Models of biogenic carbon (BC) flux assume that short herbivorous food chains lead to high export, whereas complex microbial or omnivorous food webs lead to recycling and low export, and that export of BC from the euphotic zone equals new production (NP). In the Gulf of St. Lawrence, particulate organic carbon fluxes were similar during the spring phytoplankton bloom, when herbivory dominated, and during nonbloom conditions, when microbial and omnivorous food webs dominated. In contrast, NP was 1.2 to 161 times greater during the bloom than after it. Thus, neither food web structure nor NP can predict the magnitude or patterns of BC export, particularly on time scales over which the ocean is in nonequilibrium conditions.  相似文献

X-ray lasers (XRLs) have experimental average gains that are significantly less than calculated values and a persistently low level of spatial coherence. An XRL has been used both as an injected signal to a short XRL amplifier and as an interferometer beam to measure two-dimensional local gain and density profiles of the XRL plasma with a resolution near 1 micrometer. The measured local gain is in agreement with atomic models but is unexpectedly spatially inhomogeneous. This inhomogeneity is responsible for the low level of spatial coherence observed and helps explain the disparity between observed and simulated gains.  相似文献

In the non‐breeding season, LH release is reduced via dopaminergic systems in the ram. On the other hand, our previous studies demonstrated an opioidergic inhibition of LH release in stallions outside the breeding season. Thus, in the present study we investigated the dopaminergic regulation of LH and prolactin secretion in stallions, considering interactions between dopamine and opioids. To achieve this, stallions (n=8) were treated with the dopamine antagonist sulpiride (0.6 mg/kg), the opioid antagonist naloxone (0.5 mg/kg), sulpiride plus naloxone or saline in December, March and June. Two hours after the respective treatments, they received a GnRH agonist. Sulpiride induced a significant prolactin release which was most pronounced in December, indicating seasonal variations in the inhibition of prolactin secretion by dopaminergic systems. Prolactin concentrations were not changed by naloxone. Neither during nor outside the breeding season, a dopaminergic regulation of LH release could be demonstrated. In contrast, naloxone caused a significant (p < 0.05) LH release, confirming an opioidergic inhibition of LH release. In conclusion, opioidergic regulation of LH and dopaminergic inhibition of prolactin secretion undergo seasonal changes. Neither during nor outside the breeding season, dopaminergic effects on LH release exist in the stallion.  相似文献

This paper presents quantitative results of canine pregnancy diagnoses performed by abdominal palpation and/or linear-array real-time echography. Each animal was first palpated by an experienced clinician and then another investigator performed ultrasound scanning using a 3 MHz transducer. In other dogs echography was undertaken only when the result of abdominal palpation was negative or doubtful or when the owner requested ultrasound scanning. Using all tests performed between days 20 and 49 of pregnancy (day 0 = day of first mating or insemination), the results of abdominal palpation (n = 116) versus real-time scanning (n = 135) were: sensitivity: 89.0% v 92.9%; specificity: 93% v 96.0%; positive predictive value: 95.6% v 97.5%; negative predictive value: 83.3% v 88.9%. Results of ultrasound tests performed in 97 other dogs were: sensitivity: 92.4%; specificity: 97.7%; positive predictive value: 98.0% and negative predictive value: 91.4%. Almost identical figures were found when only the tests performed between days 25 and 35 of pregnancy were included. It was concluded that real-time ultrasound scanning is an accurate method for discriminating between pregnant and non-pregnant bitches; sensitivity and specificity of the abdominal palpation technique executed by an experienced clinician approach that of ultrasound scanning; small litter size and lack of knowledge of conception date are the main sources of false negative diagnoses.  相似文献

The outcome of E. coli mastitis in cows ranges from mild to severe in individual animals. This study explored the hypothesis that milk from individual cows differs in its growth medium properties for E. coli, and whether possible variation could be related to specific milk constituents. To mimic the early phase of intramammary E. coli infection, a low inoculum size and a short incubation period were used. Cell-reduced, cell- and fat-free (skim) and cell- and fat-free and protein-reduced (whey) fractions were prepared from whole milk samples (n=18). Ten ml of whole milk, milk fractions and brain heart infusion broth (BHI) were inoculated with approximately 100cfu E. coli. After 6h of incubation, bacterial counts were assessed by dilution plating in triplicate. Bacterial counts in whole milk differed up to a 100-fold between cows, which was not associated with SCC. Bacterial counts were significantly higher in whey fractions than in whole milk, cell-reduced and skim fractions and variation in whey was smaller, indicating that the acid-precipitable protein fraction contains the milk constituents of major relevance for inhibition of and variation in bacterial growth. The presence of fat and cells added to bacterial growth inhibition to a lesser extent. In conclusion, in vitro growth of E. coli in milk differs substantially between individual cows within an incubation period comparable with the early phase of intramammary infection. This suggests that the growth medium properties of milk could be of importance in the pathogenesis of E. coli mastitis and subsequent outcome of disease.  相似文献

