ABSTRACT

Temperatures and solar radiation during ripening critically affect grain appearance in rice (Oryza sativa L.). Climatic factors to induce chalky grains were analyzed under the experimental conditions of high-temperature and shading treatment and also under the ambient condition in a high-temperature-prone region of Japan. The frequency of white-back (WB) and basal-white (BW) grains correlated with temperature and solar radiation, whereas that of milky-white (MW) grains was not correlated, suggesting that complicated climatic factors are involved in the formation of MW grains. Further investigation was carried out to identify the parameters that distinguish perfect and MW grains grown in high-temperature versus those grown in low-solar-radiation conditions. As reported previously, the chalk phenotypes in the transverse section of the MW grains were quite different between environments: oval-shaped chalk for MW grains grown in low-solar-radiation condition and center chalk for MW grains grown in high-temperature condition. Grain hardness and amylopectin chain-length distribution did not explain the difference in MW grains between environments. MW grains subjected to high temperatures had a lower protein content without a consistent reduction in the single-grain weight, whereas those from the low-solar-radiation condition had a lower amylose content with a consistent reduction in the single-grain weight, when compared with perfect grains that developed in either environmental condition. Overall, our results suggest that MW grains are formed through different physiological mechanisms with altered starch and protein synthesis under high-temperature and low-solar-radiation conditions.

Endosperm texture is an important factor governing the end-product quality of cereals. The texture of wheat (Triticum aestivum L.) endosperm is controlled by puroindoline a and b genes which are both absent in rice (Oryza sativa L.). It has been reported that the endosperm texture of rice can be modified by puroindoline genes. The mechanism, however, by which puroindolines affect the ultrastructure of rice endosperm cells remains to be investigated. In this study, we observed the ultrastructure of endosperm cells and the morphology of isolated starch granules of the transgenic rice expressing the puroindoline b gene. SEM and TEM observations indicated that compound starch granules were embedded within the matrix material in non-transgenic rice, Nipponbare, whereas they were surrounded by spaces in the transgenic rice. The morphology and size of each starch granule were not different between non-transgenic and the transgenic rice. However, the transgenic rice flour showed smaller particle size, higher starch damage, and lower viscosity during gelatinization than that of non-transgenic rice. These results confirm that puroindoline b reduces the grain hardness in rice. Moreover, the results also suggest that puroindoline b functions at the surface of compound starch granules, and not on polygonal starch granules in rice endosperm.  相似文献

A 12-year-old neutered male shih tzu developed progressive pelvic limb paraparesis. Computed tomography showed a radiolucent mass lesion in the spinal canal at the left side of the 11th thoracic vertebra. The mass was not enhanced by intravenous contrast medium injection. It was hyperintense on both T1- and T2-weighted magnetic resonance images. The signal intensity of the mass was decreased with a fat suppression technique, indicating a fatty origin. After removal of the mass via T11-T12 hemilaminectomy, chronic panniculitis was confirmed by histopathological examination. This case demonstrates the utility of computed tomography and magnetic resonance imaging for the diagnosis of spinal canal pyogranulomatous inflammation.  相似文献

RNA interference (RNAi) technology using small interfering RNAs (siRNA) has been widely used as a powerful tool to knock down gene expression in various organisms. In pig preimplantation embryos, no attempt to suppress the target gene expression with such technology has been made. The purpose of this study is to demonstrate that the RNAi technology is useful for suppression of endogenous target gene expression at an early stage of development in pigs. Alpha-1,3-Galactosyltransferase (α-GalT) is an enzyme that creates the Galα1-3Gal (α-Gal) epitope on the cell surface in some mammalian species, and removal of the epitope is considered to be a prerequisite for pig-to-human xenotransplantation. We decided to suppress the endogenous α-GalT mRNA expression in pig early embryos, since reduction of α-GalT synthesis is easily monitored by cytochemical staining with Bandeiraea simplicifolia isolectin-B(4), a lectin that specifically binds to the α-Gal epitope, and by RT-PCR analysis. Cytoplasmic microinjection of double-stranded RNA and pronuclear injection of an siRNA expression vector into the embryos generated in vitro resulted in a significant reduction in expression of the α-GalT gene and α-Gal epitope in blastocysts, at which stage the α-Gal epitope is abundantly expressed. Somatic cell nuclear transfer of embryonic fibroblasts stably transfected with an siRNA expression vector also led to a significant reduction in the level of α-GalT mRNA synthesis together with decreased amounts of the α-Gal epitope at the blastocyst stage. These results indicate that the RNAi technology is useful for efficient suppression of a target gene expression during embryogenesis in pigs and suggest the possibility of production of siRNA-expressing pigs for use in xenotransplantation.  相似文献

