Neonatal foals have unique pharmacokinetics, which may lead to accumulation of certain drugs when adult horse dosage regimens are used. Given its lipophilic nature and requirement for hepatic metabolism, metronidazole may be one of these drugs. The purpose of this study was to determine the pharmacokinetic profiles of metronidazole in twelve healthy foals at 1–2.5 days of age when administered as a single intravenous (IV) and intragastric (IG) dose of 15 mg/kg. Foals in the intravenous group were studied a second time at 10–12 days of age to evaluate the influence of age on pharmacokinetics within the neonatal period. Blood samples were collected at serial time points after metronidazole administration. Metronidazole concentration in plasma was measured using LC‐MS. Pharmacokinetic parameters were determined using noncompartmental analysis and compared between age groups. At 1–2.5 days of age, the mean peak plasma concentration after IV infusion was 18.79 ± 1.46 μg/mL, elimination half‐life was 11.8 ± 1.77 h, clearance was 0.84 ± 0.13 mL/min/kg and the volume of distribution (steady‐state) was 0.87 ± 0.07 L/kg. At 10–12 days of age, the mean peak plasma concentration after IV infusion was 18.17 ± 1.42 μg/mL, elimination half‐life was 9.07 ± 2.84 h, clearance was 1.14 ± 0.21 mL/min/kg and the volume of distribution (steady‐state) was 0.88 ± 0.06 L/kg. Oral approximated bioavailability was 100%. Cmax and Tmax after oral dosing were 14.85 ± 0.54 μg/mL and 1.75 (1–4) h, respectively. The elimination half‐life was longer and clearance was reduced in neonatal foals at 1–2.5 days as compared to 10–12 days of age (P = 0.006, P = 0.001, respectively). This study warrants consideration for altered dosing recommendations in foals, especially a longer interval (12 h).  相似文献

Reasons for performing study: Equine keratomycosis in the western USA has received little study, probably owing to its low prevalence. Objectives: To determine clinical features, predominant fungal isolates, treatment modalities and outcomes of horses with keratomycosis in California and compare these with results from different geographic regions. Methods: Records of horses presented to the University of California‐Davis Veterinary Medical Teaching Hospital (UCD‐VMTH) with confirmed keratomycosis between 1987 and 2010 were reviewed for this retrospective study. Information retrieved from the record included background, ophthalmic examination findings, treatment prior to and following presentation, visual outcome, and ocular survival. Results: A total of 48 eyes in 47 horses met the inclusion criteria and comprised 2% of cases presented to the UCD‐VMTH ophthalmology service. Prior to presentation, 20 horses (43%) received at least one topically administered anti‐inflammatory medication. Keratomycosis was confirmed by fungal culture in 38 horses (81%), by histopathology in 2 horses (4%) and by cytology in 7 horses (15%). Forty‐four isolates were identified in the 38 horses cultured; Aspergillus was the most common isolate (64%) and a novel isolate, Papulospora, was identified in 2 horses. Treatment consisted of medication only (73%), medical and surgical treatment (25%), or immediate enucleation (2%). Globe retention was 77% and vision retention was 53%. Corneal perforation was significantly associated with loss of vision (P<0.001). Conclusions: Keratomycosis is relatively uncommon in horses presented for ophthalmic conditions at UCD‐VMTH. Corneal perforation was a negative prognostic indicator for vision in this population of northern Californian horses.  相似文献

