The effects of concentrations of Rosmarinus officinalis essential oil (EO) and nisin (N) as well as temperature and storage time on growth of Streptococcus iniae and Lactococcus garvieae in Oncorhynchus mykiss were evaluated. According to analysis of the rosemary EO, the 1, 8-cineol and α-pinene were the predominant components. The growth of S. iniae was significantly decrease by EO concentrations at 4 ºC. For L. garvieae, the viable count was significantly inhibited by EO and N singly and in combinations, incubated at bath storage of 0.25 and 0.5 µg/mL proved insufficient to act against S. iniae and L. garvieae. The combinations of the rosemary EO at 0.0015% with N at 0.5 µg/mL showed stronger antimicrobial effect against two bacterial than the rosemary EO at 0% but lower than the combination with N at 0.5 µg/mL and EO at 0.045% which in turn was lower than of the rosemary EO at 0.135%. In its turn, rosemary EO showed lower antimicrobial activity than its combinations with N, which showed a bactericidal effect against the pathogens. The best inhibitory effects of EO in combinations with N for two bacterial were obtained at combinations of EO=0.135% and N=0.5 µg/mL.  相似文献

Understanding the volatile profile of melon is an ever increasingly important task for breeders for enhancing their flavour component. For this purpose, F1 progeny and parental lines were analysed using headspace-solid phase microextraction gas chromatography–mass spectrometry-based analysis. Diallel analysis showed that most of the investigated compounds were controlled via non-additive genetic effects with general combining ability to specific combining ability ratios of less than 0.4. High heterosis results were observed in alcohols such as ethanol and octanol in both rind and mesocarp samples and benzylalcohol in the rind. It was also noted that the ester-based compounds with high heterosis values in mesocarp samples differed from those with heterosis in rind samples. These observations further emphasize the non-additive control of these compounds. It can be concluded that non-additive genetic control is strongly governing the genetic profile within the studied progeny. This finding indicated that dudaim lines as a valuable genetic material enhanced the volatile profile of odourless commercial types of ameri and inodorus melon.  相似文献

The goal of the present work was to test if there is genetic variability between precocious and superprecocious cycle maize genotypes in terms of grain productivity and energy profile. Data from two separate experiments were used. An experiment was conducted with 36 precocious cycle genotypes and another experiment was conducted with 22 superprecocious cycle genotypes. The following variables were measured: grain productivity (PROD), ethereal extract (EE), starch (ST), amylose (AML), and nitrogen-corrected apparent metabolizable energy (AMEn). For all variables, basic statistics and analysis of variance was performed. For each experiment, the genotypic correlation matrix was calculated and the multicollinearity was evaluated. Then, the Mahalanobis generalized distance matrix was calculated, the clustering of the genotypes was performed and the averages of the groups formed were compared, separately for each experiment. There was genetic divergence between precocious genotypes, as well as between superprecocious genotypes, especially in terms of grain PROD, EE concentration, and AML concentration. Groups of more productive genotypes presented lower EE and higher AML grain concentrations. The results of the present study indicated that it is possible to plan crossings between groups of genotypes in terms of PROD, EE and AML, with the goal of maximizing heterosis.  相似文献

The role of cropping systems practices in agronomic biofortification programs with the aim of increasing micronutrient density in food plants has to be clarified. In these field experiments, the effect of four preceding crops, i.e., sunflower (Heliantus annus L. cv. Allstar), Sudan grass (Sorghum bicolor L. cv. Speed Feed), clover (Trifolium pratense L.), and safflower (Carthamus tinctorius L. cv. Koseh-e-Isfahan), on the total amino acids (AA) and dissolved organic carbon (DOC) concentration in rhizosphere soil solution and grain Zn content of successive wheat (Triticum aestivum cvs. Back Cross and Kavir) was investigated during 2009–2010 and 2010–2011 growing seasons. A fallow treatment was also considered as the control. In both growing seasons, preceding crops increased the concentrations of AA and DOC in the soil solution in comparison with the fallow control treatment; although the magnitude of this increase varied upon the preceding crop type and wheat cultivar. In general, clover and sunflower had greater effect on increasing soil solution DOC probably due to higher decomposability of their litter residues in soil. Preceding crops increased the total AA concentration, on average, by 45.9 % for the first year and 10.8 % for the second year. The preceding sorghum and clover had the highest and lowest influence on the concentration of AA in wheat rhizosphere soil solution, respectively. The preceding crops increased grain wheat Zn concentration and content over the fallow control treatment, although this effect was dependent on the crop type. For “Back Cross”, a positive and significant correlation was found between grain Zn concentration and soil solution DOC concentration (r?=?0.60, P?<?0.05) and particularly AA (r?=?0.76, P?<?0.001), while no such correlation was found for “Kavir”. At the second growing season, the concentration of AA in the rhizosphere of Back Cross was greater than that of Kavir, probably due to higher release of these compounds from the roots. According to the results, the preceding crop significantly affect grain Zn density of the successive wheat, that is, at least in part, by releasing soluble organic ligands into soil solution.  相似文献

