Summary Barriers to interspecific hybridization in Trifolium were investigated by manipulation of mentor pollen treatments, ploidy levels, and compatibility and male sterility systems. Crosses involving the addition of mentor pollen produced fewer seeds and hybrids than crosses involving normal pollination. Lower seed set with mentor pollen was deduced to result from the use of less viable pollen, approximately half the pollen having been killed by alcohol. Pollinations at the diploid level resulted in more hybrids than at the tetraploid level, perhaps because genes for male sterility produced higher female sterility in the tetraploids. The self-compatible stock produced more seeds, mostly selfs, than the self-incompatible stock, but produced more hybrids only in one cross, T. pratense L. × T. diffusum Ehrh. The use of male-sterile female parents reduced selfing but produced fewer hybrids than male-fertile female parents. Techniques of this study were designed to affect prefertilization barriers, but the lack of effect may indicate that postfertilization barriers in Trifolium are of greater importance.Journal Article No. 98-3-208 of Kentucky Agricultural Experiment Station. Genetic evaluation aims to identify genotypes with high empirical breeding values (EBVs) for selection as parents. In this study, 2157 potato genotypes were evaluated for tuber yield using 8 years of early‐stage trial data collected from a potato breeding programme. Using linear mixed models, spatial parameters to target greater control of localised spatial heterogeneity within trials were estimated and variance models to account for across‐trial genetic heterogeneity were tested. When spatial components improved model fit, correlations of errors were mostly small and negative for marketable tuber yield (MTY) and total tuber yield (TTY), suggesting the presence of interplot competition in some years. For the analysis of multi‐environment trials, a variance model with a simple correlation structure (with heterogeneous variances) was the most favourable variance structure fitted for TTY and PTY (per cent marketable yield). There was very little difference in model fit when comparing a factor analytic structure of order 2 (FA2) with either FA1 or simple correlation structures for MTY, indicating that simple variance models may be preferable for early‐stage genetic evaluation of potato yield.  相似文献

Summary Seeds of early generations of three reciprocal congruity-backcross (CBC) pedigrees, developed by backcrossing Phaseolus vulgaris-P. acutifolius hybrids to each of the parent species in alternate generations, exhibited a preponderance of traits (size, shape, color, and pattern) of the cytoplasmic parent. The large size of Red Cloud (V1), the P. vulgaris parent common to all of the pedigrees, dominated pedigrees with V1 as the cytoplasmic parent, while the small size and rounded or square shapes of the tepary parents, wild P. acutifolius var. acutifolius PI 263590 or G400445 (A19), wild P. acutifolius var. latifolius PI 406622 (A10), or cultivated P. acutifolius var. latifolius Serowi PI 319443 (A9), were the majority phenotypes when P. acutifolius was the cytoplasmic parent. Continuing through the second cycle of CBC, that is the second backcross with each of the parent species or the fourth backcross, began an amelioration of the apparent cytoplasmic effect on gene expression, as reciprocal pedigrees became more alike, usually with intermediate expression of parental traits or the appearance of new traits. The large seed size of V1 was recovered in hybrids with P. acutifolius cytoplasm and the kidney shape of V1 became rare in hybrids with P. vulgaris cytoplasm. Although the tepary-bean parents represented two subspecies and both cultivated and wild P. acutifolius, the three sets of reciprocal-hybrid pedigrees with P. vulgaris Red Cloud are surprisingly similar. It may be that the exotic parent used to develop a CBC pedigree should be selected more for combining ability in the interspecific cross than for specific economic traits. While the number of generations (six or more) required to produce fertile, intermediate CBC hybrids (that did not require embryo rescue) may preclude routine use of this method by practical plant breeders, the crossability of advanced hybrids with both parental species and the amount of variability apparent in advanced-hybrids progenies suggests that CBC would be valuable for maintaining exotic germplasm in immediately useful forms.  相似文献

The objectives of this study were to determine total radioactive residues and chlorate residues in edible tissues of cattle administered at three levels of sodium [36Cl]chlorate over a 24-h period and slaughtered after a 24-h withdrawal period. Three sets of cattle, each consisting of a heifer and a steer, were intraruminally dosed with a total of 21, 42, or 63 mg of sodium [36Cl]chlorate/kg of body weight. To simulate a 24-h exposure, equal aliquots of the respective doses were administered to each animal at 0, 8, 16, and 24 h. Urine and feces were collected in 12-h increments for the duration of the 48-h study. At 24 h after the last chlorate exposure, cattle were slaughtered and edible tissues were collected. Urine and tissue samples were analyzed for total radioactive residues and for metabolites. Elimination of radioactivity in urine and feces equaled 20, 33, and 48% of the total dose for the low, medium, and high doses, respectively. Chlorate and chloride were the only radioactive chlorine species present in urine; the fraction of chlorate present as a percentage of the total urine radioactivity decreased with time regardless of the dose. Chloride was the major radioactive residue present in edible tissues, comprising over 98% of the tissue radioactivity for all animals. Chlorate concentrations in edible tissues ranged from nondetectable to an average of 0.41 ppm in skeletal muscle of the high-dosed animals. No evidence for the presence of chlorite was observed in any tissue. Results of this study suggest that further development of chlorate as a preharvest food safety tool merits consideration.  相似文献

