The objective of this study was to evaluate the effect of retinol on the in vitro development of early embryos of cultured Bos indicus (Expt 1) to the blastocyst stage in medium simplex of optimization (KSOM) or sintetic fluid of oviduct (SOF) or co-cultured (Expt 2) with an oviduct cell monolayer (OCM) in KSOM or SOF. A total of 3149 cumulus-oocyte complexes obtained by aspirating follicles (2-5 mm diameter) from ovaries of slaughtered animals were selected for IVM and incubated in TCM 199 supplemented with 25 mM HEPES at 39 degrees C in air with 5% CO(2) and maximum humidity for 24 h. In vitro fertilization (IVF) was performed in modified defined medium (mDM) medium. Eighteen hours after IVF, cumulus cells were removed and presumptive zygotes were randomly allocated to the experimental groups. Zygotes cultured (Expt 1) in KSOM + retinol, KSOM, SOF + retinol and SOF were incubated in maximum humidity at 39 degrees C, 5% CO(2), 5% O(2) and 90% N(2). Zygotes co-cultured (Expt 2) in KSOM + retinol + OCM, KSOM + OCM, SOF + retinol + OCM and SOF + OCM were incubated at 39 degrees C, 5% CO(2). In both experiments media were partially changed 48 h after IVF and unfertilized ova were removed. Afterwards embryos were kept in culture or co-culture for further 9 days. In Expt 1, blastocyst rates (day 7) were 14.6% (KSOM + retinol), 15.8% (KSOM), 16.4% (SOF + retinol) and 15.9% (SOF). In Expt 2, the blastocyst rates (day 7) were 25.4% (KSOM + retinol + OCM) 14.2% (KSOM + OCM), 24.3% (SOF + retinol + OCM) and 15.9% (SOF + OCM). The same influence profile of retinol was observed in the formation of the expanded (day 9) and hatched (day 11) blastocysts. The results obtained in Expt 2 demonstrated that the addition of 0.28 microg/ml retinol to the embryo culture media used in this study had a significant (p < 0.05) positive effect on bovine early embryonic development, under the conditions tested, and can be used to enhance in vitro embryo production.  相似文献

The pregnancy rates obtained after the transfer of cryopreserved in vitro‐produced (IVP) embryos are usually low and/or inconsistent. The objective of this study was to evaluate the pregnancy rates of Holstein, Gyr and Holstein × Gyr cattle after the transfer of vitrified IVP embryos produced with X‐sorted sperm. Seventy‐two Gyr and 703 Holstein females were subjected to ovum pickup (OPU) sessions, followed by in vitro embryo production using semen from sires of the same breeds. Embryos (1636 Holstein, 241 Gyr and 1515 Holstein × Gyr) were exposed to forskolin for 48 h prior to vitrification. The pregnancy rate achieved with Gyr dam and sire was 46.1%, which was similar (p = 0.11) to that of Holstein dam and Gyr sire (40.3%). Crossing Gyr dams with Holstein sires resulted in a pregnancy rate of 38.9% and did not differ (p = 0.58) from the pregnancy rate obtained with the cross between Holstein dams and Gyr sires. The rate obtained with Holstein dam and sire was 32.5%. The average pregnancy rate was 36.6%, and no difference was found in the proportion of female foetuses (88.8%, in average) among breeds (p > 0.05). In conclusion, transfer of cryopreserved X‐sorted embryos represents an interesting choice for dairy cattle. Despite the small differences between pregnancy rates, we highlight the efficiency of this strategy for all of the racial groups studied.  相似文献