The corpus luteum (CL) of the pig lacks luteolytic sensitivity (LS) to prostaglandin (PG) F‐2α until after day 12 of the oestrous cycle, but the mechanisms underlying this phenomenon are poorly understood. As luteolysis involves apoptosis, we hypothesized that critical apoptotic proteins may be deficient in CLs that lack LS. The specific aim of these studies was to examine mRNA expression and protein levels of apoptosis genes/proteins (BAX/Bax, BCLX/Bcl‐x, CASP3/Caspase‐3, CASP8/Caspase‐8, NFΚB1/NFκB, TP53/p53) in porcine CLs collected at different stages of the oestrous cycle. CLs were collected surgically, mRNA and protein extracted, and expression/levels analyzed by semi‐quantitative (SQ) PCR and Western blots, respectively. At the mRNA expression level, only BAX (maximal on day 4) and TP53 (maximal on day 7) showed significant variations during the oestrous cycle. At the protein level, only Bcl‐x and Caspase‐3 showed significant changes during the cycle; Bcl‐x decreased on day 13 and Caspase‐3 increased on day 13. It is concluded that apoptosis‐associated proteins (i.e. Bcl‐x and Caspase 3) may play a critical role in luteolytic sensitivity in the pig.  相似文献

Pregnancy diagnosis was carried out in sheep by means of transabdominal linear-array real-time ultrasound scanning. Animals were restrained standing, and the transducer was placed on the hairless area of the ventral abdominal wall just in front of the udder. Of a total of 818 tests, 724 were performed between days 29 and 89 of pregnancy, 598 animals subsequently lambed and 126 were non-lambing animals. Only 8 of these tests were wrong: 3 false positive and 5 false negative diagnoses. Sensitivity, specificity, positive- and negative predictive values for these tests were 99.2%, 97.6%, 99.5%, and 96% respectively. There was evidence to indicate that the three false positive tests were caused by foetal mortality or unobserved abortions that took place after testing. Only 2 of the 5 false negative tests were carried out after day 39 of gestation. Counting of foetal numbers (1, 2 or 3) was performed in only some animals (n = 210) between days 45 and 77 of gestation. Three groups of animals (A: 89 ewes; B: 27 PMSG-treated ewes; C: 94 ewes) were analyzed separately. Overall accuracy of all predictions was 83.1%, 37.0% and 78.7% for the 3 groups respectively. Animals in group B produced only 3 or more lambs. This study investigated the possible effects of milk production level on the host resistance of dairy cows. High (n = 18) and low (n = 18) producing cows on a research farm, which respectively produced 11 443 and 7 727 kg milk in their previous lactation, were compared. To enhance the possible differences in host resistance between high and low producing cows, the animals in both groups were metabolically stressed by overfeeding during the dry period or were fed according to requirements, resulting in four groups of nine cows. The metabolic status was monitored from two weeks pre-partum until 2.5-4.5 weeks post-partum. Host resistance was assessed by measuring the severity of experimentally induced Escherichia coli mastitis. Pre-partum blood glucose levels tended to be higher in overfed cows than in cows fed according to requirements. The post-partum energy balance was significantly more negative in high producing cows than in low producers, and tended to be more negative in overfed cows compared to cows fed according to the requirements. Post-partum plasma glucose, NEFA, beta-OH-butyrate and urea concentrations were similar in the four groups. Plasma glucose concentrations were significantly lower and liver triacylglycerol concentrations were significantly higher in third than in second parity cows. Host resistance was not affected by the production level or feeding regimen. There were no significant correlations between the metabolic status and the severity of experimental E. coli mastitis, except for the relatively more severe mastitis in the cows with beta-OH-butyrate concentrations above 1.4 mmol/L. In conclusion, milk production level did not affect host resistance in dairy cows, as measured by the severity of experimental E. coli mastitis. Even in a situation where cows were metabolically stressed by overfeeding, high producers were as able as low producers to cope with the demands of milk production, without consequences for host resistance.  相似文献

Histophilus ovis was isolated from 29 sheep in 20 flocks and 2 artificial insemination (AI) centres in southern New South Wales from 1984 to 1990. The clinical and pathological findings were consistent with previous reports and included polyarthritis (7 flocks), epididymo-orchitis (5), meningoencephalitis (3), pneumonia (3), septicaemia (2), mastitis (1) and metritis (1). Six sheep had meningoencephalitis, a syndrome not previously associated with H ovis infection in sheep, which was similar pathologically to thromboembolic meningoencephalitis in cattle, caused by the related organism, Haemophilus somnus. H ovis was isolated from the semen of 12-month-old rams in a flock that had polyarthritis due to H ovis, in 4-month-old ram lambs and from the uterus of a ewe in a flock that had sporadic cases of H ovis septicaemia.  相似文献
