Selective sparing of a class of striatal neurons in Huntington's disease   总被引:29,自引:0,他引:29  
A distinct subpopulation of striatal aspiny neurons, containing the enzyme nicotinamide adenine dinucleotide phosphate diaphorase, is preserved in the caudate nucleus in Huntington's disease. Biochemical assays confirmed a significant increase in the activity of this enzyme in both the caudate nucleus and putamen in postmortem brain tissue from patients with this disease. The resistance of these neurons suggests that the gene defect in Huntington's disease may be modifiable by the local biochemical environment. This finding may provide insight into the nature of the genetically programmed cell death that is a characteristic of the disease.  相似文献

Rett syndrome is an autism spectrum disorder caused by mosaic expression of mutant copies of the X-linked MECP2 gene in neurons. However, neurons do not die, which suggests that this is not a neurodegenerative disorder. An important question for future therapeutic approaches to this and related disorders concerns phenotypic reversibility. Can viable but defective neurons be repaired, or is the damage done during development without normal MeCP2 irrevocable? Using a mouse model, we demonstrate robust phenotypic reversal, as activation of MeCP2 expression leads to striking loss of advanced neurological symptoms in both immature and mature adult animals.  相似文献

The genome of the invertebrate chordate Ciona intestinalis was found to be a stable mosaic of methylated and nonmethylated domains. Multiple copies of an apparently active long terminal repeat retrotransposon and a long interspersed element are nonmethylated and a large fraction of abundant short interspersed elements are also methylation free. Genes, by contrast, are predominantly methylated. These data are incompatible with the genome defense model, which proposes that DNA methylation in animals is primarily targeted to endogenous transposable elements. Cytosine methylation in this urochordate may be preferentially directed to genes.  相似文献

Asteroid 21 Lutetia was approached by the Rosetta spacecraft on 10 July 2010. The additional Doppler shift of the spacecraft radio signals imposed by 21 Lutetia's gravitational perturbation on the flyby trajectory were used to determine the mass of the asteroid. Calibrating and correcting for all Doppler contributions not associated with Lutetia, a least-squares fit to the residual frequency observations from 4 hours before to 6 hours after closest approach yields a mass of (1.700 ± 0.017) × 10(18) kilograms. Using the volume model of Lutetia determined by the Rosetta Optical, Spectroscopic, and Infrared Remote Imaging System (OSIRIS) camera, the bulk density, an important parameter for clues to its composition and interior, is (3.4 ± 0.3) × 10(3) kilograms per cubic meter.  相似文献

The preatmospheric mass of the Tagish Lake meteoroid was about 200,000 kilograms. Its calculated orbit indicates affinity to the Apollo asteroids with a semimajor axis in the middle of the asteroid belt, consistent with a linkage to low-albedo C, D, and P type asteroids. The mineralogy, oxygen isotope, and bulk chemical composition of recovered samples of the Tagish Lake meteorite are intermediate between CM and CI meteorites. These data suggest that the Tagish Lake meteorite may be one of the most primitive solar system materials yet studied.  相似文献

Plants often impact the rate of native soil organic matter turnover through root interactions with soil organisms; however the role of root-microbial interactions in mediation of the “priming effect” is not well understood. We examined the effects of living plant roots and N fertilization on belowground C dynamics in a California annual grassland soil (Haploxeralf) during a two-year greenhouse study. The fate of 13C-labeled belowground C (roots and organic matter) was followed under planted (Avena barbata) and unplanted conditions, and with and without supplemental N (20 kg N ha−1 season−1) over two periods of plant growth, each followed by a dry, fallow period of 120 d. Turnover of belowground 13C SOM was followed using 13C-phospholipid fatty acid (PLFA) biomarkers. Living roots increased the turnover and loss of belowground 13C compared with unplanted soils. Planted soils had 20% less belowground 13C present than in unplanted soils after 2 cycles of planting and fallow. After 2 treatment cycles, unlabeled soil C was 4.8% higher in planted soils than unplanted. The addition of N to soils decreased the turnover of enriched belowground 13C during the first treatment season in both planted and unplanted soils, however no effect of N was observed thereafter. Our findings suggest that A. barbata may increase soil C levels over time because root and exudate C inputs are significant, but that increase will be moderated by an overall faster C mineralization rate of belowground C. N addition may slow soil C losses; however, the effect was minor and transient in this system. The labeled root-derived 13C was initially recovered in gram negative (highest enrichment), gram positive, and fungal biomarkers. With successive growing seasons, the labeled C in the gram negative and fungal markers declined, while gram positive markers continued to accumulate labeled belowground C. The rhizosphere of A. barbata shifted the microbial community composition, resulting in greater abundances of gram negative markers and lower abundances of gram positive, actinobacteria and cyclopropyl PLFA markers compared to unplanted soil. However, the longer-term utilization of labeled belowground C by gram positive bacteria was enhanced in the rhizosphere microbial community compared with unplanted soils. We suggest that the activities of gram positive bacteria may be major controllers of multi-year rhizosphere-related priming of SOM decomposition.  相似文献

The water retention characteristic provides the traditional data set for the derivation of a soil's pore‐size distribution. However, the technique employed to achieve this requires that assumptions be made about the way pores interconnect. We explore an alternative approach based on stray field nuclear magnetic resonance (STRAFI‐NMR) to probe the water‐filled pores of both saturated and unsaturated soils, which does not require information relating to pore connectivity. We report the relative size distributions of water‐occupied pores in saturated and unsaturated samples of two sets of glass beads of known particle size, two sands, and three soils (a silty loam, a sandy loam and a loamy sand), using measurements of the NMR T1 proton relaxation time of water. The T1 values are linearly related to pore size and consequently measured T1 distributions provide a measure of the pore‐size distribution. For both the sands and the glass beads at saturation the T1 distributions are unimodal, and the samples with small particle sizes show a shift to small T1 values indicating smaller voids relative to the samples with larger particles. Different matric potentials were used to reveal how the water‐occupied pore‐size distribution changes during drainage. These changes are inconsistent with, and demonstrate the inadequacies of, the commonly employed parallel‐capillary tube model of a soil pore space. We find that not all pores of the same size drain at the same matric potential. Further, we observe that the T1 distribution is shifted to smaller values beyond the distribution at saturation. This shift is explained by a change in the weighted average of the relaxation rates as the proportion of water in the centre of water‐filled pores decreases. This is evidence for the presence of pendular structures resulting from incomplete drainage of pores. For the soils the results are similar except that at saturation the T1 distributions are bimodal or asymmetrical, indicative of inter‐aggregate and intra‐aggregate pore spaces. We conclude that the NMR method provides a characterization of the water‐filled pore space which complements that derived from the water retention characteristic and which can provide insight into the way pore connectivity impacts on drainage.  相似文献

Fifty-five isolates of Edwardsiella ictaluri were examined for the presence of plasmid DNA by a rapid alkaline extraction procedure. All 49 isolates from channel catfish and a single isolate from Bengal danio carried 2 plasmids with molecular masses of approximately 3.2 and 3.7 megadaltons (Mdal). Five E ictaluri isolates from other fish contained 1 to 3 plasmids, which had molecular masses ranging from 2.5 to 45 Mdal. The 2 plasmids (3.2 and 3.7 Mdal) from the type strain of E ictaluri (ATCC 33202) were ligated into pUC19 cloning vectors, and restriction endonuclease maps of each insert were prepared.  相似文献
