The first objective of this study was to evaluate intrauterine nitric oxide (NO) and endometrial inducible NO synthase (iNOS) in mares susceptible or resistant to persistent breeding‐induced endometritis (PBIE) within 24 h after breeding. Mares susceptible (n = 6) or resistant (n = 6) to PBIE were inseminated over five cycles, and uterine secretions and endometrial biopsies were collected before and 2, 6, 12 and 24 h after insemination. Uterine secretions were analysed for NO and biopsies were analyzed for iNOS expression. A second experiment evaluated the effect of treatment with dexamethasone or mycobacterial cell wall extract (MCWE) on uterine NO production and endometrial iNOS mRNA expression. Six susceptible mares were inseminated over three cycles with (i) killed spermatozoa without treatment (control), (ii) killed spermatozoa with 50 mg of dexamethasone IV or (iii) MCWE IV 24 h prior to insemination with killed spermatozoa. Six resistant mares were inseminated with killed spermatozoa as a control. Six hours after breeding, uterine biopsies and secretions were collected and evaluated for NO and iNOS mRNA. In Experiment 1, resistant mares had an increase in iNOS mRNA expression 2 h post‐breeding compared to baseline (p = 0.045), 12 h (p = 0.014) and 24 h (p = 0.001). Susceptible mares had higher expression 2 h compared to 6 h (p = 0.046). No differences were observed in mRNA or protein expression of iNOS between resistant and susceptible mares. Resistant mares had a relatively steady amount of total intrauterine NO over 24 h, while susceptible mares had an increase over time, with a significantly higher increase in total NO than resistant mares at 6 (p = 0.04) and 12 h (p = 0.032). In Experiment 2, no differences were observed for iNOS mRNA expression. Susceptible mares had increased NO when compared to resistant mares (p = 0.008) and MCWE decreased NO (p = 0.047).  相似文献

Real‐time PCR was used to investigate the role of progesterone (P4) and oestradiol (E2) in regulation of endometrial cytosolic, secretory and calcium‐independent phospholipase A2 (PLA2G4A, PLA2G2A and PLA2G6, respectively) gene expression. Ovariectomized mares underwent 6 days of E2 pre‐treatment followed by 14 days of P4 supplementation. At the start of P4 treatment (Day 1), mares were assigned in a 2 × 2 factorial design to receive either E2 or vehicle starting on Day 11 and endometrial biopsy collection on either Day 14 when P4 concentrations remained high (>4 ng/ml) or Day 16 when P4 concentrations had declined (0.5–2 ng/ml). Additional biopsies were collected from ovariectomized mares on Day 8, which served as control. Blood samples were collected for P4 determination. PLA2G4A expression was higher (p < 0.05) on Day 14 compared with Day 8. In contrast, PLA2G2A did not change significantly (p < 0.12). PLA2G4A and PLA2G2A gene expression increased (p < 0.05), as P4 concentration dropped, on Day 16. In contrast, PLA2G6 gene expression did not show differences between days. Treatment with oestradiol did not increase PLA2 isoforms expression when compared to treatment with the vehicle. PLA2G4A and PLA2G2A were positively correlated with each other and negatively correlated with P4 concentrations. In conclusion, P4 withdrawal upregulated PLA2G4A and PLA2G2A gene expression, and this was not affected by E2. PLA2G4A and PLA2G2A but not PLA2G6 gene expression may be involved in controlling prostaglandin F2 alpha synthesis and luteolysis.  相似文献

There is a growing interest of industry to replace synthetic chemicals by natural products with bioactive properties from plant origin. The aim of this study was to determine the chemical composition of European pennyroyal (Mentha pulegium) essential oil and to characterize the in vitro antioxidant and antimicrobial activities of its water (hot and cold) and ethanolic extracts and of the essential oil. The essential oil revealed menthone, pulegone and neo-menthol as the main constituents, comprising 35.9, 23.2 and 9.2% of the essential oil, respectively. The hot water extract exhibited the highest antioxidant activity and phenol content. In contrast, the extracts were not very effective to inhibit the growth of the seven foodborne spoilage and pathogenic bacteria tested, but the essential oil showed antibacterial activity against all bacterial strains. In conclusion, extracts and essential oil of M. pulegium from Mediterranean origin have huge potential as an alternative to chemical additives for the food industry.  相似文献

