An indirect enzyme-linked immunosorbent assay (I-ELISA) is described for simultaneous screening of bovine sera for detection of antibodies to bluetongue (BT) and epizootic hemorrhagic disease of deer (EHD) viruses (V). Optimal dilutions of BTV and EHDV antigens were combined and allowed to absorb on to the wells of microtiter plates. Appropriately diluted (1:100) bovine sera were allowed to incubate and the bound antibodies were detected by a murine monoclonal antibody (MAb) to bovine immunoglobulin (H-Chain) conjugated with horseradish peroxidase. The performance of the combined (C) I-ELISA in detecting antibodies to BTV and EHDV in sequential serum samples from calves experimentally inoculated with BTV, serotype 10, EHDV, serotype 1 (New Jersey) or EHDV serotype 2 (Alberta) was evaluated. Comparable antibody profiles were demonstrable by the CI-ELISA and separate I-ELISAs using either BTV or EHDV antigens. The results suggest that the CI-ELISA offers many advantages over the standard agar gel immunodiffusion (AGID) test and has potential application as a rapid, sensitive, inter-group-specific and inexpensive test for simultaneous screening of bovine sera for antibodies to BTV and/or EHDV. 相似文献

Oxidative stress has detrimental effects on semen quality during spermatogenesis and semen processing for artificial insemination. This work was conducted to study the effect of different levels of vitamin E on the semen traits, oxidative status and trace minerals in Beetal bucks. Thirty‐six bucks of similar body weight and age (1 year) were randomly divided into four groups. One group was kept as control with no supplementation (group 1), and the others were supplemented with 200 (group 2), 400 (group 3) and 800 IU (group 4) vitamin E/animal/day for 2 months. At the end of the experiment, semen samples were collected and evaluated. Seminal plasma was separated to study the concentration of superoxide dismutase (SOD), glutathione peroxidase (GPx), aspartate aminotransferase (AST), alanine aminotransferase (ALT) and trace minerals (Zn, Cu, Mn and Fe). Group 3 showed significantly higher (p < 0.05) semen volume and per cent motility and lower dead sperm percentage compared to control group. Superoxide dismutase, GPx, Zn, Cu and Mn were higher in the same group. The level of AST decreased in group 3 without any change on the concentration of ALT. It is suggested that vitamin E at the rate of 400 IU/buck/day supported higher semen volume, per cent motility, per cent live spermatozoa, antioxidants (SOD, GPx) and trace mineral levels (Zn, Cu, Mn) in the seminal plasma. The increased supplementation from 0 to 400 showed a general increasing trend in improving semen quality. We analyzed the mortality impacts and greenhouse gas (GHG) emissions produced by household energy use in Africa. Under a business-as-usual (BAU) scenario, household indoor air pollution will cause an estimated 9.8 million premature deaths by the year 2030. Gradual and rapid transitions to charcoal would delay 1.0 million and 2.8 million deaths, respectively; similar transitions to petroleum fuels would delay 1.3 million and 3.7 million deaths. Cumulative BAU GHG emissions will be 6.7 billion tons of carbon by 2050, which is 5.6% of Africa's total emissions. Large shifts to the use of fossil fuels would reduce GHG emissions by 1 to 10%. Charcoal-intensive future scenarios using current practices increase emissions by 140 to 190%; the increase can be reduced to 5 to 36% using currently available technologies for sustainable production or potentially reduced even more with investment in technological innovation. 相似文献

□ Effects of different arsenic (As) concentration (0–30 mg L?1) on seed germination, root tolerance index, relative shoot height, root and shoot biomass, photosynthetic pigments and arsenic accumulation in two wheat varieties were investigated. Low concentrations of arsenic (0–2.5 mg L?1) stimulated germination percentage, shoot and root elongation, plant biomass as well as chlorophyll content as compared with control, however, these factors all decreased gradually at high concentrations of arsenic (5–30 mg L?1). ‘Zarin’ variety had a significantly higher tolerance to arsenic than ‘Sardari.’ Arsenic accumulation by plants root and shoot increased with the increasing arsenic concentrations in medium, which ‘Zarin’ had a higher ability to absorb and translocate arsenic to the shoots. Root accumulated more arsenic than shoot. The similar trend of chlorophyll content and wheat growth under different arsenic concentration suggesting that arsenic toxicity affects the photosynthesis which ultimately results in the reduction of wheat growth and yield. 相似文献

An experiment was conducted to study the effect of organic manure and chemical fertilizers on soil properties and vegetable crops in the cabbage-brinjal-red amaranth cropping pattern at the homestead in a Grey Terrace Soil (Aric Albaquept) of Bangladesh. There were eight treatments: poultry manure (PM) at 5 t ha?1, cowdung (CD) at 10 t ha?1, household waste (HW) at 10 t ha?1, PM at 2.5 t ha?1 + chemical fertilizers (CF), CD at 5 t ha?1 + CF, HW at 5 t ha?1 + CF, CF, and Control. The lone CF treatment indicates 100% chemical fertilizers and any manure + CF indicates supplementary or reduced rate of fertilizers. The PM at 2.5 t ha?1 + CF treatment performed the best yield for cabbage and brinjal, and HW at 5 t ha?1 + CF yielded the highest for red amaranth. The highest uptake of nitrogen, phosphorus, potassium, and sulfur was also found in the treatment that produced the highest yield. Bulk density, organic carbon, and nutrient availability in soil as determined after two-crop cycles were improved due to the applications of manure. 相似文献

With regard to the low cation-exchange capacity and large saturated hydraulic conductivity of sandy soils, a field experiment was carried out in 2006–2007 to determine the impact of zeolite on nitrogen leaching and canola production. Four nitrogen (N) rates (0, 90, 180, and 270 kg ha–1) and three zeolite amounts (3, 6 and 9 t ha?1) were included as treatments. The results demonstrated that the highest growth parameters and seed yield were attained with 270 kg N ha?1 and 9 t zeolite ha?1. However, the highest and the lowest seed protein percentage and oil content were obtained with 270 kg N ha?1 accompanied by 9 t zeolite ha?1, respectively. Nitrate concentration in drained water was affected by nitrogen and zeolite. The lowest and highest leached nitrate values were found in control without N and zeolite (N0Z0) and in treatments with the highest N supply without zeolite (N270Z0), respectively. In general, nitrogen-use efficiency decreased with an increase in N supply. Application of 9 t zeolite ha?1 showed higher nitrogen use efficiency than other zeolite amounts. Also, application of more N fertilizer in soil reduced nitrogen uptake efficiency. In total, application of 270 kg N ha?1 and 9 t zeolite ha?1 could be suggested as superior treatment. 相似文献
