Retinoids show antitumor effects on human acute promyelocytic leukemia and other tumors via retinoid receptors. In dogs, the role of retinoid receptors in inhibiting tumor development remains unclear. To evaluate the correlation between the degree of expression of retinoic acid receptor alpha (RARalpha) mRNA and the antiproliferative effects of all-trans retinoic acid (ATRA) treatments, expression analysis of RARalpha mRNA and cell growth inhibition assay were performed on 17 established canine tumor cell lines, including 6 mammary gland tumor (MGT) cell lines, 3 osteosarcoma cell lines, 5 melanoma cell lines, and 3 mast cell tumor (MCT) cell lines. Among the cell lines investigated, all 3 MCT cell lines showed high expression of RARalpha, and the most effective cell growth inhibition was observed in ATRA-treated MCT cell lines. However, remarkable antiproliferative effects of ATRA treatments were not observed on other tumor cell lines with moderate or low RARalpha mRNA expression. As a result of the relationship between RARalpha mRNA expression and ATRA treatment with regression analysis, statistically significant correlation was suggested. Furthermore, real-time quantitative polymerase chain reaction analysis of RARalpha was performed on MCT tissue samples of dogs with spontaneous disease, and 5 of 9 tissues showed high expression. These results suggest that ATRA may be an effective antitumor agent for MCT in dogs, and that prior measurement of expression of RARalpha mRNA may be a good indicator of the effectiveness of ATRA treatment.  相似文献

Appearance of albino seedlings caused by plastome-nuclear genome incompatibility was avoided by using polyploid parents in intersubgeneric hybridization between evergreen azaleas and deciduous Rhododendron japonicum f. flavum. Intra- and interploid crosses of 2x or 4x evergreen azaleas with 2xR. japonicum f. flavum showed high capsule set, but their reciprocal crosses failed to set capsules. Green and albino hybrids were obtained from 2x evergreen azaleas × 2xR. japonicum f. flavum crosses where ptDNAs of the green and albino plants were derived from R. japonicum f. flavum (paternal parent) and evergreen azaleas (maternal parents), respectively. All the progenies from the crosses of 4x evergreen azaleas × 2xR. japonicum f. flavum were green triploids with evergreen azalea-specific ptDNA. The efficiency of obtaining green hybrids in interploid crosses was higher than that in intraploid crosses. These results suggest that interploid crosses of 4x evergreen azaleas × 2xR. japonicum f. flavum can overcome plastome-nuclear genome incompatibility between evergreen azalea-specific plastome and the nuclear genome in the resulting hybrid.  相似文献

The motion of a magnetic domain wall in a submicrometer magnetic wire was detected by use of the giant magnetoresistance effect. Magnetization reversal in a submicrometer magnetic wire takes place by the propagation of a magnetic domain wall, which can be treated as a "particle." In several primates and carnivores, pronation/supination angles of the forearm skeleton were examined, and it is thought that a larger angle is useful to acquire dexterous behaviors in feeding and/or life style, including climbing. In this study, the pronation/supination angles in Asiatic black, brown and polar bears were nondestructively examined. These specimens were classified as adult or non-adult. Three or four carcasses of each group of Asiatic black and brown bears were used for CT analysis, whereas only one adult polar bear was used. The forearms were positioned within the gantry of a CT scanner in both maximally supinated and pronated states. Extracted cross-sectional CT images of two positions were superimposed by overlapping the outlines of each ulna. The centroids of the radii were detected, and then the centroid of each radius and the midpoint of a line which connects between both ends of the surface of each radius facing the ulna, were connected by lines to measure the angle of rotation as an index of pronation/supination. In adult brown and polar bears, the angles were smaller as compared with the other groups (Asiatic black and non-adult brown bears). Asiatic black and non-adult brown bears can climb trees, whereas adult brown bears and polar bears cannot. This suggests that the pronation/supination angle is related to arboreal activity in Ursidae.  相似文献

