We developed a model of 545 components (nodes) and 1259 interactions representing signaling pathways and cellular machines in the hippocampal CA1 neuron. Using graph theory methods, we analyzed ligand-induced signal flow through the system. Specification of input and output nodes allowed us to identify functional modules. Networking resulted in the emergence of regulatory motifs, such as positive and negative feedback and feedforward loops, that process information. Key regulators of plasticity were highly connected nodes required for the formation of regulatory motifs, indicating the potential importance of such motifs in determining cellular choices between homeostasis and plasticity.  相似文献

Among marine organisms, sponges are the richest sources of pharmacologically-active compounds. Stemming from a previous lead discovery program that gathered a comprehensive library of organic extracts of marine sponges from the off-shore region of Portugal, crude extracts of Erylus cf. deficiens collected in the Gorringe Bank (Atlantic Ocean) were tested in the innovative high throughput screening (HTS) assay for inhibitors of indoleamine 2,3-dioxygenase (IDO) and showed activity. Bioassay guided fractionation of the dichloromethane extract led to the isolation of four new glycolipids, named erylusamide A–D. The structures of the isolated compounds were established by 1D and 2D nuclear magnetic resonance (NMR) spectroscopy, high-resolution electrospray ionization mass spectrometry (HR-ESI-MS) and chemical derivatization. The metabolites shared a pentasaccharide moiety constituted by unusual highly acetylated d-glucose moieties as well as d-xylose and d-galactose. The aglycones were unprecedented long chain dihydroxyketo amides. Erylusamides A, B and D differ in the length of the hydrocarbon chain, while erylusamide C is a structural isomer of erylusamide B.  相似文献

Four enyne derivatives (1-4) and quercitrin were isolated during a bioassay-guided chromatographic separation of a methanolic extract of Erigeron apiculatus. Matricarialactone (1) and lachnophyllumlactone (2) showed a high fungitoxic activity against Pyricularia oryzae. Matricaria acid methyl ester (3) and lachnophyllum acid methyl ester (4) were, instead, less active.  相似文献

Liquors from rice husk autohydrolyis, containing xylooligosaccharides, other saccharides, and nonsaccharide compounds, were subjected to two selected processing schemes to increase the proportion of substituted xylooligosaccharides in refined liquors. Nanofiltration through a ceramic membrane with a molecular mass cutoff of 1000 Da allowed simultaneous concentration and purification; this latter derived from the preferential removal of monosaccharides and nonsaccharide compounds. When liquors were nanofiltered to achieve a volume reduction factor of 5 operating at a transmembrane pressure of 14 bar, 58.6% of the nonsaccharide components and 20.9-46.9% of monosaccharides were kept in retentate, in comparison with 92% of xylooligosaccharides and glucooligosaccharides. When nanofiltered liquors were subjected to double ion-exchange processing, a final product with a nonsaccharide content near 9 kg/100 kg of nonvolatile components was obtained at a yield of 10.90 kg/100 kg oven dry rice husks. Alternatively, when nanofiltered liquors were subjected to ethyl acetate extraction and further double ion-exchange processing, a purified product with a nonsaccharide content of 5.66 kg/100 kg of nonvolatile components was obtained at a yield of 9.94 kg/100 kg oven dry rice husks. The nonsaccharide components remaining in the final concentrate were mainly made up of phenolic and nitrogen-containing compounds.  相似文献

In Reserva Natural de Castro Marim e Vila Real de Santo António (SE of Portugal), most of the salt marshes and saltpans are abandoned contributing to their degradation and, consequently, alteration of some ecological conditions. Rehabilitation of these areas by a Salicornia crop can contribute to their economic and environmental improvement by stimulation of biogeochemical processes and biomass commercialization. However, the development of agro-techniques adjusted to species and variable environmental conditions from Mediterranean are needed in order to improve the Salicornia crop. This study aimed to evaluate: i) potential use of seedlings from greenhouse in the field cultivation; and ii) survival, growth and yield of Salicornia under shading and different salinity conditions of the soils and irrigation waters. Autochthonous Salicornia species (S. ramosissima and S. patula) were tested. Germination tests under controlled conditions and in situ mesocosm assays were carried out. Assays were realized with a low density of seedlings from natural conditions and greenhouse, and soils and waters from estuary both with different salinities. Intermediate and high salinities (25–45?dS/m) affected only S. patula germination. In general, Salicornia growing in soils with low salinity had great fresh biomass accumulation independently of seedling source (from natural conditions and greenhouse) and/or growing season. The shading seems to improve the fresh biomass accumulation and yield. Under Mediterranean conditions, the transplantation of Salicornia from greenhouse to the field was a promising technique, independently of the salinity conditions. This study demonstrated that the abandoned and saline areas can be rehabilitated by a sustainable crop of Salicornia.  相似文献
