Continual discharge of textile wastewaters loaded with a variety of synthetic dyes and metals is considered as a huge threat to surrounding ecosystems. In order to treat these undesirable pollutants, microbial bioremediation is considered as an efficient and economical technique. This study was conducted to evaluate the use of bacterial strains for simultaneous removal of azo dyes and hexavalent chromium [Cr(VI)]. Fifty-eight bacterial strains were isolated from Paharang drain wastewater and tested for their potential to decolorize reactive red-120 (RR-120) in the presence of 25 mg L?1 of Cr(VI). Among the tested isolates, FA10 decolorized the RR-120 most efficiently and was identified as Acinetobacter junii strain FA10. Based on quadratic polynomial equation and response surfaces given by the response surface methodology (RSM), Cr concentration and pH were found to be the main factors governing the RR-120 decolorization by FA10. The strain FA10 also exhibited a substantial salt resistance since it showed a considerable decolorization of RR-120 even in the presence of 150 g L?1 of NaCl. Moreover, the strain FA10 also showed the potential to simultaneously remove the Cr(VI) and the selected azo dyes in the same medium. More than 80 % of the initially added Cr(VI) was removed over 72 h of incubation along with the appreciable decolorization efficiency. The strain FA10 also exhibited good tolerance to considerable levels of different heavy metals. The findings of this study suggest that the strain FA10 might serve as an efficient bioresource to develop the biotechnological approaches for simultaneous removal of different azo dyes and heavy metals including Cr(VI).  相似文献

Slag-type silicate fertilizer, which contains high amount of active iron oxide, a potential source of electron acceptor, was applied at the rate of 0, 2, 6, 10, and 20 Mg ha−1 to reduce methane (CH4) emission from rice planted in potted soils. Methane emission rates measured by closed chamber method decreased significantly with increasing levels of silicate fertilizer application during rice cultivation. Soil redox potential (Eh) decreased rapidly after flooding, but floodwater pH and soil pH increased significantly with increasing levels of silicate fertilizer application. Iron concentrations in potted soils and in percolated water significantly increased with the increasing levels of silicate fertilizer application, which acted as oxidizing agents and electron acceptors, and thereby suppressed CH4 emissions. Silicate fertilization significantly decreased CH4 production activity, while it increased carbon dioxide (CO2) production activity. Rice plant growth, yield parameters, and grain yield were positively influenced by silicate application levels. The maximum increase in grain yield (17% yield increase over the control) was found with 10 Mg ha−1 silicate application along with 28% reduction in total CH4 flux during rice cultivation. It is, therefore, concluded that slag-type silicate fertilizer could be a suitable soil amendment for reducing CH4 emissions as well as sustaining rice productivity and restoring the soil nutrient balance in rice paddy soil.  相似文献

为了解决太阳能喷灌系统应用中无法对喷头水力性能进行有效预测的问题,以光照强度为影响因素,太阳能喷灌系统泵出口流量、泵出口压力、喷洒射程、水量分布及系统灌溉均匀性系数为评价指标,通过在夏季典型天气25~36℃下的系统水力性能试验,寻找不同光照强度下系统喷洒水力性能的变化规律,获得系统最佳工作状态下所需光照强度.试验结果表明:随着光照强度的增大,系统流量及泵出口压力均增大,泵的流量和压力随光照强度的变化规律基本符合指数分布规律.当光照强度大于900.0 W/m2时,系统流量、泵出口压力、射程及水量分布基本保持不变.获得了平均光照强度与均匀性系数函数关系,当光照强度大于900.0 W/m2时,系统喷灌均匀系数大于88%.当光照强度为200.0~600.0 W/m2时,系统喷灌均匀系数为76%~82%.太阳能喷灌系统在光照强度大于200.0 W/m2时可正常工作.该研究为改善太阳能喷灌系统水力性能,促进太阳能喷灌系统在实际工程中的推广应用提供了参考.  相似文献

Global climate warming has been exerting impacts on agricultural pests. Pests also take some strategies to adapt to climate change. Understanding such adaptation could benefit more accurate predictions and integrated management of pest. However, adaptation to climate change has not been widely investigated in agricultural pests but has been well documented in model species, Drosophila, and reviewed by Hoffmann before 2003. To provide recent progress and references for agricultural entomologists who interested in thermal biology, here we have reviewed literatures since 2003 about adaptation to temperature changes under climate change. We mainly summarized thermal adaptation of Drosophila (especially to high temperatures) from three aspects, behaviors, plastic responses and micro-evolution and discussed how Drosophila increases their heat tolerance through these three mechanisms. Finally, we summarized the measures of thermotolerance and concluded the main progress in recent decade about the behavioral thermoregulation, mortality risks driven by limited evolutionary and plastic response under climate change, geographic distribution based on basal rather than plastic thermotolerance. We propose future work focus on better understanding adaptation of organisms including agricultural pests to climate change.  相似文献