Stachys arvensis (staggerweed) is a common, widely distributed weed of cultivated and waste land with the potential to intoxicate sheep. Two naturally occurring outbreaks of suspected staggerweed toxicity in the lower North Island were investigated. Affected lambs had been recently moved onto staggerweed-contaminated Brassica spp. crops. In total, 150/1,200 (13%) lambs developed hindlimb paresis, a fine generalised muscular tremor, and hunched posture. When forced to move, many became recumbent. Most lambs recovered within 48 h of removal from staggerweed, although a few developed clinical signs again when transported 2–3 weeks later. Grossly, affected lambs had large amounts of staggerweed plant material and seeds within the rumen. Histopathology showed mild, multifocal degeneration of the white matter tracts of the central nervous system (CNS), most commonly in the ventral funiculi of the spinal cord, and acute, mild to moderate, multifocal degeneration of skeletal muscles. Creatine kinase (CK) activity in serum was mildly to markedly elevated in affected lambs. In a feeding trial, ten 10-month-old Romney lambs were randomly assigned to equal treatment and control groups. Treated lambs were drenched with a liquid extract of staggerweed once daily for 7 days. Three of five treated lambs developed mild exercise intolerance, and 1/5 displayed mild paresis of the hindlimbs, slightly crouched hindlimb stance, and shortened gait, on days 6 and 7. Histologically, 4/5 treated lambs had degeneration in white matter tracts of the CNS, indistinguishable from those seen in the lambs in the outbreak, and in 1/5 lambs there was scattered regeneration of skeletal muscle. CK activity in serum in treated lambs was not significantly higher than that in control lambs. None of the control lambs developed significant clinical signs, histological changes or increases in CK activity in serum. The clinical signs and lesions observed in both the outbreaks and feeding trial were similar to those previously described in studies in Australia, with the exception that myodegeneration was more prominent in the outbreaks in New Zealand. Further characterisation of the pathogenesis of staggerweed toxicity and its potential role as a food safety hazard will be facilitated through identification of the toxic principle(s).  相似文献

A 5‐week‐old foal was evaluated for fever and hematuria of 3 days duration. Cystoscopy localized the blood to be originating from the left ureter. Abdominal ultrasonography revealed left hydronephrosis, hydroureter, and omphaloarteritis of the left umbilical artery with abscess formation that communicated with an arterial structure. Computed tomography (CT) revealed a large aortic aneurysm within the center of the abscess. An exploratory celiotomy was performed and the infection was nonresectable. The prognosis for life was grave; therefore the colt was euthanized. Necropsy findings confirmed the antemortem diagnosis. Ultrasound and CT imaging in this case provided an accurate antemortem diagnosis.  相似文献

Measurements of physiologic parameters, such as blood pressure or lactate concentration, are useful to detect occult derangements, such as tissue hypoxia and dysoxia. These tools are also useful in determining the effects of therapy. Monitoring techniques are now widely available for the intensive management of critically ill horses and foals. A number of these have evolved into noninvasive or minimally invasive devices and procedures and provide information that can be used for earlier and more dynamic therapeutic intervention. The goal of increased monitoring is to improve the level of care in the ICU; L ultimately. increased survival of critical patients is the motivation behind enhanced monitoring of physiology, with particular attention being paid to trends or alterations over time. This review highlights practical and informative monitoring tools and techniques and provides normal reference values from the literature.  相似文献

Objective: To assess the toxicity of residues of ivermectin and moxidectin in cattle faeces collected at intervals after treatment.
Design: Replicated bioassays of faeces using larvae of the bush fly, Musca vetustissima and the house fly, Musca domestica .
Animals: Two groups of five Murray Grey x Aberdeen Angus steers were treated with injectable formulations of ivermectin and moxidectin respectively. A third group was used as an untreated control.
Procedure: Newly emerged fly larvae were reared in the dung of treated animals.
Results: Drug residues in faeces collected 3 to 35 days after treatment with an injectable formulation of moxidectin had no significant effect on the survival of larvae of M vetustissima . Similarly, faeces dropped up to seven days after treatment caused no significant reduction in larval survival in M domestica . In day 2 dung, residues of moxidectin delayed development of M vetustissima larvae, but had no effect on their survival. In contrast, ivermectin-treated steers, produced dung that inhibited larval development of both M vetustissima and M domestica for 7 to 14 days after treatment. Significant reductions in survival of M vetustissima larvae occurred in dung collected on days 21 and 28 after treatment, but by day 35 survival did not differ from that in control dung.
Conclusion: Excreted faecal residues of moxidectin are relatively innocuous to larvae of both M vetustissima and M domestica . Those of ivermectin inhibit survival for 7 to 14 days after treatment and are likely to have adverse effects on non-target organisms.  相似文献

A neurological disorder in Merino sheep, characterised clinically by progressive posterior ataxia and microscopically by Wallerian degeneration in thoracic segments of the spinal cord, is described. Animals of both sexes were affected, with the earliest onset of disease being at 5 months of age. Most affected animals died before 2 years of age. The clinical, pathological and epidemiological features suggest that this degenerative thoracic myelopathy is a previously unrecognised entity differing from other reported causes of ataxia in sheep in Australia.  相似文献