ABSTRACT Sixty strains of Xanthomonas oryzae pv. oryzae, collected from 29 locations in Sri Lanka in 1995, were analyzed by restriction fragment length polymorphism using either polymerase chain reaction-amplified 16S and 23S rDNA or the repetitive DNA element IS1112 from X. oryzae pv. oryzae as hybridization probes. Two different ribogroups were observed in the Sri Lankan strains using rDNA probes, whereas five clusters were identified by the IS1112 probe. Bootstrap analysis revealed that the five clusters defined by IS1112 were relatively robust. Our results suggest that the Sri Lankan strains are phylogenetically composed of five different groups. Each cluster was partially associated with climatic conditions (intermediate zone and wet zone) and was related to groups based on ribotyping. Based on virulence analysis using 12 rice cultivars, each containing a single resistance gene, 14 pathotypes were identified among the Sri Lankan strains. All strains were virulent to resistance genes Xa1, Xa2, Xa4, Xa10, Xa11, and Xa14. Only one strain (pathotype 1) was virulent to all major resistance genes including Xa21, while strains of the other pathotypes were all avirulent to Xa21. A partial relationship was found between the determined phylogenetic groups using the IS1112 probe and pathotypes for all but two clusters. The results of this study will facilitate the further understanding of the population structure of X. oryzae pv. oryzae in Sri Lanka.  相似文献

Changes in stand structure with altitude have been studied at ten sites between 800 and 2,500 m above sea level (a.s.l.) in temperate forests on Mount Norikura (36°06′N, 137°33′E, 3,026 m a.s.l.) in central Japan. Vegetation in this range of altitudes was roughly classified as a montane deciduous broad-leaved forest zone between 800 and 1,600 m a.s.l. and a subalpine coniferous forest zone between 1,600 and 2,500 m a.s.l. at the timberline. The frequency distribution of trunk height was an L-shaped pattern at 800 m a.s.l., changed to a flat pattern with increasing altitude up to 2,000 m a.s.l., but changed to an L-shaped pattern again from 2,000 to 2,500 m a.s.l. This change in frequency distribution of trunk height with altitude was related to the change in maximum trunk height with altitude. The maximum trunk height did not change with altitude between 800 and 2,000 m a.s.l. but it decreased markedly from 2,000 to 2,500 m a.s.l. Although mechanical damage to conifer trunks and branches was not observed between 800 and 2,000 m a.s.l., the proportion of damaged trees increased from 2,000 to 2,500 m a.s.l., suggesting that subalpine conifers cannot grow in height near the timberline because of mechanical damage. The increase in the number of small trees from 2,000 m a.s.l. to the timberline was therefore because of a less developed canopy structure, i.e. small trees can grow without shading by canopy trees. This study suggests that mechanical damage, probably because of strong winds in the winter, affects changes with altitude in regeneration and formation of the timberline.  相似文献

PCR-RFLP analysis was conducted to clarify the relationship between the leaf color of progenies and their ptDNA inheritance in interspecific three-way crosses, (Rhododendron kiusianum × R. eriocarpum) × R. japonicum f. flavum. All albino progenies contained maternal ptDNA, whereas green and pale-green progenies contained paternal ptDNA. Sectorial chimeric progenies, of which the leaf and shoot color was turned from green to albino during the culture, contained both maternal and paternal ptDNA in green segments and maternal ptDNA in albino segments. These results suggest that albino progenies are caused by the incompatibility between plastome from F1 hybrids of R. kiusianum × R. eriocarpum and nuclear genome from R. japonicum f. flavum. Protein expression and subcellular localization of E-cadherin, alpha-catenin, and beta-catenin in 8 feline mammary tumor cell lines were examined by western blot analysis and fluorescence immunocytochemistry. A low E-cadherin expression was observed in FNN-m cells. Furthermore, compared to other cell lines, two E-cadherin bands existed in FMC-p1 cells and were localized in the perinuclear region; distinct radial lines were observed in the cytoplasm. A low alpha-catenin expression was observed in FON-m cells, but there were no apparent abnormalities in its localization. In contrast, similar levels of beta-catenin expression and cytoplasmic localization were observed in all cell lines.  相似文献

Interleukin-3 (IL-3) binds to its receptor with high and low affinities, induces tyrosine phosphorylation, and promotes the proliferation and differentiation of hematopoietic cells. A binding component of the IL-3 receptor was cloned. Fibroblasts transfected with the complementary DNA bound IL-3 with a low affinity [dissociation constant (Kd) of 17.9 +/- 3.6 nM]. No consensus sequence for a tyrosine kinase was present in the cytoplasmic domain. Thus, additional components are required for a functional high affinity IL-3 receptor. A sequence comparison of the IL-3 receptor with other cytokine receptors (erythropoietin, IL-4, IL-6, and the beta chain IL-2 receptor) revealed a common motif of a distinct receptor gene family.  相似文献