为了能够得到高效降解二甲基二硫醚(Dimethyl disulfide,DMDS)的菌种资源,降低其残留在环境中带来的不利影响,从苏州某废弃农药厂附近的土壤中分离、纯化出一株能够降解DMDS的细菌,并对该细菌进行分子生物学和生理生化鉴定,研究其在不同的初始浓度、转速、pH、温度及外加碳氮源条件下的降解性能。结果表明:将分离纯化得到的DMDS降解菌命名为SZT-1,经过表型分析及16S rDNA基因序列同源性比对鉴定,该菌株序列与芽孢杆菌属(Bacillus Cohn)有98%以上的同源性。SZT-1菌株可以在以DMDS为唯一碳源的无机盐培养基中生长,56 h后进入生长稳定期。经摇床接种培养和一级动力学分析,接种SZT-1可以有效提高DMDS的降解速率,使其半衰期由346.5 h缩短至86.6 h;通过单因素环境条件分析,该菌株在DMDS初始浓度为250mg·L-1,转速为130 r·min-1,pH为5,温度为30℃,外加碳源为淀粉,外加氮源为蛋白胨时,对DMDS的降解效果最佳,降解率达50%。试验首次提出了SZT-1对DMDS具有一定的降解效果,在进行相关技术完善后,有望用于治理土壤、地下水中DMDS带来的危害。  相似文献

Background: The present study aimed to fabricate surface-modified chitosan nanoparticles with two mucoadhesive polymers (sodium alginate and polyethylene glycol) to optimize their protein encapsulation efficiency, improve their mucoadhesion properties, and increase their stability in biological fluids. Method: Ionotropic gelation was employed to formulate chitosan nanoparticles and surface modification was performed at five different concentrations (0.05, 0.1, 0.2, 0.3, 0.4% w/v) of sodium alginate (ALG) and polyethylene glycol (PEG), with ovalbumin (OVA) used as a model protein antigen. The functional characteristics were examined by dynamic light scattering (DLS), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM)/scanning transmission electron microscopy (STEM). Stability was examined in the presence of simulated gastric and intestinal fluids, while mucoadhesive properties were evaluated by in vitro mucin binding and ex vivo adhesion on pig oral mucosa tissue. The impact of the formulation and dissolution process on the OVA structure was investigated by sodium dodecyl-polyacrylamide gel electrophoresis (SDS-PAGE) and circular dichroism (CD). Results: The nanoparticles showed a uniform spherical morphology with a maximum protein encapsulation efficiency of 81%, size after OVA loading of between 200 and 400 nm and zeta potential from 10 to 29 mV. An in vitro drug release study suggested successful nanoparticle surface modification by ALG and PEG, showing gastric fluid stability (4 h) and a 96 h sustained OVA release in intestinal fluid, with the nanoparticles maintaining their conformational stability (SDS-PAGE and CD analyses) after release in the intestinal fluid. An in vitro mucin binding study indicated a significant increase in mucin binding from 41 to 63% in ALG-modified nanoparticles and a 27–49% increase in PEG-modified nanoparticles. The ex vivo mucoadhesion showed that the powdered particles adhered to the pig oral mucosa. Conclusion: The ALG and PEG surface modification of chitosan nanoparticles improved the particle stability in both simulated gastric and intestinal fluids and improved the mucoadhesive properties, therefore constituting a potential nanocarrier platform for mucosal protein vaccine delivery.  相似文献

Cotton is a crop of tropical and subtropical regions but the seed cotton yield is highly influenced by abiotic stresses like drought and heat. Response of cotton genome to abiotic stresses is highly complex and involve many genes. A comprehensive study, involving cotton genotypes developed through conventional and synthetic tetraploid method, was designed to (i) study the introgression of heat and water stress tolerance by using wild relatives (ii) evaluate genetic markers for marker assisted selection against water and heat stress. Two separate experiments for water and heat stress tolerance with a common control were established. Treatments in each experiment include a control and a stress treatment. Heat stress was applied by sowing crop two month earlier than the control treatment, whereas water stress was imposed by withholding alternate irrigation. Analyses of variance depicted highly significant (P ≤ 0.01) effect of genotypes and both stresses on boll retention, boll weight and seed cotton yield. Interaction of genotypes with stress in both experiments was also highly significant (P ≤ 0.01). Genotypes derived from interspecific crosses performed consistently in stress conditions compared to control which prove it a reliable method to introgress stress related genes from wild parents. Four genes reported for water stress tolerance and five genes reported for heat stress tolerance were evaluated by field results for efficient marker assisted selection (MAS). Results verified drought stress genes but heat stress genes could not explain genetic variability caused by heat stress. It is concluded from the results that separate genes may be responsible for heat stress tolerance for vegetative and reproductive stages, therefore, selection criteria should include both the traits.  相似文献

