Papaya is a climacteric fruit that has high amounts of benzylglucosinolates (BG) and benzylisothiocyanates (BITC), but information regarding levels of BG or BITC during fruit development and ripening is limited. Because BG and BITC are compounds of importance from both a nutritional and a crop yield standpoint, the aim of this work was to access data on the distribution and changes of BG and BITC levels during fruit development and ripening. BG and BITC levels were quantified in peel, pulp, and seeds of papaya fruit. Volatile BITC was also verified in the internal cavity of the fruit during ripening. The influence of the ethylene in BG and BITC levels and mirosinase activity was tested by exposing mature green fruits to ethylene and 1-methylcyclopropene (1-MCP). The highest BG levels were detected in seeds, followed by the peel and pulp being decreased in all tissues during fruit development. Similarly, the levels of BITC were much higher in the seeds than the peel and pulp. The levels of BG for control and ethylene-treated fruit were very similar, increasing in the pulp and peel during late ripening but not changing significantly in seeds. On the other hand, fruit exposed to 1-MCP showed a decrease in BG amount in the pulp and accumulation in seed. The treatments did not result in clear differences regarding the amount of BITC in the pulp and peel of the fruit. According to the results, ethylene does not have a clear effect on BITC accumulation in ripening papaya fruit. The fact that BG levels in the pulp did not decrease during ripening, regardless of the treatment employed, and that papaya is consumed mainly as fresh fruit, speaks in favor of this fruit as a good dietary source for glucosinolate and isothiocyanates. 相似文献

Intensive culture of the hybrid surubim (Pseudoplatystoma corruscans × Pseudoplatystoma fasciatum) in Brazil is responsible for the occurrence of diseases and consequent economic losses. However, the causative agents are not well known. The objective of this study was to isolate and to characterize the pathogenic agent responsible for mortalities in cultured surubim and to demonstrate its virulence. Ten fish from a fish farm located in the Mato Grosso do Sul State (Brazil) were collected and 14 haemolytic bacteria characterized as Aeromonas hydrophila were isolated from the kidneys (eight) and brain (six). As an experimental challenge, fish weighing 98.1±23.6 g were injected with 1 mL of saline solution and 2 × 102, 2 × 104, 2 × 106 and 2 × 108 CFU A. hydrophila mL?1. Fish infected with 2 × 108 CFU showed increased external and internal symptoms and mortality of 50±12.5% after 96 h. Increased A. hydrophila concentration was responsible for a decrease in haematocrit percentage and erythrocyte number, lymphocytes and eosinophils, as well as an increase in monocytes, neutrophils, serum agglutination titre and serum antimicrobial activity. It was concluded that A. hydrophila was responsible for characteristic symptoms of bacterial haemorrhagic septicaemia as well as important haematological and immunological alterations, which led to surubim mortality. 相似文献

This paper aimed to evaluate the efficiency of resistance elicitors in the management of grapevine downy mildew (Plasmopara viticola), identify the action of the elicitors on host metabolism, and determine their economic viability. The experiments were performed in a commercial vineyard variety ‘Isabel’ (Vitis labrusca) at Vale do Sirijí [Natuba, Paraiba State in Brazil, in the period of September 2009 to January 2010 (first season) and February to June 2010 (second season)]. The statistical design of randomized blocks consisted of eight treatments (untreated control, fungicide (pyraclostrobin+metiram), potassium phosphite, Agro-Mos?, Fungicide+potassium phosphite, Fungicide+Agro-Mos?, potassium phosphite+AgroMós? and Fungicide+potassium phosphite+Agro-Mos?) with four replications, with the experimental unit consisting of 45 leaves. Applications were made every 7?days, starting 20?days after pruning (DAP) with a total of 12 sprays. The evaluations were carried out biweekly, analyzing the following variables: incubation period, disease incidence and severity, area under the disease progress curve, and efficiency of control. The enzymatic determination was performed using pulp extracts from three fruits harvested at 45, 60, 90 and 120 DAP for each treatment. The resistance elicitors were able to reduce the disease incidence under different climatic conditions, indicating their viability as an alternative for the management of Plasmopara viticola. 相似文献