Samples from four market lots of hard red winter and soft red winter wheat containing sprout- and scab-damaged kernels were used to test a prototype single-kernel density micropycnometer. Fifteen kernels for each damage type and an equal number of healthy kernels were weighed to the nearest 0.01 mg, then measured for volume to the nearest 1.0 μL. Volume measurements for all kernels were performed three consecutive times with the micropycnometer, then kernels were evaluated for weight, size, moisture, and hardness using a Single Kernel Characterization System. The structure of the sampling plan and the goals of the study indicated that a mixed-model statistical analysis was needed. The fixed effects were wheat class and type of kernel, and the random effects included lot, the interaction of lot with class and kernel type, kernels within each lot, and repeated measures of single-kernel density. Results indicated that variability of the three measurements per kernel did not depend on type of kernel or class of wheat. The standard deviation for repeated density measurements was 0.0029 g/cm3. Kernel-to-kernel variability changed depending on the type of kernel; healthy and sprout-damaged kernels showed similar variability in density, whereas scab damaged kernels had a variance about four to five times higher. Type of kernel significantly affected mean density; healthy kernels averaged 1.28 g/cm3, sprout-damaged kernels averaged 1.19 g/cm3, and scab-damaged kernels averaged 1.08 g/cm3. Wheat class did not exert a significant influence on singlekernel density. Attempts to predict single-kernel density using kernel weight, size, moisture, and hardness found no relationships of practical importance.  相似文献

Twelve hard red winter wheat cultivars were grown at four locations in central Kansas to evaluate the role of foliar fungal diseases on wheat end-use quality in 1995. Disease was allowed to develop naturally on control plots and was controlled partially on plots treated with a systemic fungicide. After harvest, wheat samples were evaluated for the impact of the disease complex (leaf rust, tan spot, speckled leaf blotch) on physical grain quality, grain protein, milling properties, flour absorption, and peak mixing time. Data were analyzed using a mixed model to account for random (location and block) and fixed (cultivar and fungicide) effects. Location significantly influenced quality characteristics except kernel size and peak mixing time. The magnitudes of variation among random effects on all quality characteristics were larger for location than for the interactions between location × cultivar and location × fungicide. The fixed effects portion of the analysis revealed that the cultivar × fungicide treatment interaction significantly affected test weight, kernel protein, and flour absorption. Fungicide treatment resulted in significant increases in yield and kernel weight. Cultivar significantly affected all quality characteristics except kernel size and peak mixing time. Disease resistance exerted a significant influence on yield and test weight. The economic benefit associated with improved wheat quality from fungicide treatment was variety specific. Three cultivars (TAM 107, Karl 92, and Ike), which account for 50% of the 1997 planted wheat acres in Kansas, demonstrated positive improvements in test weight and protein in response to fungicide treatment.  相似文献

Phase-matched harmonic conversion of visible laser light into soft x-rays was demonstrated. The recently developed technique of guided-wave frequency conversion was used to upshift light from 800 nanometers to the range from 17 to 32 nanometers. This process increased the coherent x-ray output by factors of 10(2) to 10(3) compared to the non-phase-matched case. This source uses a small-scale (sub-millijoule) high repetition-rate laser and will enable a wide variety of new experimental investigations in linear and nonlinear x-ray science.  相似文献

OBJECTIVE: To describe clinical signs and results of treatment in cats with patellar luxation. DESIGN: Retrospective case series. ANIMALS: 42 cats in which patellar luxation had been diagnosed on the basis of results of palpation of the stifle joints. PROCEDURES: Degree of luxation was graded on a scale from 1 to 4, and severity of lameness was graded on a scale from 0 to 5. Radiographs of stifle joints were evaluated for signs of osteoarthritis. Long-term function was classified as poor, fair, good, or excellent. RESULTS: 34 cats had bilateral luxation and 8 had unilateral luxation. Only 7 (17%) cats had a history of trauma. Mean age of the cats was 3.3 years, and mean weight was 4.26 kg (9.4 lb); 26 (62%) were domestic shorthairs. Seventy-three of the 76 (95%) affected joints had medial patellar luxation. Luxation grades could be assigned to 65 joints, with grade 2 (30 joints) and 3 (22 joints) luxation being most common. Lameness grades could be assigned to 73 joints, with grade 1 lameness (27 joints) most common. Outcome was excellent for 8 of 17 joints treated without surgery and for 23 of 35 joints treated surgically. Complications attributable to surgery were reported in 8 cats. CONCLUSIONS AND CLINICAL RELEVANCE: Patellar luxation should be considered as a cause of hind limb lameness in cats. Low-grade luxation can be associated with lameness of the same severity as high-grade luxation. Surgical correction of patellar luxation in cats with grade 2 or 3 lameness can result in a favorable outcome.  相似文献
