ABSTRACT:   In molluscs, mantle epithelial cells secrete organic matrix proteins to form shells. In this study, we established a culture of mantle epithelial cells by using the mantle pallial layer of scallops. We aimed to identify the mantle epithelial cells expressing scallop shell matrix proteins and establish a culture system of epithelial cells. After the mantle pallial layer was carefully isolated from the mantle tissue, explant culture was performed at 4°C. Most cells that migrated from the explant tissue were round cells. Most of the adhered cells retained round morphology, while some of the cells adhered to the dish and showed morphology similar to that of epithelial-like and fibroblast-like cells. When the cultured cells were immunostained with a polyclonal antibody against the shell matrix protein, the antibody recognized many of the adhered cells. An estimation of the number of epithelial cells revealed that approximately 70% of the adhered cells were epithelial cells. This is the first report to describe epithelial cells in cultured mantle cells, which express shell matrix proteins. This culture system may be a useful method for characterization of the mantle epithelial cells.  相似文献

Crucifer species, which include widely consumed vegetables, contain glucosinolates as secondary metabolites. Cruciferous vegetables are consumed in Japan in salt-preserved or pickled form as well as cooked and raw fresh vegetables. In this study, changes in contents of glucosinolates during the pickling process were investigated. 4-Methylthio-3-butenyl glucosinolate, a major glucosinolate in the root of Japanese radish, daikon (Raphanus sativus L.), was detected in pickled products with a short maturation period but not in those with a long maturation period. As a model pickling experiment, fresh watercress (Nasturtium officinale) and blanched watercress were soaked in 3% NaCl solution for 7 days. The results showed that the ratio of indole glucosinolates to total glucosinolates increased during the pickling process, whereas total glucosinolates decreased. Myrosinase digestion of glucosinolates in nozawana (Brassica rapa L.) indicated that indole glucosinolates, especially 4-methoxyglucobrassicin, were relatively resistant to the enzyme. The effect of pickling on glucosinolate content and the possible mechanism are discussed in view of degradation by myrosinase and synthetic reaction in response to salt stress or compression during the pickling process.  相似文献

A total of 518 fecal samples collected from 183 apparently healthy cattle, 180 pigs and 155 broilers throughout Japan in 1999 were examined to determine the prevalence and antimicrobial susceptibility of Salmonella. The isolation rates were 36.1% in broilers, 2.8% in pigs and 0.5% in cattle. S. enterica Infantis was the most frequent isolate, found in 22.6% of broiler fecal samples. Higher resistance rates were observed against oxytetracycline (82.0%), dihydrostreptomycin (77.9%), kanamycin (41.0%) and trimethoprim (35.2%). Resistance rates to ampicillin, ceftiofur, bicozamycin, chloramphenicol and nalidixic acid were <10%. CTX-M-2 β-lactamase producing S. enterica Senftenberg was found in the isolates obtained from one broiler fecal sample. This is the first report of cephalosporin-resistant Salmonella directly isolated from food animal in Japan.  相似文献

BACKGROUND: Trifluralin‐resistant biotypes of water foxtail (Alopecurus aequalis) have been identified in wheat fields from northern Kyushu, Japan. Water foxtail is a winter‐annual grassy weed, causing substantial crop losses. This study reports on mutation in α‐tubulin (TUA) genes from water foxtail, the site of action of trifluralin. RESULTS: Two trifluralin‐sensitive (S) Chikugo and Ukiha biotypes and four trifluralin‐resistant (R) Asakura‐1, Asakura‐2, Tamana and Tosu biotypes of water foxtail were used for herbicide resistance analysis. R biotypes showed 5.7–30.7‐fold trifluralin resistance compared with the S biotypes. No differences in the uptake and translocation of 14C‐trifluralin were observed between Chikugo (S) biotype and Asakura‐1 (R) biotype. Most of the 14C detected in the plant material was in the root tissue, and no substantial increases were noted in shoot tissues. Comparative TUA sequence analysis revealed two independent single amino acid changes: change of Val into Phe at position 202 in TUA1 and change of Leu into Met at position 125 in TUA3 in Asakura‐1 biotype. In the Tamana (R) biotype, two amino acid changes of Leu to Phe at position 136 and Val to Phe at position 202 were observed in the predicted amino acid sequence of TUA1, compared with Chikugo (S) biotype. CONCLUSION: The results provide preliminary molecular explanation for the resistance of water foxtail to trifluralin, a phenomenon that has arisen as a result of repeated exposure to this class of herbicide. Gingival mass lesions developed when cyclosporine was administered for 600 days to a female, 7-year-old, longhaired dachshund diagnosed with intractable immune-mediated hemolytic anemia (IMHA). Histopathology indicated hyperplastic suppurative gingivitis. As the anemia improved, the dosage of cyclosporine A (CsA) was markedly decreased, and the mass lesions decreased in size and disappeared, thus suggesting that the mass lesions were an adverse reaction to CsA.  相似文献

A phytotoxin isolated from the Stretomyces sp. causing potato russet scab in Japan (Kamenoko-byo in Japanese) was identified as a 16-membered macrolide, FD-891. It induced necrosis in potato tuber slices above 50 µg/disk, which is about 1/100 the activity of thaxtomin A. The phytotoxin was detected in potatoes infected by the pathogen and was produced by other pathogenic strains with different geographic origins.  相似文献

To quantify the spatial variation and spatial structure of nitrous oxide (N2O) and nitric oxide (NO) emission from forest soils, we measured N2O and NO emission rates from surface soil cores taken at 1 m intervals on a cross-line transect (65 m × 20 m) on a slope of Japanese cedar ( Cryptomeria japonica ) forest in a temperate region of central Japan and analyzed the spatial dependency of N oxide gas emissions using geostatistics. We divided N2O emission into N2O from denitrification and N2O from nitrification using the acetylene inhibition method. According to the geostatistical analysis, N2O emission rates on the slope had large spatial variation and weak spatial dependency. This weak spatial dependency was caused by the inordinately high N2O emissions on the slope, which were derived mainly from denitrification. In contrast, NO emission rate on the slope had large spatial variation, but strong spatial dependency and a distinct spatial distribution related to slope position, that is, high in the middle of the slope and low in the shoulder and the foot of the slope. The CN ratio and water-filled pore space were the dominant factors controlling NO emission rate on a slope. Our results suggest that spatial information about topographic factors helps to improve the estimation of both N2O emission and NO emission from forest soils.  相似文献
