A mongrel dog, aged 2 years, was found to have only a small number of sperm, immobilization of all sperm, and many sperm agglutinations in its ejaculates, and scrotal palpation revealed a small nodule in the left cauda epididymis. Addition of the dog's seminal plasma or serum to the semen of 2 normal dogs caused immobilization and agglutination of their sperm. Histological examination showed that the nodule was a sperm granuloma. Many lymphocytes were seen in the stroma around the sperm granuloma. Anti-sperm antibodies are presumed to be present in the semen and serum of the asthenozoospermic dog.  相似文献

The plant eukaryotic translation-initiation factors eIF4E and eIF(iso)4E play key roles in infection by plant RNA viruses, especially potyviruses. Mutations in the genes that encode these factors reduce susceptibility to the viruses. In the amphidiploid plant tobacco (Nicotiana tabacum L.), eIF4E1-S deletion mutants resist Potato virus Y (PVY), but resistance-breaking strains (RB-PVY) have appeared. In an earlier study, we demonstrated that the loss-of-function of eIF(iso)4E-T reduces susceptibility to RB-PVY. Here, we show that simultaneous inhibition of eIF4E1-S and eIF(iso)4E-T synergistically confers enhanced resistance to both PVY and RB-PVY without host growth or development defects. PVY symptoms and accumulation in a tobacco line lacking eIF4E1-S were detected at 14 days post-inoculation (dpi) and RB-PVY symptoms in lines without functional eIF(iso)4E-T were observed at 24 dpi. RB-PVY emerged in a PVY-infected tobacco line lacking eIF4E1-S. In contrast, lines without functional eIF4E1-S and eIF(iso)4E-T were nearly immune to PVY and RB-PVY, and little accumulation of either virus was detected even at 56 dpi. Thus, the lines will be promising for PVY-resistance breeding. This study provides a novel strategy to develop tobacco highly resistant to PVY and RB-PVY, and insights into the mechanisms responsible for high-level resistance.  相似文献

It has been suggested that soil-thawing and snow-melting are critical triggers for vigorous emissions of nitrous oxide (N2O) from soils in cold regions. However, because soil freezing is affected by air temperature and snow cover, accurate predictions that estimate subsequent emissions of this important greenhouse gas are difficult to make. In this study, we measured in situ soil gas N2O and oxygen (O2) concentrations at two experimental sites in northern Japan over the period of a year, from November 2008 to October 2009, to clarify the factors stimulating N2O production in soil at low temperatures. The sites were N-fertilized bare arable lands with different soil frost depths and snowmelt rates, according to the snow cover management imposed. Winter-to-spring net N2O fluxes, ranging from −0.10 to 1.95 kg N2O-N ha−1, were positively correlated with the annual maximum soil frost depth (ranging from 0.03 to 0.41 m; r = 0.951***). In the plots with deeper maximum soil frost, winter-to-spring N2O fluxes represented 58% to 85% of the annual values. Soil N2O production was stimulated when the soil frost depth was greater than 0.15 m or the daily mean soil temperature at 0.05-m depth was below −2.0 °C. In the soil with the greatest frost depth, soil gas N2O concentrations at the depth of 0.10 m peaked at 46 ppm when soil gas O2 concentrations fell down to 0.12 m3 m−3 under soil temperature below 0.0 °C. Snowmelt acceleration had no stimulating effect on N2O production in the soil during the winter-to-spring period.  相似文献

Heat shock proteins are evolutionarily highly conserved polypeptides that are produced under a variety of stress conditions to preserve cellular functions. A major antigen of tubercle bacilli of 65 kilodaltons is a heat shock protein that has significant sequence similarity and cross-reactivity with antigens of various other microbes. Monoclonal antibodies against this common bacterial heat shock protein were used to identify a molecule of similar size in murine macrophages. Macrophages subjected to various stress stimuli including interferon-gamma activation and viral infection were recognized by class I-restricted CD8 T cells raised against the bacterial heat shock protein. These data suggest that heat shock proteins are processed in stressed host cells and that epitopes shared by heat shock proteins of bacterial and host origin are presented in the context of class I molecules.  相似文献

