福建沿海牡蛎养殖群体的多重种类特异性PCR分析和形态参数比较 总被引:1,自引:0,他引:1
应用多重种类特异性PCR方法对福建沿海养殖牡蛎的6个自然苗群体(福鼎FD、连江LJZ、福清FQ、莆田门、石狮SSZ、厦门XM)和4个人工苗群体(宁德ND、连江LJR、石狮SSR、诏安ZA)进行种类鉴定,并应用方差分析和Tukey多重比较对10个养殖群体的4个壳形态参数进行比较分析。每个群体随机选取30个个体,共分析10个养殖群体300个个体。多重种类特异性PCR分析结果显示,自然苗群体中共发现熊本牡蛎17个,其余为葡萄牙牡蛎;人工苗群体均为葡萄牙牡蛎。福建沿海养殖牡蛎种类为葡萄牙牡蛎和熊本牡蛎,其中葡萄牙牡蛎为优势种类。应用方差分析和Tukey多重比较分析结果表明,葡萄牙牡蛎的壳形态参数明显大于熊本牡蛎;FD和SSZ群体间差异不显著(P〉0.05),SSR和xM群体间差异不显著(P〉0.05);FD与FQ、ND和LJR群体间差异显著(P〈0.05)。 相似文献

研究了杏SSR-PCR反应体系的主要成分对扩增结果的影响,并进行了体系验证.优化后的反应体系为:总体积20 μL,1×buffer、2.0 mM/L Mg2+、0.25 mM/L dNTP、0.20 μM/L Primer、60 ng/20μL模板DNA和0.05 U/μL Taq酶.利用32个杏品种验证此反应体系,6%的变性聚丙稀酰胺凝胶电泳检测结果显示,扩增产物在184~267 bp之间,不同品种间DNA谱带具有多态性, 且反应体系的稳定性和可重复性好. 相似文献

To investigate the relationship between stable carbon isotope discrimination (Δ) of different organs and water use efficiency (WUE) under different water deficit levels, severe, moderate and low water deficit levels were treated at bud burst to leafing, flowering to fruit set, fruit growth and fruit maturation stages of field grown pear-jujube tree, and leaf stable carbon isotope discrimination (ΔL) at different growth stages and fruit stable carbon isotope discrimination (ΔF) at fruit maturation stage were measured. The results indicated that water deficit had significant effect on ΔL at different growth stages and ΔF at fruit maturation stage. As compared with full irrigation, the average ΔL at different growth stages and ΔF at fruit maturation stage were decreased by 1.23% and 2.67% for different water deficit levels, respectively. ΔL and ΔF among different water deficit treatments had significant difference at the same growth stage (P < 0.05). Under different water deficit conditions, significant relationships between the ΔL and WUEi (photosynthesis rate/transpiration rate, Pn/Tr), WUEn (photosynthesis rate/stomatal conductance of CO2, Pn/gs), WUEy (yield/crop water consumption, Y/ETc) and yield, or between the ΔF and WUEy and yield were found, respectively. There were significantly negative correlations of ΔL with WUEi, WUEn, WUEy and yield (P < 0.01) at the fruit maturation stage, or ΔL with WUEi and WUEn (P < 0.01) over whole growth stage, respectively. ΔF was negatively correlated with WUEy, WUEn and yield (P < 0.05), but positively correlated with ETc (P < 0.01) over the whole growth stage. Thus ΔL or ΔF can compare WUEn and WUEy, so the stable carbon isotope discrimination method can be applied to evaluate the water use efficiency of pear-jujube tree under the regulated deficit irrigation. 相似文献

The quality and safety of agricultural products is the technique focus of the strategy of rural vitaliza⁃tion. Carbamate pesticide residues are seriously threatening the safety of agricultural product and human life and health. Nowadays, the common detection methods are instrument-based for carbamate pesticide residue, which dis⁃ satisfy the request of an on-site rapid, sensitive and simultaneous detection for multiple carbamate pesticide resi⁃ dues. Based on the home-made monoclonal antibodies against carbofuran and carbaryl respectivity, a microarray for rapid and simultaneous detection of multiple carbamate pesticide residues was developed within 30 min. Under the optimized conditions, the limits of detection(LODs)were found to be 0.11 and 0.09 ng/g, while the linear ranges were 0.33-3000 and 0.18-1500 ng/g for carbofuran and carbaryl respectively. By using spiked peanut samples, it recorded recoveries of 88.4%-98.3%, 90.1%-118.8% for carbofuran and carbaryl respectively. The RSD of intraarray were found at 9.0%, 3.5%, while the RSD of inter-array were recorded at 12.3%, 16.3% respectively. The de⁃
tection results of the microarray were highly consistent with that of HPLC-MS/MS. This proposed microarray provid⁃ ed an on-site high-sensitivity, simultaneous and rapid detection of carbofuran and carbaryl, and could be widely used in the fields of agricultural product quality safety, food safety and environmental monitoring. 相似文献