A 70 days feeding trial was carried out on juvenile gilthead seabream (Sparus aurata) (mean initial body weight: 362 ± 59 g) to evaluate whether the simultaneous replacement of fishmeal and fish oil by vegetable ingredients affected the muscle's proteolytic potential. Fish were fed either a fishmeal/fish oil diet, a fishmeal/vegetable oil diet, a plant protein/fish oil or a plant protein/vegetable oil diet. The use of dietary plant proteins resulted in lower sulphated glycosaminoglycans' content and lower glycogen phosphorylase activity in the muscle, while both plant proteins and vegetable oil increased muscle pH and reduced cathepsin B activity. Our results indicate that high replacement of fish meal and/or fish oil with plant ingredients has a significant impact on early postmortem metabolic processes and proteolytic potential of seabream muscle, especially when replacing both fish meal and fish oil.  相似文献

The effect of the replacement of corn silage with spineless cactus associated with Tifton 85 hay or sugarcane bagasse was evaluated on the performance, body weight components and sheep meat quality. Twenty-one Santa Inês male sheep with initial body weight of 22.9 ± 2.9 kg were used. They were confined in individual stalls for 74 days receiving the following diets: corn silage + concentrate (CS); spineless cactus + Tifton 85 hay + concentrate (TH); spineless cactus + sugarcane bagasse + concentrate (SB). After the feedlot period, the animals were slaughtered and their body components were weighed and sampled. Data were submitted to Tukey’s test at 5% probability. The average daily gain (0.25 vs 0.14 kg), cold carcass weight (13.3 vs 13.7 kg), loin muscularity (0.6 vs 0.4 kg) and liver weight (0.7 vs. 0.5 kg) were higher (P < 0.05) for the TH diet when compared to that for the CS. The physical-chemical parameters of meat were not influenced (P > 0.05) by the treatments and presented about 20.1% of crude protein, 6.1% of crude fat and shear force of 2.0 kgf/cm2. In complete diets for sheep, Tifton 85 hay associated with spineless cactus provides greater weight gain, cold carcass muscularity and organ weight when compared to those for animals fed with corn silage as exclusive roughage. Sugarcane bagasse associated with spineless cactus may replace corn silage in complete diets for sheep.  相似文献

The objectives of this study were to evaluate the effect of heat treatment of eucalypt wood (Eucalyptus grandis Hill ex Maiden) on the dynamic modulus of elasticity by using the stress wave nondestructive method and also to determine the air-dry density variation, weight loss and equilibrium moisture content following treatment. Heat treatments were performed at four different temperatures (180, 200, 215 and 230°C) and for three different durations (15?min, 2 and 4?h). The results revealed a significant reduction in air-dry density following heat treatment independent of temperature and time. A significant weight loss was observed between and within treatments. The treatment at 230°C for 2 and 4?h produced a weight loss of 20.5 and 26.5%, respectively, which was statistically different from other treatment conditions. The dynamic modulus of elasticity decreased by about 13% in the most severe treatment (230°C for 4?h). Depending on the temperature and time, the equilibrium moisture content was significantly reduced within the range of 40–74%.  相似文献

To meet demand for processes that minimize the environmental impact generated by waste, efficient systems that degrade such substances and use them as an alternative source for renewable energy generation are increasingly becoming needed. Increased food production to meet the needs of the world’s increasing population has encouraged the use of agrochemicals in order to ensure productivity in crops. However, excessive use of pesticides has caused contamination of natural systems and, therefore, of living beings. In this context, this work presents an alternative plan for an integrated system that simultaneously remediates contaminated environments and generates electricity using a Cu/CuO electrode as a photocatalyst. The materials were prepared from reagents and accessible metals, which reduced costs and contributed to a clean process, without using organic additives. The results showed that the generation of current in an area 6.9 cm2 was 193.37 μA for potassium hydrogen phthalate degradation. The Aminol 806® and Connect® pesticides were degraded by 54.46 and 21.02%, respectively, after 90 min in the system, under ultraviolet radiation. The results showed that, at pH 2.0, the generation of current was 2493.2 mA (36.165 mA m?2) for Aminol 806® and 7.894 mA (0.114 mA m?2) for Connect®. The degradation of organic contaminants and simultaneous power generation of energy in the integrated system provides a self-sustaining form of environmental remediation and energy recovery, and its use is possible on a large scale.  相似文献