Atherosclerosis is a vascular disorder involving inflammation, a narrowed vascular lumen in the entire tunica intima, and reduced elasticity of the arterial wall. It has been found that Hsp60 from Chlamydia pneumoniae, an obligate bacterial pathogen associated with atheroma lesions, mimics human Hsp60, thereby causing attacks by immune cells on stressed endothelial cells expressing endogenous Hsp60 on their surface. Furthermore, Hsp60 from C. pneumoniae has been shown to promote the growth of vascular smooth muscle cells (VSMCs). To explore probes that can be used for studying signal transduction elicited by the chlamydial Hsp60, we have tested several natural products for their inhibitory actions on the Hsp60-induced proliferation of rat arterial smooth muscle cells. Sesamol, vanillyl alcohol, and trans-ferulic acid exhibited moderate inhibitory actions on the Hsp60-induced cell proliferation; zerumbone, humulene, and caryophylene effectively inhibited it at low concentrations with IC(50) values of 529, 122, and 110 nM, respectively. The results indicated that the 11-membered alicyclic ring is favorable for interactions with receptors involved in the Hsp60-induced VSMC proliferation.  相似文献

This paper investigated the components, especially aldehydes, in the fume condensates from four kinds of cooking oil using ultraviolet (UV) spectrometry and gas chromatography-mass spectrometry (GC-MS). It was observed that there was a great change of the UV absorption spectra from the results of the unheated oil to the results of the fume after heat treatment (190-200, 230-240, and 270-280 degrees C). There was a strong peak within the wavelength range of 260-270 nm in each condensate sample. From the GC-MS results, it was tentatively deduced that there were some 2,4-dialkylenaldehydes and other conjugated compounds in the condensates. The results showed there were large amounts of hexanal and 2-heptenal in the cooking oil fume and that the total aldehyde peak areas of the condensates from four kinds of oil were around 30-50% of the total peak area at 270-280 degrees C.  相似文献

Phosphate buffer-extractable organic nitrogen (PEON), which can be extracted from soil with a 67-mM neutral phosphate buffer solution, is considered to be a major source of mineralizable nitrogen (N) that is ubiquitous in soil. We have developed a Western blot analysis and enzyme-linked immune sorbent assay (ELISA) method for determining PEON. First, anti-PEON antibody (IgG) from a rabbit was produced using purified PEON from a volcanic ash soil. PEON was detected as a ladder-like staining pattern in the high-molecular-weight region and as a single band under 14 kDa in Western blot analysis using the anti-PEON IgG. In the ELISA assay, the anti-PEON IgG could specifically react with PEON irrespective of soil type and management. Furthermore, the absorbance at 450 nm in ELISA was highly correlated to the concentration of organic N (Norg) in phosphate extracts of different soils. We propose the immune assay using the anti-PEON IgG as a possible novel method for evaluating available N in soil.  相似文献

The effects of initial tree spacing on wood density at breast height were examined for 22-year-old Japanese larch (Larix kaempferi). The experiment involved the use of three plots with different initial tree spacing densities (300, 500, and 1000 trees/ha). For five trees selected from each plot, the total tree height, diameter at breast height, height to the base of the live crown, and crown diameter were measured. Ring width and wood density for individual growth rings were determined by X-ray densitometry. A mixed-effects model was applied for fitting the radial variation in wood density in relation to initial spacing. Models having various mean and covariance structures were tested in devising an appropriate wood density model. The model, consisting of the mean structure with quadratic age effects and heterogeneous first-order autoregressive covariance, was able to describe the radial variation in wood density. Closer spacing of trees (1000 trees/ha) resulted in a faster increase in wood density from the pith outward than for more widely spaced trees, indicating that initial tree spacing may influence the age of transition from juvenile to mature wood.  相似文献
