Intracameral injection of tissue plasminogen activator has been suggested as a means of hastening the clearance of blood from the anterior chamber and reducing the risk of synechia and fibrotic membrane formation in horses with hyphaema. The following report describes a case of ocular trauma in which intracameral injection of tenecteplase (TNK), a new‐generation plasminogen activator, resulted in rapid resolution of hyphaema and successful outcome. However, a large dose (1 mg) was administered and may have been associated with retinopathy and keratopathy. TNK may be an effective treatment for equine hyphaema and further controlled clinical or experimental studies are required to determine its efficacy and safety profiles.  相似文献

Castration is an ancient husbandry procedure used to produce docile cattle for draught work, to reduce unwanted breeding, and to modify carcass quality. All the physical methods used to castrate cattle have side-effects and cause pain. The plasma cortisol response to castration using Burdizzo clamps and, by inference, the acute pain experienced, is less than that caused by surgical, rubber-ring or latex-band castration. The cortisol response may be influenced by the age of the animal castrated, but this has not been well defined. Local anaesthesia virtually eliminates the cortisol response, and thus the acute pain, caused by rubber-ring or latex-band castration, but needs to be combined with a systemic analgesic such as the non-steroidal anti-inflammatory drug ketoprofen to eliminate the cortisol response to Burdizzo or surgical castration. When used alone, ketoprofen sometimes reduces the cortisol response to Burdizzo or surgical castration but may need to be accompanied by local anaesthesia to eliminate the pain-induced behaviour seen during the castration process itself. Thus, pharmacological methods are available to virtually eliminate the acute pain experienced by calves during the 12 h following castration. The use of these methods is an additional cost for farmers and may be limited by the availability of drugs for farmers to use and the scarcity of veterinarians in farm animal practice.  相似文献

The objective of this study was to characterize porcine beta1- and beta2-adrenergic receptors (beta1-AR and beta2-AR) in heart, skeletal muscle, and adipose tissue by measuring the binding of a radioligand to cell membrane fragments. In skeletal muscle (LM), [3H]CGP12177 labeled a homogeneous population of beta2-AR as evidenced by the rank order of affinity of catecholamines [(-)isoproterenol > (-)epinephrine > (-)norepinephrine], a high affinity of the binding site for the beta2-AR-agonist clenbuterol (equilibrium dissociation constant, Kd = 16 nM), and a low affinity of the binding site for the beta1-AR-antagonist CGP20712A (Kd = 21 microM). The affinity of ICI118551, a ligand selective for beta2-AR in other species, was uncharacteristically low in porcine LM (Kd = 441 nM), but was consistent with a value reported for the cloned porcine beta2-AR. In heart ventricle, ligand binding revealed a predominant population of beta1-AR, judged by the rank order of affinity of catecholamines [(-)isoproterenol > (-)epinephrine > or = (-)norepinephrine] and high-affinity binding to CGP20712A (Kd = 40 nM). The Kd for ICI118551 (731 nM) was close to that observed at beta2-AR in LM, confirming that ICI118551 is not subtype-selective in the pig. Displacement studies using (-)propranolol, clenbuterol, and (-)isoproterenol revealed a second high-affinity binding site in the heart that was not a beta2-AR and could not be eliminated by guanosine 5'-triphosphate or guanylyli-midodiphosphate. In adipose tissue, an equal number of beta1- and beta2-AR was identified through the binding of clenbuterol and CGP20712A, whereas ICI118551 could not discriminate between these sites. In further experiments, we used 10 microM CGP20712A to eliminate beta1-AR binding and allow accurate Kd values to be determined at beta2-AR for nonselective ligands. Under these conditions, another binding site was observed that had a high affinity for (-)propranolol (Kd = 20 pM), which is inconsistent with beta3- or beta4-AR binding reported elsewhere. Our results indicate that porcine adipose tissue contains beta1-AR, beta2-AR, and an atypical binding site in the proportions 50, 34, and 16%, respectively, of the total binding sites labeled by [3H]CGP12177.  相似文献

Biopsy specimens were collected from skin or spleen from 8 dogs, 7 cats, and 1 cow with mast cell neoplasms. Following histopathologic grading, the neoplasms were examined by transmission electron microscopy with the intention of reviewing their fine structure and recording new findings. For 2 feline specimens, short-term cell cultures were established, and adherent cells were fixed in situ and examined with the electron microscope. In addition to the usual array of mast cell organelles, including Golgi apparatus, secretory granules, mitochondria, vesicles, tubules, microfilaments, and ribosomes, important findings included coarse interdigitation of cytoplasmic projections between adjacent mast cells; intracytoplasmic parallel stacks of 10 nm diameter filaments as well as parallel arrays of coarse, 120 to 150 nm diameter tubules; the appearance of coated vesicles and caveolae on cell surfaces; the appearance of endocytosed erythrocytes; and the formation of giant secretory granules.  相似文献