Genetic parameters for 54 carcass and chemical traits, such as general composition (moisture, crude fat and crude protein), fatty acid composition and water‐soluble compounds (free amino acids, peptides, nucleotides and sugars) of 587 commercial Japanese Black cattle were assessed. Heritability estimates for carcass traits and general composition ranged between 0.19–0.28, whereas those for fatty acid composition ranged between 0.11–0.85. Most heritability estimates for water‐soluble compounds were lower than 0.30; these traits were affected by aging period. Moderate heritability was observed for glutamine, alanine, taurine, anserine, inosine 5′‐monophosphate (IMP), inosine and myo‐inositol. In particular, heritability estimates were the highest (0.66) for taurine. Traits with moderate heritability were unaffected by aging period, with the exception of IMP, which was affected by aging period but exhibited moderate heritability (0.47). Although phenotypic correlations of water‐soluble compounds with carcass weight (CW), beef marbling standard (BMS) and monounsaturated fatty acid were generally low, genetic correlations between these traits were low to high. At the genetic level, most of the water‐soluble compounds were positively correlated with monounsaturated fatty acid but negatively correlated with CW and BMS. Thus, our results indicate that genetic variance and correlations could exist and be captured for some of the water‐soluble compounds.  相似文献

Soil microbial communities in an apple orchard and its adjacent boundary bush with or without infestation by violet root rot were investigated for 2 years. Effects of season (spring, summer, and fall), land-use (apple orchard and boundary bush), and violet root rot (infested and healthy) on soil microbial populations, microbial activity, and microbial community structures were determined using physiological, cytochemical, and molecular (PCR-DGGE) approaches. Seasonal fluctuations were significant (P<0.05) in viable bacteria and fungal populations, bacterial FAME, fluorescein diacetate (FDA) hydrolysis, and diversity (H′) and evenness (J′) of community-level physiological profile (CLPP) in both years. However, seasonal differences of soil microbial guilds that utilize carbon substrate groups observed in the first year were not reproduced in the second year. The land-use factor differentiated the apple orchard from the boundary bush where viable bacterial population, bacterial FAME and FDA hydrolysis were significantly greater in both years. Infestation status of violet root rot, on the other hand, significantly increased bacterial FAME and FDA hydrolysis in both years. In addition, neither the land-use nor the disease infestation factor significantly influenced the utilization patterns of individual substrate guilds for the 2 years. In both years, saturated fatty acids were significantly more abundant in the orchard than in the bush soil, and monosaturated fatty acids vice versa. Principal component analyses for CLPP, FAME, and denaturing gradient gel electrophoresis (DGGE) consistently exhibited that, although the violet root rot influenced the soil microbial community structures both in the apple orchard and the boundary bush, overall magnitude of the difference in communities between the violet root rot infested and non-infested sites in the bush were greater than in the orchard, irrespective of the season. These results suggested that the seasonal and the land-use factors affected soil microbial community both quantitatively and qualitatively, whereas the impact of the violet root rot on the soil microbial community was mainly qualitative and more pronounced in the adjacent bush than in the orchard.  相似文献

Genomic DNAs of 14 mammary tumors were analyzed by Southern blot hybridization using a human c-yes-1 oncogene probe. The amplification was successful in half of the cases (7 adenocarcinomas). The degree of amplification was approximately 4-fold, and a high proportion was seen in malignant tumors. In addition, DNA polymorphism was detected in two adenocarcinomas.  相似文献

The influence of transport stress on rectal temperature, respiratory rate and serum concentrations of cortisol, thyroxine and triiodothyronine (T3) in normal pigs (NN) and in heterozygous (Nn) pigs with Halothane gene (n), a major gene that causes the pale, soft, exudative meat (PSE), were examined in a hot environment (32–34°C). Easily releasable myofilament content of Longissimus dorsi taken by needle biopsy was also measured as an index of myofibrillar disassembly. Rectal temperature and respiratory rate were significantly increased by the transport. However, the influence was higher (P < 0.01) in Nn pigs. Serum concentration of cortisol was increased (P < 0.01) by the transport despite the genotypes. Serum concentration of T3 was not changed by the transport in Nn pigs, whereas in NN pigs it was significantly decreased after the transport (P < 0.01). Easily releasable myofilament content after the transport was higher in Nn pigs than that of NN pigs (P < 0.05). These results suggest that response of thyroid function might relate to the stimulation of muscle proteolysis and PSE in Nn pigs under stressful conditions.  相似文献

