A new device for evaluating the continuity of taste was developed with the use of surface plasmon resonance (SPR). The model of lingual cells was constructed with liposomes immobilized onto an L1 sensor chip for SPR. Using this device, we classified food components into three categories according to the sensorgram pattern and residual ratio on lipid bilayer. Samples in group A strongly interacted with lipid bilayer, those in group B poorly interacted, and those in group C belong to neither group A nor group B. Sweet proteins and gymnemic acids that prolonged sweet perception were categorized in group A. Almost all the carbohydrates investigated and aspartame, of which the taste perception does not continue, belonged to group B. This device made it possible to detect the interaction with lipid bilayer and dissected the mechanism of taste continuity. 相似文献

Radical scavenging activities of extracts and constituents in Cornus capitata adventitious root cultures were evaluated by using 1,1-diphenyl-2-pycrylhydrazyl (DPPH) and superoxide anion radicals. Inhibitory activity against peroxidation of linoleic acid was assayed by using the thiobarbituric acid (TBA) method. Ethyl acetate and aqueous fractions were prepared from adventitious roots cultured in Murashige-Skoog liquid medium with 0.1 microM Cu(2+) (0.1CuMS) or 10 microM Cu(2+) (10CuMS). The highest scavenging activities on DPPH and superoxide anion radicals were observed in the ethyl acetate fraction from 0.1CuMS. In the inhibitory activity against linoleic acid oxidation, the ethyl acetate fraction from 10CuMS was highest among the fractions tested. The ethyl acetate fraction of adventitious roots cultured in 0.1CuMS contained mainly galloylglucoses (1,2,3,6-tetragalloylglucose and 1,2,3,4,6-pentagalloylglucose). The ethyl acetate fraction of adventitious roots cultured in 10CuMS contained mainly ellagic acid derivatives [3,3'-di-O-methylellagic acid 4-(5"-acetyl)-alpha-L-arabinofuranoside and stenophyllin H1]. Aqueous fractions prepared from both media contained iridoid glycosides (dihydrocornin and cornin). Tetra- and pentagalloylglucoses showed strong inhibitory activities (61.9 and 85.2%, respectively) against linoleic acid oxidation relative to those of butylated hydroxytoluene (BHT) (91.1%) or alpha-tocopherol (49.5%) at 50 microM concentration. Although both ellagic acid derivatives had weak activities (<50%) on DPPH and superoxide anion radical scavenging, 3,3'-di-O-methylellagic acid 4-(5"-acetyl)-alpha-L-arabinofuranoside was stronger (74.7%) than alpha-tocopherol (49.5%) in inhibiting linoleic acid oxidation at 50 microM concentration. Iron overload has been well recognized to cause oxidant-mediated cellular/tissue injury; however, little is known about the effects of iron overload on the blood coagulation system. We encountered an unexpected bleeding tendency in rats fed a high-iron diet in a set of studies using iron-modified diets. In this study, we investigated the mechanism of hemorrhagic diathesis induced by dietary iron overload in rats. Six-week-old F344/DuCrlCrlj male rats were fed a standard (containing 0.02% iron) or a high-iron diet (containing 1% iron) for 6 weeks and were then sampled for hematological, blood biochemical, coagulation, and pathological examinations. Serum and liver iron levels increased in rats fed the high-iron diet (Fe group) and serum transferrin was almost saturated with iron. However, serum transaminase levels did not increase. Moreover, plasma prothrombin time and activated partial thromboplastin time were significantly prolonged, regardless of the presence of hemorrhage. The activity of clotting factors II and VII (vitamin K-dependent coagulation factors) decreased significantly, whereas that of factor VIII was unaltered. Blood platelet levels were not influenced by dietary iron overload, suggesting that the bleeding tendency in iron-overloaded rats is caused by secondary hemostasis impairment. In addition, hemorrhage was observed in multiple organs in rats fed diets containing more than 0.8% iron. Our results suggest that iron overload can increase the susceptibility of coagulation abnormalities caused by latent vitamin K insufficiency. 相似文献

The effects of dietary medicinal herbs on growth and some non-specific immunity were investigated in juvenile red sea bream
Pagrus major. The fish (mean body weight 24.0±0.2g) were fed fishmeal diets supplemented with either Massa medicata (Mm), Crataegi fructus (Cf), Artemisia capillaries (Ac), Cnidium officinale (Co), or a mixture of all the herbs (HM), and a control diet without medicinal herbs, for 12 weeks. Survival, specific growth
rate, feed efficiency, condition factor and hemoglobin levels were higher in fish given herbal diets than fish given the control
diet without herbs. Significantly higher serum high density lipoprotein-cholesterol level and lysozyme activity were detected
in HM and Co diet groups, and alternative complement pathway activity was detected in the HM diet group. However, significantly
lower serum alanine aminotransferase and aspartate aminotransferase activities were obtained in all herbal diet groups compared
with the control diet group. Pathogen challenge test by intraperitoneal injection of Vibrio anguillarum indicated that highest survival was obtained in the HM diet group followed by Ac, Co, Cf, and Mm diet groups. The lowest
survival was obtained in the control group. These results reveal that medicinal herbs in diets enhance growth and some non-specific

