African elephants (Loxodonta africana) and large trees have ecological and human value, but elephant impact on large trees may influence conservation strategies. We used a self-administered questionnaire to examine perceptions of elephants, large trees, and elephant management endeavors among tourists and residents in the Associated Private Nature Reserves, South Africa to inform management approaches. The questionnaire included photographs of different elephant types and of vegetation with varying degrees of elephant-impact. Respondents reported high attractiveness rankings for all elephant types, but they scored non-impacted trees higher than photos of trees with visible elephant impact. Results also revealed a significant positive correlation between elephant types known to cause high tree impact and attractiveness of non-impacted trees. Residents favored more intrusive elephant management methods, with environmental manipulation representing the most supported and balanced option for both interest groups. Respondents advocated management that focuses on ecosystem functioning more so than elephant population control.  相似文献

In this study, in-house isolated laccase isoforms, i.e., Lac-I and Lac-II of the basidiomycete Pycnoporus sanguineus (CS43), were evaluated in relation to their Remazol Brilliant Blue R (RBBR) dye degradation capacity. A modified Dhouib medium additionally supplemented with 3% ethanol as a secondary inducer was used to propagate P. sanguineus CS43 for enhanced production of laccase under liquid state fermentation. The crude laccase extract was purified by passing through ion exchange diethylaminoethanol (DEAE)-Sepharose and gel filtration-based Sephadex G-200 column chromatography. The purified laccase fractions were subjected to the electrophoresis, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed two laccase isoforms Lac-I and Lac-II with 66 and 68 kDa, respectively. To explore the industrial applicability, for RBBR dye, degradation efficiencies ranged from 82 to 88% after 3 h of incubation for both; Lac-I and Lac-II at both concentrations were recorded. However, with 8 U/mL, the degradation ranged between 70 to 80% during the first 5 min of incubation. Enhanced degradation of RBBR dye was obtained in the presence of violuric acid and N-hydroxypthalamide as laccase mediators. Finally, using RBBR as a substrate kinetic characterization of both Lac-I and Lac-II isoforms was performed that revealed K m (0.243 and 0.117 mM for Lac-I and Lac-II) and V max (1.233 and 1.012 mM/Sec for Lac-I and Lac-II) values, respectively.  相似文献

BACKGROUND: The toxicity of a fusion protein, ButalT/GNA, comprising a venom toxin (ButaIT) derived from the red scorpion, Mesobuthus tamulus (F.), and Galanthus nivalis agglutinin (GNA), was evaluated under laboratory conditions against several pest insects. Insecticidal activity was compared with SFI1/GNA, a fusion comprising a venom toxin (SFI1) derived from the European spider Segestria florentina (Rossi) and GNA, which has been previously demonstrated to be effective against lepidopteran and hemipteran pests, and to GNA itself. RESULTS: Injection assays demonstrated that both fusion proteins were toxic to lepidopteran larvae, dipteran adults, coleopteran adults and larvae and dictyopteran nymphs. ButalT/GNA was more toxic than SFI1/GNA in all cases. GNA itself made a minor contribution to toxicity. Oral toxicity of ButalT/GNA towards lepidopteran pests was confirmed against neonate Spodoptera littoralis (Boisd.), where incorporation at 2% dietary protein resulted in 50% mortality and > 85% reduction in growth compared with controls. ButaIT/GNA was orally toxic to Musca domestica L. adults, causing 75% mortality at 1 mg mL?1 in aqueous diets and, at 2 mg g?1 it was orally toxic to Tribolium castaneum (Herbst.), causing 60% mortality and a 90% reduction in growth. CONCLUSIONS: Toxicity of the ButaIT/GNA recombinant fusion protein towards a range of insect pests from different orders was demonstrated by injection bioassays. Feeding bioassays demonstrated the potential use of the ButaIT/GNA fusion protein as an orally active insecticide against lepidopteran, dipteran and coleopteran pests. These experiments provide further evidence that the development of fusion protein technology for the generation of new, biorational, anti‐insect molecules holds significant promise. © Crown Copyright 2009. In this study three different approaches were employed to identify key odorants in Sauvignon blanc wines. First, the concentrations of the odorants were compared to their respective aroma detection thresholds. The resulting odor activity values (OAV) were transformed into a normalized and weighted measure that allows the aroma profiles of different wines to be compared and the contribution of a single aroma in a complex mixture to be evaluated. Based on their OAV, 3-mercaptohexanol and 3-mercaptohexyl acetate were the two most important aroma compounds in many Marlborough Sauvignon blanc wines. Due to limitations with the OAV approach, the study was extended to include aroma extract dilution analysis (AEDA), which revealed that β-damascenone, together with the varietal thiols, esters, and higher alcohols, are key odorants in Sauvignon blanc wines. The final approach undertaken was aroma reconstitution and omission tests using a deodorized wine base and the creation of a model Marlborough Sauvignon blanc. Single compounds and groups of compounds were omitted from the model to study their impact on the sensory properties of the model wine. Reconstitution and omission confirmed that varietal thiols, esters, terpenes, and β-damascenone are all important contributors to Sauvignon blanc aroma. The methoxypyrazines showed an important but relatively low impact in all three of the approaches undertaken in this study.  相似文献

Global declines in shark populations have created uncertainty in the future status of many species and conservation efforts are urgently needed. Marine protected areas (MPAs) are used increasingly as conservation tools around the world, but how they benefit mobile and wide ranging species like sharks remains unclear. To evaluate the degree of protection MPAs may provide for sharks, we used an array of acoustic receivers to examine the movements and spatial use of two tropical coastal species within two MPAs in the Great Barrier Reef Marine Park, Australia. Juvenile pigeye (Carcharhinus amboinensis) and adult spottail (Carcharhinus sorrah) sharks were fitted with acoustic transmitters from 2009 to 2010. Both species displayed long-term use of MPAs, with some individuals detected for longer than 600 days. The mean percentage of time C. amboinensis and C. sorrah spent inside MPAs was 22% and 32%, respectively. MPA use varied seasonally, with C. amboinensis spending a higher percentage of time inside MPAs in summer (mean = 28%) and C. sorrah spending a higher percentage of time inside MPAs in winter (mean = 40%). Although sharks used large areas inside MPAs, most individuals tended to use only half of the available protected space. In addition, all sharks made excursions from MPAs and individuals exited and re-entered at consistent locations along the MPA boundaries. These results demonstrate that MPAs have conservation benefits for shark populations by providing protection across different species and life stages, and tracking studies can be used to help tailor MPA design to maximize effectiveness.  相似文献