Background:Bispecific antibodies represent an important class of mAbs, with great therapeutic potentials due to their ability to target simultaneously two distinct epitopes. The generation of functional bispecific antibodies with the highest possible yields is particularly critical for the production of these compounds on industrial scales. Anti- CD3 × CD19 bsAb is a bispecific T-cell engager (BiTE) currently used for treating ALL. Herein, we have tried to optimize the expression level of this antibody in mammalian hosts. Methods:WPRE sequence was incorporated at the 3’ end of the expression cassette. This modification resulted in a notable about two-fold increase in the expression of the bsAb in the Expi293 cell line. Results & Conclusion:Follow-up flow cytometry analysis demonstrated the binding properties of the produced antibody at acceptable levels, and in vitro bioactivity assays showed that this product is potent enough for targeting and destroying CD19-positive cells. Our findings show that WPRE enhances the expression of this type of bispecific mAbs in HEK-293 family cell lines. This approach can be used in biopharma industry for the mass production of anti-CD3 × CD19 bispecific antibody. The occurrence of mycobacteriosis caused by Mycobacterium marinum in a commercial breeding farm of bullfrogs (Rana catesbeiana) in Rio de Janeiro, Brazil is described. Ten animals presented skin lesions on the head and extremities. These and 38 other asymptomatic adult animals from various tanks were killed and at necropsy disseminated granulomatous lesions were observed in the 10 clinically affected animals and in 16 (42.1%) of the asymptomatic frogs. Acid-fast bacilli were observed in all smears of the 10 symptomatic frogs and in all but one from the 16 asymptomatic animals with visceral lesions. Ten samples from the 25 positive animals were randomly selected for culture which yielded four isolates of fast-growing (<7 days) mycobacteria. Those purified isolates were characterised by biochemical traditional means as M. marinum. Identification of the strains was confirmed using reverse-phase high-performance liquid chromatography and a polymerase chain reaction (PCR) restriction enzyme analysis assay. It is suggested that M. marinum is an important agent of granulomatous disease in bullfrogs and that infected animals, even when asymptomatic, could act as reservoirs spreading the disease and contaminating other frogs in the farm.  相似文献

Population growth and production of the freshwater rotifer, Euchlanis dilatata, were determined after being fed 1 of 12 diets: Scenedesmus quadricauda (S), Chlorella vulgaris (C), baker's yeast (Saccharomyces cerevisiae) (Y), alfalfa meal (Medicago spp.) (A), and eight combinations of S+C, S+Y, C+Y, S+C+Y, S+A, C+A, S+C+A, and Y+A (mixture in the same ratio by equal nitrogen weight). Results showed that both microalgal and alfalfa meal alone and in combination with Y had a significantly higher (P < 0.05) population growth and production compared with Y alone. The mean maximum E. dilatata population density fed on S, S+Y, C, C+Y, S+C, S+C+Y, Y, A, S+A, C+A, S+C+A, and Y+A diets were 65 ± 7.5 (mean ± SE), 124.6 ± 17.2, 115.8 ± 7.9, 41.5 ± 7.9, 131 ± 14.1, 93.5 ± 8.7, 14.5 ± 7.6, 129.5 ± 18.5, 255.0 ± 20.5, 180 ± 30.5, 124.5 ± 25.4, and 61.5 ± 24.5 individuals/mL, respectively. Correspondingly, the mean population growth rates (K) through the 16‐d culture period were 0.29, 0.40, 0.45, 0.27, 0.34, 0.41, 0.26, 0.51, 0.58, 0.59, 0.39, and 0.45/d, respectively. Results indicated that alfalfa meal in combination with microalgae and yeast could be used as primary diets for E. dilatata culture. This may reduce the costs of rotifer production.  相似文献

ABSTRACT

Salinity is one of the most important abiotic stresses limiting agricultural production worldwide. Here, we investigated the biometric, physiological and anatomical changes of Passiflora edulis plants propagated from seeds, cuttings and grafts and exposed to salt stress (0.3 – control and 6.3 dS m?1). Saline stress significantly reduced plant height, the number of leaves, total chlorophyll, stomatal conductance, shoot dry weight, total dry weight, total water consumption and root volume. Among the propagation methods evaluated, plants propagated from seeds were more sensitive to salinity, while plants propagated from cuttings showed smaller alterations when subjected to salinity. Similar behavior was observed for anatomical characteristics, since plants propagated from seeds presented more pronounced anatomical alterations, such as greater thicknesses of the leaf mesophyll, upper epidermis, lower epidermis, palisade parenchyma and spongy parenchyma and also was observed presence of calcium oxalate crystals in the form of druses in greater quantity in the root cortex, in comparison to plants propagated from cuttings. The approach used in this study allowed identifying the type of yellow passion fruit propagation (cuttings) that was best for physiological and anatomical adaptation to salt stress and will be potentially useful in programs for improvement of the crop.  相似文献