BACKGROUND: The insulin-like growth factor (IGF) system is implicated in the pathogenesis of diabetes and cardiovascular disease. OBJECTIVE: We report the effects of total energy intake on the IGF system in two populations with markedly different dietary macronutrient intake and cardiovascular event rate. DESIGN, SUBJECTS AND SETTING: Dietary macronutrient intake was measured in a specific Gujarati migrant community in Sandwell, UK (n=205) compared with people still resident in the same villages of origin in India (n=246). Fasting IGF-I, IGF-binding protein (IGFBP)-1 and IGFBP-3, insulin and glucose (0 and 2-hour) were measured. RESULTS: Total energy and total fat intake were higher in UK migrants, as were IGFBP-3 and IGF-I (mean (95% confidence interval): 145.9 (138.1-153.6) vs. 100.9 (94.6-107.3) ng ml(-1); F=76.6, P<0.001). IGFBP-1 was lower in UK migrants (29.5 (25.9-33.0) vs. 56.5 (50.6-62.5) microg l(-1); F=48.4, P<0.001). At both sites, IGF-I correlated positively with total energy (Spearman's rho=0.45, P<0.001) and total fat (rho=0.44, P<0.001) as did IGFBP-3 with total energy (rho=0.21, P<0.05) and fat (rho=0.26, P<0.001). Conversely, in Indian Gujaratis, IGFBP-1 fell with increasing total energy (rho=-0.27, P<0.001) and fat intake (rho=-0.26, P<0.01) but not in UK Gujaratis. Multiple linear regression modelling showed that increasing quartiles of fat intake were associated with higher IGF-I (beta=0.42, P=0.007) independent of age, body mass index, plasma insulin, fatty acids and 2-hour glucose. CONCLUSION: In these genetically similar groups, migration to the UK and adoption of a different diet is associated with marked changes in the IGF system, suggesting that environmental factors profoundly modulate serum concentrations and actions of IGFs.  相似文献

A comparison was made of 15 different techniques which are used in assessing soil microbial populations and/or biomasses. These include direct observations (fungal standing crop, fluorescein diacetate active mycelia, acridine orange stained bacteria), cultural methods (bacterial plate counts), physiological methods (total microbial, bacterial and fungal biomasses, O2-uptake), soil enzyme analyses (dehydrogenase, catalase, alkaline and acid phosphatase, protease, amylase), and ATP-analyses. The various techniques were applied to six soils known to have different microbial characteristics. The results are discussed with respect to the convertability of counts and measurements into microbial biomasses, the variability of the techniques, the correlations within comparable groups of methods, and the practical limitations in application of individual methods to different soils.  相似文献

Buffers have been found to reduce non-point source pollution (NPSP) from watersheds. Hydrologic simulation models assist in predicting the effects of buffers on runoff and sediment losses from small watersheds. The objective of this study was to calibrate, validate and simulate runoff and sediment losses and compare buffer effects on NPSP losses relative to control watersheds (no buffer) for seven years. The experimental design consists of four watersheds under pasture management which were monitored from 2002 through 2008; two with agroforestry buffers (AgB 100 and AgB 300) and two control watersheds (CW 400 and CW 600). Pasture areas included red clover (Trifolium pretense L.) and lespedeza (Kummerowia stipulacea Maxim.) planted into fescue (Festuca arundinacea Schreb.) while the agroforestry buffer area included Eastern cottonwood trees (Populus deltoids Bortr. ex Marsh.) planted into fescue. The APEX model was calibrated from 2002 to 2005 and was validated from 2006 to 2008. The r 2 and NSE values for the calibration and validation periods of the runoff varied from 0.52 to 0.78 and 0.50 to 0.74, respectively. The model did not predict sediment loss very well probably due to insufficient number of measured events and low measured sediment loss. The measured runoff was 57% higher for CW watersheds compared to AgB watersheds. The measured sediment loss was 95% higher for CW watersheds compared to AgB watersheds. After calibrating and validating the model, it was run for long-term scenario analyses for 10 years from 1999 to 2008. Simulated buffer width had a significant influence on runoff. Simulated runoff decreased by 24% when the buffer width was doubled compared to losses associated with the measured buffer width. Simulated runoff from the CW watersheds was 11% higher with double stocking density (relative to measured density) compared to AgB watersheds with double stocking density. With half stocking density (relative to measured density), the AgB watershed had 18% lower runoff compared to CW. Results from this study imply that establishment of agroforestry buffers on grazed pasture watersheds reduce runoff and sediment losses compared to control watersheds without buffers.  相似文献

