Transgenic mice are essential research tools in developmental biology studies. The 2A peptide allows multiple genes to be
expressed simultaneously at comparable levels in somatic cells, but there are no reports of it being used successfully in
germ cells. We constructed a Cre/loxP-based conditional vector containing the 2A peptide to significantly enhance the
expression of a reporter and target gene from a constitutive promoter in oocytes. Mice with a transgene insertion containing
the chicken β-actin promoter, floxed EGFP-polyA cassette, mCherry reporter, 2A peptide and target gene DNA methyltransferase
3A2 (Dnmt3a2) were crossed with TNAP- or Vasa-Cre mice to produce offspring, in which mCherry and DNMT3A2
proteins were highly expressed in oocytes upon Cre-mediated removal of EGFP-polyA. This novel transgenic mouse line based on

The core histone is composed of four proteins (H2A, H2B, H3 and H4). Investigation of the modification patterns of histones
is critical to understanding their roles in biological processes. Although histone modification is observed in multiple cells
and tissues, little is known about its function in spermatogenesis. We focused on the modification patterns of histone H4
during murine spermatogenesis. We demonstrated that the individual N-terminal sites of H4 show different modification
patterns during the differentiation of male germ cells. The methylation pattern varied depending on the residues that were
mono-, di-, or tri-methylated. All the H4 modifications were high during the meiotic prophase, suggesting that histone H4
modification plays an important role during this stage of spermatogenesis. Elongating spermatids showed increased acetylation
of histone H4, which may be associated with a histone-to-protamine substitution. Our results provide further insight into the
specific relationship between histone H4 modification and gene expression during spermatogenesis, which could help to
elucidate the epigenetic disorders underlying male infertility. 相似文献

ABSTRACT: To explore the potential utility of underutilized fish in the fish sauce industry, fish sauces were prepared from both raw and heat-induced meat of Japanese anchovy Engraulis japonica and rabbit fish Siganus fuscescens using wheat malt, and their quality aspects and sensory properties were assessed. Proximate composition of the fish meat represents protein as the major component (16.0–17.8%), other than moisture. Analyses of free amino acids, peptides including oligopeptides, and organic acids contents in fish sauces revealed suitability of both raw and heat-induced meat of the selected species in commercial fish sauce production. The variations in taste of raw meat fish sauces were species-specific, but the taste of fish sauces from heat-induced meat were similar. 相似文献
