Melanoidins, the brown polymers formed through Maillard reaction during coffee roasting, constitute up to 25% of the coffee beverages' dry matter. In this study chemical characterization of melanoidins obtained from light-, medium-, and dark-roasted coffee beans, manufactured from the same starting material, was performed. Melanoidins were separated by gel filtration chromatography and studied by MALDI-TOF mass spectrometry. Results showed that the amount of melanoidins present in the brews increased as the intensity of the thermal treatment increased, while their molecular weight decreased. The antioxidant activity of melanoidins isolated from the different brews was studied by using different methodologies. Melanoidins antiradical activity determined by ABTS(*)(+) and DMPD(*)(+) assays decreased as the intensity of roasting increased, but the ability to prevent linoleic acid peroxidation was higher in the dark-roasted samples. Data suggest that melanoidins must be carefully considered when the relevance of coffee intake in human health is studied. 相似文献

This study reports on the first use of the "optothermistor" as a novel, precise, fast, and low-cost detector of lycopene in a wide range of commercially available processed-tomato products. The quantitative performance of the new device was evaluated by comparing data obtained to that acquired by conventional methods, namely, absorption spectrophotometry and high-performance liquid chromatography (HPLC); the linear correlation was high (R = 0.98). The variation of data obtained with the optothermistor in a series of consecutive measurements performed with the same loading of the sample was better than 1%. However, the repeatability (RSD 0.5-9.0%, n = 3-5) achieved with the optothermistor by independent analyses (multiple loading) is comparable to that of HPLC and spectrophotometry. Results of the studies performed on the 19 products derived from tomatoes demonstrated that the optothermistor is suitable for selective, accurate, precise, and simple determination of lycopene (range = 7-75 mg/100 g of product weight) without the need for a sample pretreatment step. The estimated sensitivity of the present optothermistor is 2 mg of lycopene/100 g of product. 相似文献

Phloroglucinol from phlorin hydrolysis for testing quality of commercial orange juices and beverages

The European Association of Juices and Nectars Producers (AIJN) is evaluating the opportunity to introduce the content of phlorin (3,5-dihydroxyphenyl beta-D-glucopyranoside), a peel marker for oranges, as a parameter for testing the quality of orange juices. Because of the lack of a commercial standard of phlorin and its laborious isolation procedures, in this contribution is developed a simple and reliable method for measuring the phlorin level as the corresponding aglycon phloroglucinol, obtained after a total enzymatic hydrolysis of the sample. The method was applied to the quantification of phloroglucinol in several industrial and commercial blond and pigmented orange juices and beverages based on 12% orange juice. Under the same extraction procedure, the phloroglucinol content in the pigmented juices was higher than in the blond ones. No significant difference was obtained between not from concentrate juices and reconstituted from concentrate juices. The marker amount increases in the highly processed orange fruits and in the byproducts of citrus processing due to the contact of the juice with the albedo, which is the major source of phlorin. In orange-based beverages the phloroglucinol content revealed a large heterogeneity and a poor quality of the raw juices used. 相似文献

The reliability and efficiency of pressurized fluid extraction (PFE) technique for the extraction of total lipid content from cod and the effect of sample treatment on the extraction efficiency have been evaluated. The results were compared with two liquid-liquid extraction methods, traditional and modified methods according to Jensen. Optimum conditions were found to be with 2-propanol/n-hexane (65:35, v/v) as a first and n-hexane/diethyl ether (90:10, v/v) as a second solvent, 115 degrees C, and 10 min of static time. PFE extracts were cleaned up using the same procedure as in the methods according to Jensen. When total lipid yields obtained from homogenized cod muscle using PFE were compared yields obtained with original and modified Jensen methods, PFE gave significantly higher yields, approximately 10% higher (t test, P < 0.05). Infrared and NMR spectroscopy suggested that the additional material that inflates the gravimetric results is rather homogeneous and is primarily consists of phospholipid with headgroups of inositidic and/or glycosidic nature. The comparative study demonstrated that PFE is an alternative suitable technique to extract total lipid content from homogenized cod (lean fish) and herring (fat fish) muscle showing a precision comparable to that obtained with the traditional and modified Jensen methods. Despite the necessary cleanup step, PFE showed important advantages in the solvent consumption was cut by approximately 50% and automated extraction was possible. 相似文献