A lectin was purified from the seaweed Gracilaria cornea by hydrophobic interaction chromatography on phenyl-Sepharose CL-4B followed by affinity chromatography on immobilized mucin. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of G. cornea lectin (GCL) revealed a single protein band of approximately 60 kDa, whereas by gel filtration on Sephadex G-100 its native molecular mass was 66 kDa. GCL exhibited a single isoeletric point of 4.3 and a 52.5% content of neutral sugars. Furthermore, the EDTA-treated lectin did not show any significant decrease in its ability to agglutinate trypsin-treated chicken erythrocytes. These data suggest that GCL is an acidic, monomeric glycoprotein that probably does not require divalent metal ions for its hemagglutinating activity. GCL hemagglutinating activity was not inhibited by any of the mono-, di-, and trisaccharides tested but was by the complex glycoproteins fetuin and porcine stomach mucin. Exposure of engorged females of the cattle tick (Boophilus microplus) to 0.1 mg mL(-1) GCL significantly (P < 0.05) reduced the female weight after the oviposition period, the egg mass weight, the hatching period, and the mean larvae survival time. 相似文献

Unripe bananas have a high content of starch (almost 20%) that is metabolized during fruit ripening with a concomitant synthesis of soluble sugars. Since starch granules are composed of amylose and amylopectin, several enzymes have to be involved in its mobilization during banana ripening, with a necessary participation of one starch-debranching enzyme (DBE) to hydrolyze the alpha-1,6-branches of amylopectin. Banana DBE seems to be an isoamylase-type enzyme, as indicated by substrate specificity and the cloning of a 1575 bp cDNA, similar to the isoamylase sequences from potato, Arabdopsis, and maize. The assays for DBE indicated only minor changes in activity during ripening, and the results of the northern and western blots with antiserum against the recombinant banana isoamylase were in agreement with the steady-state level of activity, since no significant changes in gene expression were observed. The high activity on beta-limit dextrin and the similarity to the potato isoform 3 suggest that during banana ripening the hydrolysis of alpha-1,6-linkage of amylopectin results from the activity of a pre-existing isoamylase-type debranching enzyme in coordination with other amylolitic enzymes. An analytical procedure for the separation and quantification of 17 short-chain, medium-chain, and long-chain acids in cacha?as and various spirits has been developed involving C18 solid phase extraction, derivatization with 9-anthryldiazomethane, and reverse phase HPLC using fluorescence detection. The limit of detection was between 5 and 15 fmol, whereas the recovery of nonanoic acid as internal standard was >95%. In theory, habitat fragmentation alters plant community dynamics by influencing both local (within patch) and regional (among patch) processes. However, the lengthy generation times of plant taxa relative to the short duration of most experiments has precluded studies from assessing the impact of fragmentation at both local and regional scales. Due to their accelerated life cycles, high rates of local extinction, and naturally patchy substrates, epiphyllous bryophyte assemblages are an appropriate plant guild for empirically testing metacommunity-based predictions associated with habitat fragmentation. By examining the local abundance and regional distribution patterns of 67 epiphyllous (leaf-inhabiting) bryophyte species in an experimentally fragmented landscape in Amazonia, we demonstrate that changes in local abundance wrought by habitat fragmentation are best explained by fragment size rather than proximity to forest edge. Furthermore, evidence of a simultaneous inter-specific decline in epiphyll local abundance and regional distribution in small (1- and 10-ha) forest fragments corroborate with metapopulation-based predictions highlighting the importance of immigration in buffering from patch extinction risk (i.e., the rescue effect). Collectively, these results provide indirect evidence that dispersal limitation, rather than compromised habitat quality attributable to edge effects, likely account for species loss from small tropical forest fragments. Whether dispersal limitation is due to increased insularity from regional sources for epiphyll recolonization or rather to lowered within-fragment dispersal potential is unknown; nonetheless, the long-term persistence of these microscopic plant metacommunities depends on the preservation of rain forest reserves of at least 100-ha in size. 相似文献
