Diagnosis of corpus luteum (CL) function by rectal palpation (RP) has been widely used for recipient selection of embryo transfer (ET), a technology essential for genetic improvements in cattle. To examine the accuracy of RP diagnosis method, the relationship between RP‐based CL function and reproductive performance was compared in this study. In Experiment 1, CL of Holstein heifers on day 7 after estrus was classified into functional or hypoplastic by RP, and the results were compared with ultrasonographic (US) images and plasma progesterone (P4) levels. As a result, heifers with functional CL judged by RP had a mean maximum CL diameter of 20.1 ± 3.1 mm on US and a mean P4 concentration of 8.1 ± 2.3 ng/mL. These values were significantly greater than those of heifers with hypoplastic CL (12.4 ± 5.4 mm, 4.0 ± 2.8 ng/mL) (P < 0.001). In Experiment 2, the length of the estrus cycle was examined between functional CL and hypoplastic CL. The rate of heifers with a normal estrus cycle length with 18–25 days was significantly lower with hypoplastic CL than with functional CL (16/24 vs. 43/46, P < 0.01). In Experiment 3, 543 inseminated heifers were similarly classified by CL function by RP 7 days after estrus. The heifers with functional CL showed higher pregnancy rate compared with the heifers with hypoplastic CL (75.2 vs. 47.9%, P < 0.0001). Finally, the CL function of 66 heifers was examined by RP on day 7 post‐estrus, and ET was performed in 49 (74.2%) heifers with functional CL. As a result, 27 (55.1%) of them became pregnant. Taken together, these results reconfirm that RP on day 7 after estrus is useful for selection of heifers with functional CL.  相似文献

To assess the effect of changes in organic litter stock on seasonal changes in heterotrophic respiration (R H), soil respiration (R S), and total ecosystem respiration (R E), we measured seasonal changes in leaf litter respiration (R LL) by the chamber method and estimated the seasonal change in total R H using the RothC model in a warm-temperate mixed deciduous?Cevergreen forest in Japan. Both R E and R S had seasonal hysteresis and were higher in spring than at the same temperature during autumn. Under warm and humid conditions, the rate of decomposition of newly supplied leaf litter in one?year was high (60% loss). Consequently, R LL and R H were higher in spring after leaf drop, when more fresh material was available, than in autumn. In this study, 42 and 88% of the difference in R E and R S between spring and autumn (soil temperature 16?C18°C) could be accounted for by the difference in R H, respectively, and 71% of the difference in R H could be accounted for by the difference in R LL. This study showed that seasonal changes in heterotrophic respiration (R LL and R H) could be a major factor in the seasonal hysteresis of R E and R S.  相似文献

The Ppd-A1 genotype of 240 Japanese wheat cultivars and 40 foreign cultivars was determined using a PCR-based method. Among Japanese cultivars, only 12 cultivars, all of which were Hokkaido winter wheat, carried the Ppd-A1a allele, while this allele was not found in Hokkaido spring wheat cultivars or Tohoku-Kyushu cultivars. Cultivars with a photoperiod-insensitive allele headed 6.9–9.8 days earlier in Kanto and 2.5 days earlier in Hokkaido than photoperiod-sensitive cultivars. The lower effect of photoperiod-insensitive alleles observed in Hokkaido could be due to the longer day-length at the spike formation stage compared with that in Kanto. Pedigree analysis showed that ‘Purple Straw’ and ‘Tohoku 118’ were donors of Ppd-A1a and Ppd-D1a in Hokkaido wheat cultivars, respectively. Wheat cultivars recently developed in Hokkaido carry photoperiod-insensitive alleles at a high frequency. For efficient utilization of Ppd-1 alleles in the Hokkaido wheat-breeding program, the effect of Ppd-1 on growth pattern and grain yield should be investigated. Ppd-A1a may be useful as a unique gene source for fine tuning the heading time in the Tohoku-Kyushu region since the effect of Ppd-A1a on photoperiod insensitivity appears to differ from the effect of Ppd-B1a and Ppd-D1a.  相似文献