The critical load concept is now accepted throughout Europe as a means of estimating the sensitivity of key components of aquatic and terrestrial ecosystems to atmospheric inputs of sulphur (S) and nitrogen (N). Current UK freshwater maps, based on steady-state water chemistry, are derived using a critical acid neutralising capacity (ANCLIM) value of zero eql–1, which is based on the probability of occurrence of salmonid fish in lakes. In practice most acidification damage to salmonid fish occurs in nursery streams at the emergence and first feeding stages. In general a clear relationship exists between salmon (Salmo salar L.) and trout (S. trutta L.) densities in Scottish streams and ANC values. However, differences between sites depend on which ANC value is used (eg maximum, minimum or mean). By contrast, when the exceedance of critical loads is compared with salmonid densities the relationship is less clear because many exceeded sites have good salmonid densities. Many of these latter sites are found in north-west Scotland where sea-salt inputs are high and ANC is usually greater than zero eql–1, although diatom-based studies indicated slight acidification of these waters, with a point of change in diatom flora close to ANC=20 eql–1. These false exceedances are probably due to preferential adsorption of acidic SO4 deposition which results in an overestimate of exceedance values. All sites with a mean ANC 0 are fishless but some sites with negative minimum ANC values had normal salmonid densities. Consequently a mean ANCLIM value of zero in the critical load equations for UK freshwaters appears to be too low to protect salmonid stocks. Values between 20–50 eql–1 represent a more realistic range if prevention of long term damage to salmonid stocks is to be achieved.  相似文献

Soil compaction is one of the major problems facing modern agriculture. Overuse of machinery, intensive cropping, short crop rotations, intensive grazing and inappropriate soil management leads to compaction. Soil compaction occurs in a wide range of soils and climates. It is exacerbated by low soil organic matter content and use of tillage or grazing at high soil moisture content. Soil compaction increases soil strength and decreases soil physical fertility through decreasing storage and supply of water and nutrients, which leads to additional fertiliser requirement and increasing production cost. A detrimental sequence then occurs of reduced plant growth leading to lower inputs of fresh organic matter to the soil, reduced nutrient recycling and mineralisation, reduced activities of micro-organisms, and increased wear and tear on cultivation machinery. This paper reviews the work related to soil compaction, concentrating on research that has been published in the last 15 years. We discuss the nature and causes of soil compaction and the possible solutions suggested in the literature. Several approaches have been suggested to address the soil compaction problem, which should be applied according to the soil, environment and farming system.

The following practical techniques have emerged on how to avoid, delay or prevent soil compaction: (a) reducing pressure on soil either by decreasing axle load and/or increasing the contact area of wheels with the soil; (b) working soil and allowing grazing at optimal soil moisture; (c) reducing the number of passes by farm machinery and the intensity and frequency of grazing; (d) confining traffic to certain areas of the field (controlled traffic); (e) increasing soil organic matter through retention of crop and pasture residues; (f) removing soil compaction by deep ripping in the presence of an aggregating agent; (g) crop rotations that include plants with deep, strong taproots; (h) maintenance of an appropriate base saturation ratio and complete nutrition to meet crop requirements to help the soil/crop system to resist harmful external stresses.  相似文献

Nitrogen (N) is a major factor limiting grain production in the high rainfall zone (HRZ, 450–700 mm annual average rainfall of southwestern Australia (SWA). Transient waterlogging and leaching of applied N fertilizer are hazards faced in most years by crop producers. The major crops are wheat (Triticum aestivum L.), barley (Hordeum vulgare L.), canola (Brassica napus L.) and lupin (Lupinus angustifiolius L.) grown in rotation. Two series of experiments involving, levels and timing of N fertilizer application and levels of plant population were done. The first series, in 2003–2004, consisted of 3 experiments in 3 growing seasons (early May to late-October) to measure the grain yield (GY) increase (response) of wheat and barley to various methods of N fertilizer application (methods of split N application were compared to N applied at sowing). The aim of the experiments was to determine the optimal N fertilizer application strategy for maximum GY and quality in situations where transient waterlogging was a frequent occurrence. The second series of four experiments, from 2007–2009, measured the GY of wheat sown at three levels of plant population to 4 levels of N applied after transient waterlogging (taken to be rainfall events in which >25 mm of rain was recorded in 24 to 48 hours).

Applying the N fertilizer after high rainfall and transient waterlogging (tactical N application) increased GY and protein percentage of grain compared to applying all of the N fertilizer at sowing. Where transient waterlogging was not frequent, applying the N after waterlogging was not always better than applying part of the N according to growth stage of the crop or according to fixed times after sowing. When the crop was water-logged three or more times, N uptake by the crop at anthesis and apparent fertilizer N recovery in the crop was substantially increased by applying the N after waterlogging compared to applying the entire N at sowing. This study found that a tactical N management strategy for the HRZ of SWA is to apply some N at sowing with subsequent applications made after heavy rainfall that leads to transient waterlogging. Split N fertilizer applied either according to time after sowing or to growth stage of the crop was equally effective for increasing GY in situations where waterlogging was less frequent.

The observation from these experiments, that grain yield increases due to splitting the N dose were associated with increases in ear numbers, lead to a further set of experiments where plant population was increased in conjunction with N applied after waterlogging events. The combined strategy of increased plant population with strategic N application decreased the amount of N required for maximum GY where more than 3 heavy rainfall events occurred in a growing season.

One practical outcome of this research is to indicate that farmers can withhold applications of N fertilizer after sowing in seasons when transient waterlogging does not occur.  相似文献