Since 1985, frequent mass mortalities due to a bacterial infection have occurred in zoeal larvae of swimming crab Portunus trituberculatus reared at the Tamano Station of the Japan Sea-Farming Association. A bacterium was predominantly isolated from diseased larvae and was demonstrated by experimental infection to be the causative agent of the disease. Based on morphological, physiological, biochemical, serological and genetic characteristics of the pathogen, it was thought to be a new species and tentatively named Vibrio sp. Zoea. Challenge experiments with the bacterium in several aquatic animals indicate that Vibrio sp. Zoea appears to be a pathogen specific for crustaceans. 相似文献

ABSTRACTTemperatures and solar radiation during ripening critically affect grain appearance in rice (Oryza sativa L.). Climatic factors to induce chalky grains were analyzed under the experimental conditions of high-temperature and shading treatment and also under the ambient condition in a high-temperature-prone region of Japan. The frequency of white-back (WB) and basal-white (BW) grains correlated with temperature and solar radiation, whereas that of milky-white (MW) grains was not correlated, suggesting that complicated climatic factors are involved in the formation of MW grains. Further investigation was carried out to identify the parameters that distinguish perfect and MW grains grown in high-temperature versus those grown in low-solar-radiation conditions. As reported previously, the chalk phenotypes in the transverse section of the MW grains were quite different between environments: oval-shaped chalk for MW grains grown in low-solar-radiation condition and center chalk for MW grains grown in high-temperature condition. Grain hardness and amylopectin chain-length distribution did not explain the difference in MW grains between environments. MW grains subjected to high temperatures had a lower protein content without a consistent reduction in the single-grain weight, whereas those from the low-solar-radiation condition had a lower amylose content with a consistent reduction in the single-grain weight, when compared with perfect grains that developed in either environmental condition. Overall, our results suggest that MW grains are formed through different physiological mechanisms with altered starch and protein synthesis under high-temperature and low-solar-radiation conditions.Abbreviations: BW: basal-white; DAH: days after heading; DP: degree of polymerization; MW; milky-white WB: white-back 相似文献

Iron (Fe) is an important micronutrient for living organisms. Fe deficiency severely impairs plant growth and is a widespread human dietary problem, with particularly high numbers of affected children and females. Rice (Oryza sativa) is a source of energy for more than half of the world’s population. Thus, understanding the mechanisms of Fe uptake and translocation in rice is of utmost importance in the development of rice varieties that are tolerant to low Fe availability and with high seed levels of Fe. In recent years, the mechanisms underlying Fe transport and homeostasis have been revealed, providing opportunities to increase the Fe content of rice grain. As excess Fe is toxic to cells, plants have developed sophisticated mechanisms to control Fe flow, making it difficult to alter Fe transport. Thus, choosing appropriate chelators and Fe transporters driven by appropriate promoters seems to be the key in developing rice that is tolerant to low Fe availability and which accumulates high grain levels of Fe. Many recent studies have been aimed at increasing the Fe content of rice. Here, we summarize these efforts and review recent progress in understanding the mechanisms of Fe transport. 相似文献

Red sea bream Pagrus major (RSB) weighing 0.09 g were fed on test diets, composed of 65% fish meal supplemented with 0.49% Crataegi fructus (Cf) and a mixture of Massa medicata, Cf, Artemisia capillaries and Cnidium officinale (HM) or a control diet without herbs, in trial 1. After air exposure and anaesthesia treatment, higher stress tolerances were obtained in herbal groups than control group. Moreover, after challenge test with Vibrio anguillarum, mortalities of the herbal groups were lower than the control group. There was no significant difference in growth performance among the treatments. Feed efficiency (FE) of the test groups was 124–141%. In trial 2, RSB weighing 0.11 g were fed on diets composed of 35% fish meal and 30% enzyme treated fish meal (EFM) with the same herbal treatments as trial 1. The herbal groups revealed higher final body weight and specific growth rate (SGR) and lower daily feed intake than those of the control group. The FE of test groups was 172–203%. These results indicate that the dietary herbs supplementation acts not only as recovery enhancer for some rearing stresses but also as growth enhancer, which is accelerated with dietary EFM in the early juvenile stage of red sea bream. 相似文献

The skin fluke Neobenedenia girellae has become a serious problem in Japan since the 1990s. Present control methods focus on the removal of the attached parasite and these post-infection treatments are often labor intensive, time consuming, and/or stressful to fish. Chub mackerel Scomber japonicus are highly susceptible to N. girellae. However, because of their sensitive nature, bath treatments may cause mortality. In this study, we investigated the efficacy of cage shading to reduce skin fluke infection and the frequency of conventional post-infection treatments. Juvenile mackerel were reared in cages with or without shade for 3 months and their skin fluke infections were monitored. We performed either freshwater baths or oral administration of praziquantel if fluke intensity exceeded the given criteria. In unshaded cages, 3 total bath treatments or 6 total drug treatments were conducted. In contrast, no treatment was required for the shaded cage. The overall fluke intensity in the shaded cage was less than half that of the unshaded cages, despite the lack of treatments. This study demonstrated for the first time the practical use of shading in fish farms to reduce skin fluke infection. 相似文献