(1)H NMR spectrometry in combination with multivariate analysis was considered to provide greater information on quality assessment over an ordinary sensory testing method due to its high reliability and high accuracy. The sensory quality evaluation of watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) was carried out by means of (1)H NMR-based metabolomics. Multivariate analyses by partial least-squares projections to latent structures-discrimination analysis (PLS-DA) and PLS-regression offered extensive information for quality differentiation and quality evaluation, respectively. The impact of watermelon and rootstock cultivars on the sensory qualities of watermelon was determined on the basis of (1)H NMR metabolic fingerprinting and profiling. The significant metabolites contributing to the discrimination were also identified. A multivariate calibration model was successfully constructed by PLS-regression with extremely high reliability and accuracy. Thus, (1)H NMR-based metabolomics with multivariate analysis was considered to be one of the most suitable complementary techniques that could be applied to assess and predict the sensory quality of watermelons and other horticultural plants.  相似文献

Asian rice (Oryza sativa L.) is widely cultivated in Asia, where it has been classified into Indica and Japonica Group, the latter is further classified into Tropical and Temperate Japonica Subgroup. O. rufipogon is believed to be the closest ancestor to O. sativa, but it remains unclear whether the two groups arose from a single ancestor or different ancestors. Therefore, here, we investigated the matrilineal ancestors of O. sativa using markers for organelle (chloroplast and mitochondrial) genomes, and 119 O. sativa landraces, 10 O. glaberrima Steud., 115 O. rufipogon Griff. from Asia, and 39 accessions from other wild rice species with AA genomes. We screened 18 organelle markers developed based on polymorphic loci in the organelle genomes. In addition, we used the open reading frame 100 of a chloroplast marker. The results indicated that O. rufipogon first differentiated into two lineages and then further differentiated into Indica and Japonica Group, respectively. Accessions of O. rufipogon (R-1f and R-2d types) from Myanmar appear to be the closest ancestors of Tropical Japonica Subgroup and Indica Group, respectively. Therefore, these wild strains may have made a strong contribution to the domestication of rice landraces in Myanmar.  相似文献

The conservation of rare species on islands is a special challenge, especially when the islands are faced with increasing human impact. Two endangered wild felids in Japan provide a contrast that is useful for examining conservation strategies. The Iriomote cat (Prionailurus bengalensis iriomotensis) is endemic to Iriomotejima Island in Okinawa, and the Tsushima leopard cat (Prionailurus bengalensis euptilurus) occurs in the Tsushima islands in Japan, although a related subspecies found in the Korean Peninsula. Population size was approximately 100 individuals for each subspecies. While the population of Iriomote cat appears relatively stable, the population and abundance of the Tsushima leopard cat has decreased in recent decades. Because of their small population sizes and restricted habitats both are listed as endangered species/subspecies in Japan’s Red List, and the Iriomote cat is listed as an endangered subspecies in the IUCN Red List. Although both are similar-sized felids living on small islands, their ecological characteristics such as food habit, habitat selection, and density differ. These differences seem to be caused by the climate, the biological environment (the species composition of fauna in each island, and the presence of competitors), and the artificial background. The threats facing these two felids are also similar. Habitat destruction, traffic accidents, and negative influences by introduced species are the principal threats to be addressed for their conservation. At the same time, there are differences in the degree of impact by each threat, in each species’ conservation programs in practice, and in the stages of progress and operation of programs. These differences are partly because they have different ecological features and partly because the social background of each island, such as human population, main industry, and historical relationship between human and wildlife, are quite different. We will compare the ecology and state of these two wild felids and discuss the different conservational situations in relation to felid ecology and human society.  相似文献

