We provide an overview of both traditional and innovative control tools for management of three Xylosandrus ambrosia beetles (Coleoptera: Curculionidae: Scolytinae), invasive species with a history of damage in forests, nurseries, orchards and urban areas. Xylosandrus compactus, X. crassiusculus and X. germanus are native to Asia, and currently established in several countries around the globe. Adult females bore galleries into the plant xylem inoculating mutualistic ambrosia fungi that serve as food source for the developing progeny. Tunneling activity results in chewed wood extrusion from entry holes, sap outflow, foliage wilting followed by canopy dieback, and branch and trunk necrosis. Maintaining plant health by reducing physiological stress is the first recommendation for long-term control. Baited traps, ethanol-treated bolts, trap logs and trap trees of selected species can be used to monitor Xylosandrus species. Conventional pest control methods are mostly ineffective against Xylosandrus beetles because of the pests’ broad host range and rapid spread. Due to challenges with conventional control, more innovative control approaches are being tested, such as the optimization of the push–pull strategy based on specific attractant and repellent combinations, or the use of insecticide-treated netting. Biological control based on the release of entomopathogenic and mycoparasitic fungi, as well as the use of antagonistic bacteria, has yielded promising results. However, these technologies still require validation in real field conditions. Overall, we suggest that management efforts should primarily focus on reducing plant stress and potentially be combined with a multi-faceted approach for controlling Xylosandrus damage.

In 11 rain‐fed arable soils of the Potohar plateau, Pakistan, the amounts of microbial‐biomass C (Cmic), biomass N (Nmic), and biomass P (Pmic) were analyzed in relation to the element‐specific total storage compartment, i.e., soil Corg, Nt, and Pt. The effects of climatic conditions and soil physico‐chemical properties on these relationships were highlighted with special respect to crop yield levels. Average contents of soil Corg, Nt, and Pt were 3.9, 0.32, and 0.61 mg (g soil)–1, respectively. Less than 1% of Pt was extractable with 0.5 M NaHCO3. Mean contents of Cmic, Nmic, and Pmic were 118.4, 12.0, and 3.9 µg (g soil)–1. Values of Cmic, Nmic, Pmic, soil Corg, and Nt were all highly significantly interrelated. The mean crop yield level was closely connected with all soil organic matter– and microbial biomass–related properties, but showed also some influence by the amount of precipitation from September to June. Also the fraction of NaHCO3‐extractable P was closely related to soil organic matter, soil microbial biomass, and crop yield level. This reveals the overwhelming importance of biological processes for P turnover in alkaline soils.  相似文献

This study was designed to compare the quality of liquid‐stored buffalo bull spermatozoa in soya lecithin based extender Bioxcell® (BIOX), milk (MILK), tris‐citric egg yolk (TEY) and egg yolk‐citrate (EYC) extender at 5°C. Semen was collected from five Nili‐Ravi buffalo (Bubalus bubalis) bulls of 6–7 years of age with artificial vagina over a period of 3 weeks (two consecutive ejaculates once in a week). Semen ejaculates having more than 60% motility were pooled, split into four aliquots, diluted (37°C; 10 × 106 motile spermatozoa/ml), cooled from 37 to 5°C in 2 h (0.275°C/min) and stored for 5 days. Sperm motility, viability, plasma membrane integrity (PMI) and normal acrosomal ridge were studied at first, third and fifth day of storage. Higher values of progressive sperm motility (%), sperm viability (%), sperm PMI (%) and normal apical ridge (%) were observed in BIOX, MILK and TEY extenders at first, third and fifth day of storage than EYC extender. Progressive sperm motility, sperm viability and sperm PMI in BIOX® extender were not different from MILK and TEY extenders at 1st and third day storage period. However, at fifth day of storage, the values for these parameters remained significantly higher (p < 0.05) in BIOX® compared with MILK, TEY and EYC extenders. At fifth day of storage, the semen quality parameters for Bioxcell® were comparable to those with MILK and TEY extenders at third day of storage. In conclusion, motility, viability and PMI of buffalo bull spermatozoa remained similar in Bioxcell®, milk and TEY extender at first and third days of storage at 5°C. Yet, the values for the aforementioned parameters in Bioxcell® were higher compared with milk, TEY and EYC extender at fifth day of storage at 5°C.  相似文献

