In this study, infrared thermography (IRT) was assessed as a means of detecting foot-and-mouth disease virus (FMDV)-infected cattle before and after the development of clinical signs. Preliminary IRT imaging demonstrated that foot temperatures increased in FMDV-infected animals. The maximum foot temperatures of healthy (n = 53), directly inoculated (DI) (n = 12), contact (CT) (n = 6), and vaccine trial (VT) (n = 21) cattle were measured over the course of FMD infection. A cut-off value was established at 34.4 °C (sensitivity = 61.1%, specificity = 87.7%) with the aim of detecting FMDV-infected animals both before and after clinical signs were observed. Seven of 12 (58%) DI and 3/6 (50%) CT animals showed maximum foot temperatures exceeding the 34.4 °C cut-off before the development of foot vesicles. In contrast, only 5/21 (24%) VT animals displayed pre-clinical foot temperatures above this cut-off possibly indicating partial vaccine protection of this group. These results show IRT as a promising screening technology to quickly identify potentially infected animals for confirmatory diagnostic testing during FMD outbreaks. Further evaluation of this technology is needed to determine the value of IRT in detecting animals with mild clinical signs or sub-clinical infections.  相似文献

The 2013 EU ban of three neonicotinoids used in seed coating of pollinator attractive crops was put in place because of concern about declining wild pollinator populations and numbers of honeybee colonies. It was also concluded that there is an urgent need for good field data to fill knowledge gaps. In the meantime such data have been generated. Based on recent literature we question the existence of recent pollinator declines and their possible link with the use of neonicotinoids. Because of temporal non‐coincidence we conclude that declines of wild pollinators and of honeybees are not likely caused by neonicotinoids. Even if bee decline does occur and if there is a causal relationship with the use of neonicotinoids, we argue that it is not possible on such short term to evaluate the effects of the 2013 ban. In order to supply future debate with realistic (field) data and to discourage extrapolating the effects of studies using overdoses that are not of environmental relevance, we propose – in addition to field studies performed by the chemical industry – to use the ‘semi‐field worst case’ treated artificial diet studies approach to free flying colonies in the field. This kind of study may provide realistic estimates for risk and be useful to study realistic interactions with non‐pesticide stressors. © 2017 The Authors. The objective of this study was to define the patterns of organogenesis and foetal haemodynamics during the normal gestation of healthy agoutis (Dasyprocta prymnolopha) kept in captivity. Thirty pregnant agoutis that ranged in size from small to medium and weighed between 2.5 and 3 kg underwent B‐mode and Doppler ultrasonography for the biometric evaluation of the foetal organs. The foetal aortic blood flow proved to be predominantly systolic, and the measured flow velocity was 78.89 ± 2.95 cm/s, with a maximum pressure gradient of 2.12 ± 0.27 mmHg. The liver was characterized by its large volume, occupying the entire cranial aspect of the abdominal cavity, and it was associated cranially with the diaphragm and caudally with the stomach. The flow velocity in the portal vein was estimated to equal 12.17 ± 2.37 cm/s, with a resistivity index of 0.82 ± 0.05. The gallbladder was centrally located and protruded cranially towards the diaphragm. The spleen was visualized as an elongated structure with tapered cranial and caudal extremities, and the foetal kidneys were visualized bilaterally in the retroperitoneal region, with the right kidney positioned slightly more cranially than the left. The morphological characterization and hemodynamic analysis of the foetal organs of black‐rumped agoutis via B‐mode and Doppler ultrasonography allow determination of the vascular network and of reference values for the blood flow required for perfusing the anatomical elements essential for maintaining the viability of foetuses at different gestational ages.  相似文献

Single layer centrifugation (SLC) has been shown to select the most robust spermatozoa from the ejaculate in several species. Here the effects of SLC prior to freezing on various parameters of frozen‐thawed bovine sperm quality are reported. Semen from 8 bulls was layered on top of a species‐specific colloid, Bovicoll. After centrifugation for 20 min at 300 g, the resulting sperm pellet was resuspended in OPTIXcell® (IMV Technologies, l′Aigle, France); the SLC‐selected sperm samples and uncentrifuged controls were frozen. On thawing, all sperm samples were analysed for membrane integrity, production of reactive oxygen species, mitochondrial membrane potential (MMP) and chromatin integrity. The SLC‐treated samples had a higher percentage of live, superoxide‐positive spermatozoa than uncentrifuged samples (27.9 ± 5.1% versus 21.7 ± 6.7%; p = .03). They had a higher proportion of spermatozoa with high mitochondrial membrane potential than uncentrifuged samples (55.9 ± 8.2% versus 40.5 ± 15.1%; p = .03) and also a lower proportion of spermatozoa with low mitochondrial membrane potential than non‐treated samples (42.0 ± 8.5% versus 55.9 ± 14.4%; p = .04). No significant effects of treatment were found for membrane integrity or chromatin integrity. The effect of bull was significant on the proportions of dead, superoxide‐positive spermatozoa and live, hydrogen peroxide‐negative spermatozoa, as well as on membrane integrity, but it was not significant for mitochondrial membrane potential or chromatin integrity. These results suggest that SLC selects the most metabolically active bull spermatozoa from the rest of the population in normal ejaculates; the pattern of reactive oxygen species production may be different in SLC‐selected spermatozoa compared to unselected samples.  相似文献