This study evaluated the use of hydrolysed rubber seed meal as a locally grown alternative feed ingredient to reduce the use of fishmeal within formulated feeds for tilapia. Five experimental diets were prepared by formulating different inclusion levels of hydrolysed rubber seed meal (0%, 25%, 50%, 75% and 100% of plant‐based protein contribution). Dried and defatted rubber seed meal was hydrolysed by adding rumen digesta liquid to the ingredient. For digestibility measurement, chromic oxide was added to each diet as an inert marker. The digestibility trial was conducted over a 14‐day period by collecting faecal matter from the rearing tanks. In addition, a growth trial was conducted over a period of 50 days using four replicates per dietary treatment. In both experiments, all fish were fed three times daily to apparent satiation. At the end of growth trial, the inclusion level of hydrolysed rubber seed meal up to 50% of plant‐based protein contribution did not give any significant difference compared to the control diet in terms of feed efficiency, specific growth rate and protein retention parameters. In the digestibility study, fish fed diets containing hydrolysed rubber seed meal up to a 50% of plant‐based protein inclusion level did not give any significant difference compared to the control diet in terms of protein and dry matter apparent digestibility. Based on the result of this growth and digestibility study, up to a 50% of plant‐based protein contribution of hydrolysed rubber seed meal can be used within formulated tilapia feeds without any adverse effect.  相似文献

BackgroundConditioned medium is the medium obtained from certain cultured cells and contained secretome from the cells. The secretome, which can be in the form of growth factors, cytokines, exosomes, or other proteins secreted by the cells, can induce the differentiation of cells that still have pluripotent or multipotent properties.ObjectivesThis study examined the effects of conditioned medium derived from E17 rat brain cells on cells with pluripotent properties.MethodsThe conditioned medium used in this study originated from E17 rat brain cells. The CM was used to induce the differentiation of primary colonies of mice blastocysts. Primary colonies were stained with alkaline phosphatase to analyze the pluripotency. The morphological changes in the colonies were examined, and the colonies were stained with GFAP and Neu-N markers on days two and seven after adding the conditioned medium.ResultsThe conditioned medium could differentiate the primary colony, beginning with the formation of embryoid-body-like structure; round GFAP positive cells were identified. Finally, neuron-like cells testing positive for Neu-N were observed on the seventh day after adding the conditioned medium.ConclusionsConditioned medium from different species, in this case, E17 rat brain cells, induced and promoted the differentiation of the primary colony from mice blastocysts into neuron-like cells. The addition of CM mediated neurite growth in the differentiation process.  相似文献

Fucoidans are sulfated, complex, fucose-rich polymers found in brown seaweeds. Fucoidans have been shown to have multiple bioactivities, including anti-inflammatory effects, and are known to inhibit inflammatory processes via a number of pathways such as selectin blockade and enzyme inhibition, and have demonstrated inhibition of inflammatory pathologies in vivo. In this current investigation, fucoidan extracts from Undaria pinnatifida, Fucus vesiculosus, Macrocystis pyrifera, Ascophyllum nodosum, and Laminaria japonica were assessed for modulation of pro-inflammatory cytokine production (TNF-α, IL-1β, and IL-6) by human peripheral blood mononuclear cells (PBMCs) and in a human macrophage line (THP-1). Fucoidan extracts exhibited no signs of cytotoxicity in THP-1 cells after incubation of 48 h. Additionally, all fucoidan extracts reduced cytokine production in LPS stimulated PBMCs and human THP-1 cells in a dose-dependent fashion. Notably, the 5–30 kDa subfraction from Macrocystis pyrifera was a highly effective inhibitor at lower concentrations. Fucoidan extracts from all species had significant anti-inflammatory effects, but the lowest molecular weight subfractions had maximal effects at low concentrations. These observations on various fucoidan extracts offer insight into strategies that improve their efficacy against inflammation-related pathology. Further studies should be conducted to elucidate the mechanism of action of these extracts.  相似文献