The ultrastructural features of lymphoproliferative disorders from 18 dogs are described and compared with those recorded in the veterinary literature. A spectrum of diseases was selected for study to demonstrate the structural heterogeneity of these disorders. The most extreme variations were noted in nuclear contour, where bizarre cerebriform-appearing nuclei were characteristic of neoplasms of T-cell lineage, or single deep nuclear clefts suggested B-cell lineage. The lesions surveyed included acute and chronic lymphocytic leukemias, plasma cell neoplasms, cutaneous and multicentric malignant lymphomas, and cutaneous lymphoproliferative disorders with histologic similarities to human mycosis fungoides.  相似文献

SUMMARY Lice were collected from 71 flocks detected as Infested at market Inspection in 1990 and 1991, from 16 flocks where resistance was suspected and from 31 flocks from Kangaroo Island. Susceptibility to cypermethrin was measured by a treated surface technique and survival of lice at 5 ppm or greater was taken as an indication of resistance. The prevalence of resistance was 34% in louse populations from the market inspection sample, 50% in flocks in which resistance was suspected and 68% in flocks from Kangaroo Island. Most resistance factors were in the range 1 to 20 although one louse population with a resistance factor of 91 was found on Kangaroo Island.  相似文献

Reasons for performing the study: L‐tryptophan is a common ingredient in equine calmative products, but its effectiveness has not been demonstrated in horses. Hypothesis: To determine whether a commercial dose of L‐tryptophan increases plasma tryptophan and alters behaviour in horses fed a roughage or concentrate meal. Methods: L‐tryptophan (6.3 g) or placebo (water) was administered per os in a cross‐over design, to 12 Thoroughbred horses (503 ± 12.1 kg bwt), just before a meal of lucerne hay or oats. Plasma tryptophan was measured by gas chromatography. Horse behaviour was observed in an empty enclosure, then in the presence of an unfamiliar person and a novel object. Results: Total plasma tryptophan increased 3‐fold in both studies, peaking 1.5‐2 h after dosing. After the peak, tryptophan remained high for several hours if the horses had been fed hay, but fell sharply if fed oats, consistent with the glycaemic responses to these meals. However, the ratio of tryptophan to 4 large neutral amino acids (phenylalanine, tyrosine, leucine and isoleucine) increased in the tryptophan‐treated horses to a similar extent and for a similar duration, with both diets. The presence of a stranger or novel object increased heart rate (P<0.05), but caused no behavioural effects that were altered by tryptophan, regardless of the diet. Conclusions: Plasma tryptophan increases when tryptophan is administered at a dose used in some commercial products, but this is not reflected by marked behavioural changes in the horse. Potential relevance: Further work is required to refine behavioural tests and identify an effective dose of L‐tryptophan in the horse.  相似文献

AIM: To assess the efficacy of toltrazuril against the Eimeria spp. affecting brown kiwi (Apteryx mantelli).

METHODS: Droppings were collected from three brown kiwi, aged <6 months old, at a captive rearing facility in the North Island of New Zealand, between 22 February and 20 April 2017, on 14 sampling dates. Only droppings (n=30) that were excreted between 03:00 and 07:00, as determined using video surveillance, were included for analysis, reflecting the peak time for shedding of coccidial oocysts for brown kiwi. Oocysts were quantified in each sample and Eimeria species identified on the basis of oocyst morphology. All samples were collected between 2 and 10 days after the birds had been treated with 25?mg/kg toltrazuril.

RESULTS: Eimeria spp. oocysts were identified in 28/30 individual samples and on 14/14 sampling dates. Oocyst counts varied from 0 to 328,080 oocysts per gram (opg), and at least one oocyst count >10,000 opg was measured on 12/14 sampling dates. Three species of Eimeria were observed, with Eimeria apteryxii and E. kiwii most commonly encountered, whereas only one sample contained E. paraurii.

CONCLUSIONS AND CLINICAL RELEVANCE: In the three birds monitored at this research site, there was a high abundance of E. apteryxii and E. kiwii oocysts in droppings despite recent administration of toltrazuril. These results suggest that the populations of Eimeria spp. affecting brown kiwi at this location appear to possess an ability to survive exposure to toltrazuril. Toltrazuril is widely used at captive rearing facilities to limit the effects of coccidiosis in juvenile kiwi. If a lack of efficacy is confirmed, it will be necessary to investigate alternative treatment regimens alongside broader environmental management strategies.  相似文献