Most methods for determining cardiac output (CO) have limited application in clinical practice due to the invasive techniques required. This study compared the thermodilution technique (TDCO) with three noninvasive methods for determining CO in anesthetized dogs: transthoracic bioimpedance (BICO), partial CO2 rebreathing (NICO), and transesophageal echocardiography (TEECO). TDCO was compared to BICO, NICO, and TEECO in six adult sevoflurane anesthetized beagle dogs (9.1–13.0 kg). All dogs were administered midazolam [0.3 mg kg?1, intravenously (IV)] and butorphanol (0.1 mg kg?1 IV), followed by ketamine (5.0 mg kg–1 IV) and sevoflurane in nitrous oxide (1 L minute–1) and oxygen (1 L minute–1) and mechanically ventilated. Dogs were maintained at 2.2% end‐tidal sevoflurane (ETsev) concentration for instrumentation and baseline measurements. Low (5.0% ETsev), intermediate (3.3% ETsev), and high cardiac output values were achieved by varying the end‐tidal sevoflurane concentration and the administration of dobutamine (3–10 g kg–1 minute–1 and 2.2% ETsev). A minimum of thirty data sets was obtained for each comparison. The correlation coefficients when compared to TDCO were 0.684 for BICO (p < 0.0001), 0.883 for NICO (p < 0.0001), and 0.991 for TEECO (p < 0.0001). Cardiac output values ranged 50–444 mL kg–1 minute–1 for TDCO, 100–253 mL kg–1 minute–1 for BICO, 64–214 mL kg–1 minute–1 for NICO, and 52–401 mL kg–1 minute–1 for TEECO. The differences when compared to TDCO ranged – 62–235 mL kg?1minute?1 for BICO, 18–220 mL kg?1 minute?1 for NICO, and – 35–32 mL kg–1 minute–1 for TEECO. Differences were maximum at the highest CO in BICO and NICO. In conclusion, this study demonstrated that BICO and NICO underestimate CO in sevoflurane anesthetized dogs. TEECO is a viable noninvasive method for determining CO in sevoflurane anesthetized dogs.  相似文献

Crude fat content of longissimus (ribeye) muscle of beef cattle was predicted from a ratio of fat area (RFA) to area of ribeye muscle calculated from computer image analysis (CIA). Cross sections of 64 ribeyes taken from the 6-7th rib from cattle at experiment station A and cross sections of 94 ribeyes taken from the 6-7th rib from cattle at Experiment Station B were used in this study. Slices (1 to 1.5 cm in thickness) of just the Longissimus dorsi were homogenized and sampled for chemical estimation of crude fat content using petroleum ether. Crude fat content as determined from chemical analysis was used as the true estimate of fat content. A CCD (charge-coupled device) camera was used as the input device at Experiment Station A, and a single-lens reflex camera was used at Experiment Station B to photograph ribeyes for CIA. The contour comparison method, which assigns a threshold value for each marbling particle, was used to obtain accurate binarization in this study. Minimum and maximum of chemical measurements of crude fat were 2.1 and 39.8%, and for CIA calculation of the RFA were 6.1 and 56.8%, respectively. This range covered almost the complete range of the beef marbling standard used in carcass grading in Japan. The equation for the regression of the crude fat content (Y) on RFA (X) calculated from CIA for all of the data was Y = .793X-3.04 with r2 = .96. Regression equations for prediction of crude fat percentage from RFA taking into consideration the effect of experiment station were Y = .741X-2.22 with r2 = .91 for Experiment Station A, and Y = .782X-2.54 with r2 = .91 for Experiment Station B. Analysis of covariance showed that the effects of experiment stations on intercepts and slopes were not significant (P > .10). The ranges of differences between actual and predicted crude fat content from the prediction equation that was calculated without consideration of the effect of station were -6.4 to 4.0%. CIA of cross sections of the ribeye muscle seems to have potential for prediction of crude fat content.  相似文献
