The mobility, bioavailability, and environmental fate of heavy metals in soil are controlled by their adsorption onto soil minerals and solid organic matter. The adsorption is strongly affected by the presence of various low-molecular-weight organic acids. In this study, effect of hydroxamate siderophore desferrioxamine B (DFOB) on cadmium (Cd) and zinc (Zn) adsorption onto two micaceous clay minerals, muscovite and phlogopite, was evaluated in batch experiments. Results showed that the presence of DFOB diminished the adsorption of Cd and Zn onto both minerals, particularly under neutral to alkaline pH conditions. For instance, at pH 8.2, the presence of DFOB caused a decrease in the adsorption of Zn onto phlogopite by nearly 50%. The equilibrium adsorption of Cd and Zn was satisfactorily described using Freundlich isotherm. The adsorption isotherms showed that the affinity of Cd and Zn onto the minerals decreased in the presence of DFOB. For example, at pH 8.0, the presence of siderophore caused a decrease in the Freundlich adsorption isotherm coefficient KF for Zn adsorption onto muscovite and phlogopite from 4.60 to 0.07 L g-1 and from 3.56 to 0.36 L g-1, respectively. These findings confirm the potential influence of siderophore on the fate of Cd and Zn in arid soils containing substantial contributions of micaceous silicate minerals.  相似文献

Field experiments were conducted to evaluate the tolerance of strawberry cultivars to terbacil. Treatments (84, 168, and 336 g ha–1) were applied at either dormant stage or 3–4 leaf. Initial crop injury at 1 and 3 weeks after treatment (WAT) ranged between 0% and 43%; all cultivars fully recovered at 6 WAT. Terbacil applied at 168 g ha–1 at dormant stage caused approximately 20% yield reduction in ‘Brunswick’, ‘Darselect’, and ‘Honeoye’, which was not observed with other treatments. A negative impact of the herbicide on yield of these cultivars is an unlikely outcome when sprayed at 3–4 leaf stage.  相似文献

This study was carried out to evaluate the use of commercial live bakers’ yeast, Saccharomyces cerevisiae as a growth and immunity promoter for Nile tilapia, Oreochromis niloticus (L.). Fish (0.33 g) were randomly distributed at 25 fish per 140-L aquarium and fed a diet containing 0.0, 0.25, 0.50, 1.0, 2.0, or 5.0 g yeast/kg diet for 12 weeks. After the 12-week experimental period, fish of each treatment were challenged by pathogenic Aeromonas hydrophila, which was given by interperitoneal (IP) injection and kept under observation for 10 days to record clinical signs and the daily mortality rate. The growth-promoting influences of bakers’ yeast were observed with fish and the optimum growth, feed utilization, and protein turn-over were obtained with 1.0–5.0 g yeast/kg diet. Also, yeast supplementation increased protein deposition in fish body. Biochemical parameters were improved in fish fed yeast up to 1.0 g/kg diet. Total fish mortality 10-days after IP injection with A. hydrophila and its count after incubation with fish serum decreased with the increase of yeast level in fish diets. However, the lowest fish mortality and bacterial counts were obtained in fish fed 5.0 g yeast/kg. These results indicate that bakers’ yeast supplement is promising as an alternative method to antibiotics for disease prevention in tilapia aquaculture, and the optimum level of live bakers’ yeast is about 1.0 g per kg diet.  相似文献

The ability of plants to extract water from soil is controlled by the water‐potential gradient between root and soil, by the hydraulic conductivity of roots, and, as the soil dries, by that of the soil near the roots (rhizosphere). Recent experiments showed that the rhizosphere turned hydrophobic after drying and it remained temporarily dry after rewetting. Our objective was to investigate whether rhizosphere hydrophobicity is associated with a reduction in root water uptake after drying and rewetting. We used neutron radiography to trace the transport of deuterated water (D2O) in the roots of lupines growing in a sandy soil. The plants were grown in aluminum containers (28 × 28 × 1 cm3) filled with a sandy soil. The soil was initially partitioned into different compartments using a 1‐cm layer of coarse sand (three vertical × three horizontal compartments). We grew plants in relatively moist conditions (0.1 < θ < 0.2). Three weeks after planting, we let the upper left compartment of soil to dry for 2–3 d while we irrigated the rest of the soil. Then, we injected D2O in this compartment and in the upper right compartment that was kept wet. We monitored D2O transport in soil and roots with time‐series neutron radiography. From the changes of D2O concentration inside roots, we estimated the root water uptake. We found that root water uptake in the soil region that was let dry and rewetted was 4–8 times smaller than that in the region that was kept moist. The reduced uptake persisted for > 1–0.5 h. We conclude that a reduction in hydraulic conductivity occurred during drying and persisted after rewetting. This reduction in conductivity could have occurred in roots, in the rhizosphere, or more likely in both of them.  相似文献