Viewing of polar bears (Ursus maritimus) from tundra vehicles has been offered at Churchill, Manitoba since the early 1980s. This form of wildlife viewing has provided a unique and safe way for tourists to learn about polar bears. However, these activities have largely been carried out without examining possible effects on polar bear behaviour. We studied vigilance behaviour (a scanning of the immediate vicinity and beyond) of resting polar bears to evaluate impacts from tundra vehicle activity. Focal animal sampling was used to examine whether a difference in vigilance behaviour existed when vehicles were present. We recorded the numbers of head-ups, vigilance bout length, and between-bout intervals for polar bears. In general, the frequency of head-ups increased, and the between-bout intervals decreased for male bears, when vehicles were present. Female bears behaved opposite to males. The vigilance bout lengths did not differ significantly between vehicle presence and absence. Vigilance behaviour of male bears was not magnified with increasing numbers of vehicles; therefore the threshold is one vehicle. We suggest that manipulative studies be conducted to examine how distances between vehicles and bears, tundra vehicle activity in the immediate vicinity of a bear during viewing, and noise of tourists affect increased vigilance. 相似文献

B-mode ultrasonography using a 7.5 megahertz (MHz ) transducer was performed on 95 equine cadaver eyes submerged in a water bath. Identification and measurement of the anterior to posterior diameters of the intraocular structures and of the axis bulbi was done. The dorsal ventral axis of the eye was also measured. The eyes were then frozen, sectioned in a saggital plane and the same measurements were made using calipers. Two sets of measurements were then compared statistically.
With the exception of the cornea and the dorsal ventral axis measurements of male horses there was no statistically significant difference in the measurements obtained by the two methods.
The use of B-mode ultrasonography at a frequency of 7.5 MHz to examine the architecture of the equine eye is suggested as a valuable clinical tool. Biometry of the intraocular structures at this frequency may be reasonably accurate. 相似文献

The objectives of this study were to determine how landscape patterns in Georgia, USA have changed through time and whether the spatial patterns varied by physiographic region. Historical aerial photography was used to analyze spatial patterns of land use from the 1930's to the 1980's. Land use patterns were quantified by: (1) mean number and size of patches; (2) fractal dimension of patches; (3) amount of edge between land uses; and (4) indices of diversity, dominance, and contagion. Forest cover increased in aerial extent and in mean patch size. The mean size of agricultural patches increased in the coastal plain and decreased in the mountains and piedmont. Edges between land uses decreased through time, indicating less dissection of the landscape. Fractal dimensions also decreased, indicating simpler patch shapes. Indices of diversity and dominance differed through time but not among regions; the contagion index differed among regions but not through time. A geographic trend of decreasing diversity and increasing dominance and contagion was observed from the mountains to the lower coastal plain. Landscape patterns exhibited the greatest changes in the piedmont region. Overall, the Georgia landscape has become less fragmented and more connected during the past 50 years. Changing patterns in the landscape may have implications for many ecological processes and resources. 相似文献

The relationship between a landscape process and observed patterns can be rigorously tested only if the expected pattern in the absence of the process is known. We used methods derived from percolation theory to construct neutral landscape models,i.e., models lacking effects due to topography, contagion, disturbance history, and related ecological processes. This paper analyzes the patterns generated by these models, and compares the results with observed landscape patterns. The analysis shows that number, size, and shape of patches changes as a function of p, the fraction of the landscape occupied by the habitat type of interest, and m, the linear dimension of the map. The adaptation of percolation theory to finite scales provides a baseline for statistical comparison with landscape data. When USGS land use data (LUDA) maps are compared to random maps produced by percolation models, significant differences in the number, size distribution, and the area/perimeter (fractal dimension) indices of patches were found. These results make it possible to define the appropriate scales at which disturbance and landscape processes interact to affect landscape patterns. 相似文献
