suggested. Recent evidence indicated that satellite cells are multipotent cells that can undergo not only myogenic, but also adipogenic differentiation. In the present study, rat satellite cells were isolated from soleus, back, extensor digitorum longus, tibialis anterior and quadriceps muscles, and their adipogenic potentials were compared by culturing them under adipogenic conditions in vitro. Cells from soleus muscle exhibited the highest adipogenic potential as judged from Oil Red-staining and immunocytochemical C/EBPalpha-staining. The adipogenic potential of satellite cells was positively correlated with type I myofiber distribution in the corresponding muscle of origin. These results demonstrated that the adipogenic potential of satellite cells differs according to the muscle of origin and suggested that its possible correlation to type I myofiber distribution may account for preferential adipose tissue development in slow oxidative muscles.  相似文献

We have previously produced human growth hormone (hGH) transgenic (TG) rats that show low circulating levels of both hGH and endogenous rat GH. Although body length of the TG rats is normal, they develop hyperphagia and severe obesity. The present study was undertaken to elucidate the causes of hyperphagia in the TG rats by focusing on temporal changes in plasma ghrelin levels and hypothalamic neuropeptide Y (NPY) contents. In both wild-type (WT) and TG rats, the highest value of plasma ghrelin levels was observed just before the dark phase, and thereafter plasma ghrelin levels were maintained higher in the TG than WT rats. Although NPY contents also showed the peak level just before the dark phase in both the arcuate (ARC) and paraventricular nuclei (PVN) of the hypothalamus, the values in the ARC, but not the PVN, of the TG rats was always lower than those of the WT rats, suggesting increased transport of NPY from the ARC to PVN in the TG rats. In addition, treatment with antagonists for Y1 and Y5 receptors for NPY reduced food intake much more effectively in the TG than WT rats. Intermittent treatment with recombinant hGH for a week significantly decreased food consumption, adipose tissue weight and plasma triglyceride concentrations in the TG rats. These results suggest that, in the TG rats, insufficiency in circulating GH stimulates the ghrelin-NPY system with a resultant increase in food intake.  相似文献

To detect allergen-specific IgE in dogs with allergic diseases, we developed a recombinant canine high affinity IgE receptor α chain (FcεRIα)-based IgE detection system. Using the recombinant protein of canine FcεRIα expressed by an Escherichia coli expression system, we could detect house dust mite (Dermatophagoides farinae) allergen-specific IgE in sera from dogs naturally and experimentally sensitized to this allergen with ELISA and western blotting. The IgE binding activity of recombinant canine FcεRIα on ELISA was impaired by heat treatment of these sera. The specificity of this recombinant canine FcεRIα-based IgE detection system was confirmed by inhibition assays with canine IgE. The recombinant canine FcεRIα-based IgE detection system established in this study offers an alternative tool to measure allergen-specific IgE in dogs.  相似文献

The absorption and dynamics of Cs in crops can be measured and examined using not only 137Cs but also 133Cs, a stable isotope which can be analyzed in a regular laboratory. When 133Cs is used, however, the concentration of added 133Cs must be set higher than that in the soil solution due to the detection sensitivity of inductively coupled plasma optical emission spectrometry (ICP-OES). There is concern that such high Cs concentrations may adversely affect crops and interfere with Cs dynamics. Using soybean as a test product, we first examined the initial growth of soybean cultivated under exposure to solutions with different 133Cs concentrations. In combination with each 133Cs concentration, two potassium (K) concentrations were tested. Results show that at both K concentrations, growth was inhibited under 133Cs concentrations higher than 1 × 10?3 mol L?1. Soybeans cultivated at 133Cs concentrations higher than 1 × 10?6 mol L?1 can be subject to ICP-OES for measuring 133Cs concentrations in the plant tissue. Next, soybeans cultivated under different 133Cs concentrations throughout growth were examined for Cs distribution in the plants. The distribution patterns of Cs in soybean plants cultivated at 133Cs concentrations equivalent to those found in the soil were almost identical to those in plants cultivated at measurable 133Cs concentrations (1 × 10?6–1 × 10?4 mol L?1) as measured with ICP-OES. Therefore, in our investigation, 133Cs distribution in soybean plants treated with up to 1 × 10?4 mol L?1 was considered equivalent to that in plants cultivated in the field.  相似文献