Blue jays learned to respond differentially to the presence or absence of Catocala moths in slides. This detection of the moths by the jays was affected by the background upon which the moth was placed and its body orientation, thus providing an objective measure of crypticity. Rodents are sources of many zoonotic pathogens that are of public health concern. This study investigated bacterial pathogens and assessed their antimicrobial resistance (AMR) patterns in commensal rodents in Qatar. A total of 148 rodents were captured between August 2019 and February 2020, and blood, ectoparasites, and visceral samples were collected. Gram-negative bacteria were isolated from the intestines, and blood plasma samples were used to detect antibodies against Brucella spp., Chlamydophila abortus, and Coxiella burnetii. PCR assays were performed to detect C. burnetii, Leptospira spp., Rickettsia spp., and Yersinia pestis in rodent tissues and ectoparasite samples. Antimicrobial resistance by the isolated intestinal bacteria was performed using an automated VITEK analyzer. A total of 13 bacterial species were isolated from the intestine samples, namely Acinetobacter baumannii, Aeromonas salmonicida, Citrobacter freundii, Citrobacter koseri, Enterobacter aerogenes, Enterobacter cloacae, Escherichia coli, Hafnia alvei, Klebsiella pneumoniae, Providencia stuartii, Proteus mirabilis, Pseudomonas aeruginosa, and Salmonella enterica. The majority of them were E. coli (54.63%), followed by P. mirabilis (17.59%) and K. pneumoniae (8.33%). Most of the pathogens were isolated from rodents obtained from livestock farms (50.46%), followed by agricultural farms (26.61%) and other sources (22.94%). No antibodies (0/148) were detected against Brucella spp., C. abortus, or C. burnetii. In addition, 31.58% (6/19) of the flea pools and one (1/1) mite pool was positive for Rickettsia spp., and no sample was positive for C. burnetii, Leptospira spp., and Y. pestis by PCR. A total of 43 (38%) bacterial isolates were identified as multidrug resistant (MDR), whereas A. salmonicida (n?=?1) did not show resistance to any tested antimicrobials. Over 50% of bacterial MDR isolates were resistant to ampicillin, cefalotin, doxycycline, nitrofurantoin, and tetracycline. The presence of MDR pathogens was not correlated with rodent species or the location of rodent trapping. Seven (11.86%) E. coli and 2 (22.2%) K. pneumoniae were extended-spectrum beta-lactamases (ESBL) producers. These findings suggest that rodents can be a source of opportunistic bacteria for human and animal transmission in Qatar. Further studies are needed for the molecular characterization of the identified bacteria in this study.

Nitrification inhibitors are widely used in agriculture to mitigate nitrous oxide(N2O)emission and increase crop yield.However,no concrete information on their mitigation of N2O emission is available under soil and environmental conditions as in Pakistan.A field experiment was established using a silt clay loam soil from Peshawar,Pakistan,to study the effect of urea applied in combination with a nitrification inhibitor,nitrapyrin(2-chloro-6-tri-chloromethyl pyridine),and/or a plant growth regulator,gibberellic acid(GA_3),on N2O emission and the nitrogen(N)uptake efficiency of maize.The experimental design was a randomized complete block with five treatments in four replicates:control with no N(CK),urea(200 kg N ha-1)alone,urea in combination with nitrapyrin(700 g ha-1),urea in combination with GA_3(60 g ha-1),and urea in combination with nitrapyrin and GA_3.The N2O emission,yield,N response efficiency,and total N uptake were measured during the experimental period.The treatment with urea and nitrapyrin reduced total N2O emission by 39%–43%and decreased yield-scaled N2O emission by 47%–52%,relative to the treatment with urea alone.The maize plant biomass,grain yield,and total N uptake increased significantly by 23%,17%,and 15%,respectively,in the treatment with urea and nitrapyrin,relative to the treatment with urea alone,which was possibly due to N saving,lower N loss,and increased N uptake in the form of ammonium;they were further enhanced in the treatment with urea,nitrapyrin,and GA_3 by 27%,36%,and 25%,respectively,probably because of the stimulating effect of GA_3 on plant growth and development and the reduction in biotic and abiotic stresses.These results suggest that applying urea in combination with nitrapyrin and GA_3 has the potential to mitigate N2O emission,improve N response efficiency,and increase maize yield.  相似文献