Background:Some resources have suggested that genetically inactivated PTs bear a more protective effect than chemically inactivated products. This study aimed to produce new version of PT, by cloning an inactive PTS1 in a fusion form with N-terminal half of the LLO pore-forming toxin. Methods:Deposited pdb structure file of the PT was used to model an extra disulfide bond. Codon-optimized ORF of the PTS1 was used to make recombinant constructs of PTS1 and LLO-PTS1 in the pPSG-IBA35 vector. The recombinant PTS1 and LLO-PTS1 proteins were expressed in BL21 DE3 and SHuffle T7 strains of E. coli and purified by affinity chromatography. Cytotoxic effects of the recombinant proteins were examined in the MCF-7 cell line. Results:The purity of the products proved to be more than 85%, and the efficiency of the disulfide bond formation in SHuffle T7 strain was higher than BL21 DE3 strain. No cytotoxicity of the recombinant proteins was observed in MCF-7 cells. Soluble recombinant PTS1 and LLO-PTS1 proteins were produced in SHuffle T7 strain of E. coli with high efficiency of disulfide bonds formation. Conclusion:The LLO-PTS1 with corrected disulfide bonds was successfully expressed in E. coli SHuffle T7 strain. Due to the safety for human cells, this chimeric molecule can be an option to prevent pertussis disease if its immunostimulatory effects would be confirmed in the future. Fusarium head blight (FHB) caused by Fusarium spp. is one of the most important fungal diseases of wheat (Triticum aestivum L.) in regions with wet climatic conditions. Improvement of the FHB resistance by developing new varieties requires sound knowledge on the inheritance of resistance. An 8 × 8 diallel analysis was performed to estimate general (GCA) and specific (SCA) combining ability of resistance to FHB. The F1s and parental lines were evaluated under artificial inoculation at the experimental field of IFA-Tulln, Austria during 2001 and 2002. Disease severity was evaluated by repeated scoring of the percentage of infected spikelets and calculating an area under the disease progress curve (AUDPC). The analysis of combining ability across two years showed highly significant GCA and non-significant SCA effects indicating the importance of additive genetic components in controlling FHB resistance. The significant GCA-by-year interaction presented the role of environmental factors in influencing the FHB reaction of wheat lines. The comparison of the crosses with low FHB infection and GCA effects of their parents showed that such crosses involved at least one parent with high or average negative GCA effect. The results revealed that it is feasible to use highly or moderately resistant genotypes and conventional breeding methods to achieve genetic improvement of FHB resistance in spring wheat.  相似文献

Potassium (K+) directly released from primary K‐bearing minerals can contribute to plant nutrition. The objective of this research was to assess short‐term K+ release and fixation on a range of intensively cropped calcareous soils. Potassium sorption and desorption properties and the contributions of exchangeable‐K+ (EK) and nonexchangeable‐K+ (NEK) pools to K+ dynamics of the soil‐solution system was measured using a modified quantity‐to‐intensity (Q : I) experiment. Release and fixation of K+ were varied among soils. The relation between the change in the amount of NEK during the experiment and the initial constrain was linear, and soil ability for K+ release and fixation (β) for all soils varied from 0.041 to 0.183, indicating that 4% to 18% of added K+ converted to NEK when fixation occurred. The equilibrium potential buffering capacity (PBC) for K+ derived from Q : I experiments had significant correlation (r = 0.75, p < 0.01) with β, indicating that PBC depends not only on exchange properties but also on release and fixation properties. The depleted soils showed higher β value than the other soils, indicating much of the added K+ was converted to NEK in case of positive constraint. The range of the amount of EK which was not in exchange equilibrium with Ca (Emin) in the experimental conditions was large and varied from 0.68 to 9.00 mmol kg–1. On average, Emin amounted to 64% of EK. This fraction of EK may not be available to the plant. The parameters obtained from these short‐term K+ release and fixation experiments can be used in plant nutrition.  相似文献