In plant breeding, correlations between the statistics of stability and adaptability of popcorn cultivars are not yet well understood. Therefore, the objectives of the present experiment was to investigate the correlations between sdi2 \sigma_{\rm di}^{2} and bi \beta_{\rm i} from Eberhart and Russell, ωi from Wricke, \textS\texti(1) {\text{S}}_{\text{i}}^{(1)} , \textS\texti(2) {\text{S}}_{\text{i}}^{(2)} and \textS\texti(3) {\text{S}}_{\text{i}}^{(3)} from Huehn, Pi from Lin and Binns and the rank-sum from Kang, and indicate the most reliable method for selecting popcorn cultivars. These statistics were estimated by data of crop yield from 19 Brazilian genotypes under 21 environments and popping expansion under 16 environments. The ωi, \textS\texti(1) {\text{S}}_{\text{i}}^{(1)} , \textS\texti(2) {\text{S}}_{\text{i}}^{(2)} , \textS\texti(3) {\text{S}}_{\text{i}}^{(3)} and sdi2 \sigma_{\rm di}^{2} were positively and significantly correlated indicating that just one in these five statistics is sufficient for selecting stable genotypes although they were not correlated with the means of crop yield and popping expansion. The bi \beta_{\rm i} was negatively and significantly correlated with Pi for crop yield indicating that the most adaptable genotypes tend to have the lowest estimates of Pi. Although Pi was not correlated with ωi, \textS\texti(1) {\text{S}}_{\text{i}}^{(1)} , \textS\texti(2) {\text{S}}_{\text{i}}^{(2)} , \textS\texti(3) {\text{S}}_{\text{i}}^{(3)} , or sdi2 \sigma_{\rm di}^{2} statistics, it displayed positive correlation with the Index 1 (crop yield and popping expansion +  \textS\texti(1) {\text{S}}_{\text{i}}^{(1)} rank) and Index 2 (crop yield and popping expansion + Wi) indicating that superior popcorn genotypes are also stable. Finally, both Pi and the rank-sum are useful statistics in breeding programmes where crop yield, popping expansion and stability are essential traits for selecting genotypes.  相似文献

Papaya seeds (Carica papaya Linn) have been found to have a significant effect on sperm characteristics in some mammals, including humans, but no studies have investigated the effects on dogs. In the present study, a significant decrease in sperm concentration was observed in a group of dogs treated with extracts from washed papaya seeds, but no decrease was observed in the group of dogs treated with non‐washed seeds. An important effect of extract components from washed seeds such as fatty acids is probably involved in the reduction of sperm production because of Sertoli cell damage, as has been suggested for langur monkeys. Dilution of the active components in the non‐washed papaya seeds or interference with some of the components may reduce the expected effect on spermatogenesis. This first report on the effects of a chloroformic extract of papaya seeds in dogs suggests that an increased dose is necessary to achieve azoospermia.  相似文献

Most isolates of foot-and-mouth disease virus (FMDV) display a broad host range. Since the late 1990s, the genetic lineage of PanAsia topotype FMDV serotype O has caused epidemics in the Far East, Africa, the United Kingdom, France, the Netherlands, and numerous other countries throughout Europe and Asia. In contrast, there are several FMDV isolates that exhibit a more restricted host range. A Cathay topotype isolate of FMDV serotype O from the 1997 epizootic in Taiwan (O/TAW/97) demonstrated restricted host specificity, only infecting swine. Methods used to evaluate infectivity and pathogenicity of FMDV isolates in cattle are well-documented, but there has been less progress studying transmission and pathogenicity of FMDV isolates in pigs. In previous studies designed to examine pathogenicity, various chimeric viruses derived from O/TAW/97 were intradermally inoculated in the heel bulb of pigs. Subsequent quantitative scoring of disease and evaluation of virus released into nasal secretions and blood was assessed. Here we prove the usefulness of this method in direct and contact inoculated pigs to evaluate infectivity, pathogenicity and transmission of different Asian FMDV isolates. Virus strains within the Cathay topotype were highly virulent in swine producing a synchronous disease in inoculated animals and were efficiently spread to in-contact naive pigs, while virus strains from the PanAsia topotype displayed more heterogeneous properties.  相似文献

Computing of landscape pattern metrics from spectrally classified digital images is becoming increasingly common. Recently, object-orientated image classification is being seen as an alternative and is tending to replace pixel-based approaches. However, object-based methods are likely to influence and produce biases in the results of these spatial analyses. In this study, the sensitivity of 85 landscape metrics to different classification methods and parameters are analyzed. A Landsat image of a complex mountainous forest region of Mexico was classified using pixel-based and object-based approaches. Nine object-based classified images were obtained using a region-growing algorithm based upon different segmentation parameters. Pixel-based classified images were smoothed using different methods (majority filtering, sieving and clumping). Accuracy assessment was carried such that classified images with similar accuracy were compared. Landscape metrics were then derived from the different classified images and compared through a coefficient of variation computing. Almost all the metrics showed variability due to classification and post-processing methods, particularly core area metrics and some proximity and contagion/interspersion indices. Caution must be observed when comparing values of metrics derived from images with slight differences in their characteristics or in the way they have been processed as, for example, in landscape monitoring studies based upon multidate imagery.  相似文献