Sipholenol A is a natural sipholane triterpenoid isolated from the Red Sea sponge, Callyspongia siphonella. Previous studies showed the antimigratory and antiproliferative activities of the semisynthetic sipholenol A esters against breast cancer cell lines. This study investigated the effects of sipholenol A-4-O-3′,4′-dichlorobenzoate (SPA) on the growth, migration and invasion of diverse human breast cancer cells. Results showed that SPA inhibited the growth of the human breast cancer cells, MDA-MB-231, MCF-7, BT-474 and T-47D, in a dose-dependent manner. Immunofluorescent analysis showed that SPA significantly reduced Ki-67-positive cells in MDA-MB-231 cells. Flow cytometry and Western blot analyses revealed that SPA treatment suppressed MDA-MB-231 cell growth by inducing cell cycle arrest at the G1 phase. In addition, SPA suppressed breast cancer cell migration, invasion and decreased Brk and FAK activation in a dose-dependent manner. Molecular docking study suggested a perfect fitting at the FAK’s FERM domain, inhibiting the main autophosphorylation site, Y397, which was further confirmed by Western blot analysis. Most known small molecule FAK inhibitors target the kinase domain, creating several off-target side effects. The in vivo studies showed that SPA treatment suppressed breast tumor growth and Ki-67, CD31, p-Brk and p-FAK expression in orthotopic breast cancer in nude mice. In conclusion, SPA inhibited the growth, invasion and migration of breast cancer cells possibly via deactivating Brk and FAK signaling, suggesting good potential for therapeutic use to control invasive breast cancer.  相似文献

Marine natural products (MNPs) are recognized for their structural complexity, diversity, and novelty. The vast majority of MNPs are pharmacologically relevant through their ability to modulate macromolecular targets underlying human diseases. Angiogenesis is a fundamental process in cancer progression and metastasis. Targeting angiogenesis through selective modulation of linked protein kinases is a valid strategy to discover novel effective tumor growth and metastasis inhibitors. An in-house marine natural products mini-library, which comprises diverse MNP entities, was submitted to the Lilly’s Open Innovation Drug Discovery platform. Accepted structures were subjected to in vitro screening to discover mechanistically novel angiogenesis inhibitors. Active hits were subjected to additional angiogenesis-targeted kinase profiling. Some natural and semisynthetic MNPs, including multiple members of the macrolide latrunculins, the macrocyclic oxaquinolizidine alkaloid araguspongine C, and the sesquiterpene quinone puupehenone, showed promising results in primary and secondary angiogenesis screening modules. These hits inhibited vascular endothelial growth factor (VEGF)-mediated endothelial tube-like formation, with minimal cytotoxicity at relevant doses. Secondary kinase profiling identified six target protein kinases, all involved in angiogenesis signaling pathways. Molecular modeling and docking experiments aided the understanding of molecular binding interactions, identification of pharmacophoric epitopes, and deriving structure-activity relationships of active hits. Marine natural products are prolific resources for the discovery of chemically and mechanistically unique selective antiangiogenic scaffolds.  相似文献