The association of the polymorphism of bovine leukocyte antigen ( BoLA-DRB3 ) genes identified by the polymerase chain reaction sequence-based typing (PCR-SBT) method with resistance and susceptibility to mastitis caused by pathogenic bacteria was investigated. Blood samples for DNA extraction were collected from 194 Holstein cows (41 healthy cows and 153 mastitis cows including 24 mixed-infection cows infected with 2 or 3 species of pathogens) from 5 districts of Chiba prefecture, Japan. Sixteen BoLA-DRB3 alleles were detected. The 4 main alleles of DRB3*0101 , *1501 , *1201 , and *1101 constituted 56.8% of the total number of alleles detected. Mastitis cows were divided into 2 groups: group 1 with single-infection cows and group 2 with all mastitis cows including 24 mixed-infection cows. The differences in the frequencies of BoLA-DRB3 alleles and the number of cows homozygous or heterozygous for each BoLA-DRB3 allele between healthy cows and the 2 groups of mastitis cows were evaluated. Furthermore, similar comparisons were performed between healthy cows and the 2 groups of mastitis cows for each mastitis pathogen. It was considered that the 4 alleles, namely, DRB3*0101 , *1501 , *1201 , and *1101 had specific resistance and susceptibility to 4 different mastitis pathogens. Thus, DRB3*0101 might be associated with susceptibility to coagulase-negative Staphylococci and Escherichia coli , and DRB3*1501 might be associated with susceptibility to Escherichia coli . However, DRB3*1101 might be associated with resistance to Streptococci and coagulase-negative Staphylococci , and DRB3*1201 , with resistance to Streptococci , Escherichia coli , and Staphylococcus aureus .  相似文献

We determined patterns of microsite suppression in dwarf bamboo Sasa nipponica when grazing deer were absent. This bamboo species is able to outcompete Hondo spruce (Picea jezoensis var. hondoensis) saplings under many environmental circumstances. We set up two 10 × 100 m plots inside a deer-proof fence within a subalpine forest on Mt. Ohdaigahara, central Japan, and two similarly sized plots outside the fence. Within the plots, we surveyed microsites where spruce saplings grew. We measured height and shoot elongation of all spruce saplings, and culm height and cover ratios of dwarf bamboo growing around each spruce sapling. Spruce sapling density and average height were higher inside the deer-proof fence than outside, as were bamboo height and cover. Thus, there was a negative effect of deer browsing on vegetation parameters outside the fence and a suppression of the negative effect of bamboo on spruce sapling growth inside the fence. Spruce sapling height was higher in tree-fall pits than in other microsites inside the fence, whereas both dwarf bamboo height and cover were lower in pits and rocky sites than elsewhere. In soil and collar microsites, spruce sapling shoot growth was lower and bamboo height and cover were higher than in pits and rocky sites. Inside the fence, dwarf bamboo cover was high, but pits and rocks suppressed its growth, allowing spruce saplings to flourish. To restore heavily damaged spruce forests with advanced saplings, it will be necessary to construct deer-proof fences and create and maintain microsites with pits and rocks.  相似文献

This study has focused on solute diffusing into cell walls in solution-impregnated wood under conditioning, process of evaporating solvent. The purpose of this paper was to clarify the RH- (relative humidity-) schedule that promotes the solute diffusion into shrinking cell walls during conditioning. The wood samples impregnated with a 20 mass% aqueous solution of polyethylene glycol (PEG1540) was conditioned with a temperature of 40?°C to the equilibrium point at the RH where the samples swelled maximally. The samples were subsequently conditioned at 40?°C under the schedules including four ways of RH-decrease steps where the cell walls shrunk. The amount of solute (PEGs) diffused into cell walls during the conditioning logarithmically increased with increasing the number of the RH-decrease steps. This was well explained by the theoretical model that describes the solute diffusion into shrinking cell walls. It is clarified from the model that the RH, or moisture content of the sample, should be decreased as gradually as possible to increase the total amount of diffused solute into shrinking cell walls, and that the amount of diffused solute is smaller for the lower moisture content. The model also suggests that effect of change in RH schedule on change in total amount of diffused solute does not depend on solute diffusivity in the sample under drying in a vacuum over phosphorous pentoxide, and that impregnated wood should be conditioned under natural convection rather than forced convection for promoting the diffusion into shrinking cell walls.  相似文献
