Phytocystatins are the plant thiol protease inhibitors involved in several reaction mechanisms of the plant system like regulation of proteolytic activity and storage of proteins. Biochemical and biophysical changes induced by fungicide SDD in phytocystatin purified from Phaseolus mungo have been investigated in terms of mass spectroscopy, Fourier transform infrared spectroscopy, and fluorescence spectroscopy, at pH 7.0, with varying fungicide concentrations (1-9 mM) and a time of incubation ranging from 2 to 8 h at 37 degrees C, with a fixed cystatin concentration (1.5 mM). Reactive oxygen species responsible for inhibitor damage were also investigated, and thiourea was found to scavenge the free radicals generated by SDD. FTIR analysis indicates a significant conformational transition from alpha-helix to beta-sheet structure; quenching of fluorescence is evident by fluorescence spectroscopy. The activity assay showed a decrease in inhibitory activity, as well as a fragmentation of the inhibitor was observed in electrophoresis. Results obtained implicate that exposure of phytocystatins to SDD involves physicochemical changes in cystatins leading to damage and a decrease in the activity of the inhibitor.  相似文献

Methodologies for the determination of vitamin B(2) in food matrixes and a premix using simple sample conditioning steps coupled with a convenient and cheap electrochemical sensing device are presented. Electrochemical analysis based on differential pulse voltammetry (DPV) coupled to carbon electrodes gave a well-defined reduction peak at -0.42 V versus a Ag/AgCl quasi-reference electrode. Using a straightforward sample preparation step, vitamin B(2) can be measured successfully in a nutritional premix and food products. Standard additions of riboflavin were used to confirm the analyte concentrations and to provide precision data.  相似文献

Leptosphaeria maculans causes blackleg disease on Brassica napus, an economically important oilseed crop. Brassica juncea has high resistance to blackleg and is a source for the development of resistant B. napus varieties. To transfer the Rlm6 resistance gene from B. juncea into B. napus, an interspecific cross between B. napus “Topas DH16516” and B. juncea “Forge” was produced, followed by the development of F2 and F3 generations. Sequence characterized amplified region (SCAR) and cleaved amplified polymorphic sequence (CAPS) markers linked to the L. maculans resistance gene Rlm6 were developed. Segregation of SCAR and CAPS markers linked to Rlm6 were confirmed by genotyping of F2 and F3 progeny. Segregation of CAPS markers and phenotypes for blackleg disease severity in F2 plants had a Mendelian ratio of 3:1 in resistant vs. susceptible plants, respectively, supporting the assumption that genetic control of resistance was by a single dominant gene. The molecular markers developed in this study, which show linkage with the L. maculans resistance gene Rlm6, would facilitate marker‐assisted backcross breeding in a variety development programme.  相似文献

Gladiolus corms were grown in media contaminated with cadmium (Cd) (50 mg kg?1) and supplemented with silicon (Si) and potassium (K). The role of Si and K for mitigation of Cd toxicity was evaluated. Cd-induced stress generated significantly increased level of oxidative stress markers including hydrogen peroxide (H2O2), and malondialdehyde (MDA) in gladiolus. The application of K and Si improved the production of protein and proline in the treated plants. Moreover, K and Si supplemented plants exhibited an improvement in the activity of antioxidant enzymes and a reduction in the level of MDA, H2O2 and Cd uptake under Cd stress. Application of K and Si also enhanced the uptake of mineral nutrients including calcium (Ca), magnesium (Mg), manganese (Mn), sulfur (S) and K. The plants supplemented with K and Si exhibited a higher amount of total phenolics and flavonoids. The combined effect of Si and K was more pronounced regarding beneficial effects on gladiolus plants compared to individual effect of these elements under Cd stress. The current research reveals that Si and K may improve gladiolus growth by decreasing the oxidative stress and Cd uptake and by increasing the activity of antioxidant defense enzymes, the quantity of secondary metabolites and plant nutrition.  相似文献

Boron (B) plays a vital role in cell division and elongation in apical meristem. Drought stress (DS) severely reduced the B-uptake and thus growth and crop-productivity of plants. The aims of this study were to evaluate whether foliar application of B corrects physiological-disorders under DS in two contrasting maize genotypes i.e. Dekalb-6525 (drought-tolerant) and Yousafwala-hybrid (drought-sensitive). Initially, foliar rate of B (0, 2, 4 and 6 mg L?1) was optimized in terms of improved plant growth under drought-stress. Then, optimized rate of B (4 mg L?1) was applied to assess the physiological and biochemical basis of B-induced improved growth of maize under deficit-moisture supply. Drought-stress reduced the growth of plants by lowering in water-status (leaf water-relations), photosynthetic capacity (gas-exchange aspects, photosynthetic-pigments), membrane permeability, imbalance in redox potential (oxidative-stress, antioxidant-defense system) and tissue-B concentration. Application of B considerably improved maize growth by improving water-status, photosynthetic capacity, tissue-B concentration as well as up-regulation of antioxidative defense-system. Moreover, ameliorative effects of B on maize was also evident from stress relieving indicators such as slight decrease in accumulation of proline, total free amino-acids, total soluble-sugars and MDA content under water-deficit conditions. In addition, cultivar Dekalb-6525 showed considerable improving response to B application over Yousafwala-hybrid.  相似文献

Camelina sativa has rejuvenated as a successful oilseed crop in the recent years. It is a low-input-requiring crop with an unusual fatty acid composition. A pot experiment was conducted in rain-out shelter to investigate the effect of different nitrogen levels (N0 = 0, N1 = 50, N2 = 100, and N3 = 150 kg ha?1) on the growth and yield of two C. sativa genotypes (Australian and Canadian) under normal [100% field capacity (FC)] and water stressed (60% FC) conditions. The experiment was laid out in a completely randomized design with factorial arrangement having three replicates in the Department of Crop Physiology, University of Agriculture, Faisalabad. The results indicated that nitrogen doses and water stress levels significantly affected the growth and yield of C. sativa. Maximum values for growth indices like leaf area index (LAI), crop growth rate (CGR), leaf area duration (LAD), net assimilation rate (NAR), and yield attributes were observed under N2 treatment (100 kg ha?1) followed by the treatment in which nitrogen was applied at the rate of 50 kg ha?1. However, growth and yield components significantly reduced under water stress conditions (60% FC). Of both the genotypes, Australian Camelina performed better as compared to Canadian Camelina under both non-stress and drought stress conditions.  相似文献

Disease associated with Maedi-Visna infection results in substantial economic losses in affected sheep producing areas of the world. A survey was conducted to estimate herd and individual seroprevalence in the province of Manitoba and evaluate risk factors for seropositive herds. Of 2207 sheep sampled from 77 selected sheep flocks, the animal level seroprevalence was 2.47% and herd level seroprevalence was 25.10%. The herd-level factors of presence of clinical skin disease, herd size of > 70, history of musculoskeletal/lameness abnormalities, and the purchase of new stock (> 50) in the last 1 to 5 y, showed significant associations with seropositive herd status. The study documented a remarkable stability of low seroprevalence in the province over a 20-year period in the absence of a systematic disease control program.  相似文献
