Abstract AIM: To investigate polymeric nanoparticles as an oral delivery system for protein biocontrol agents for control of the brushtail possum, Trichosurus vulpecula. METHODS: Insulin-loaded poly(ethyl 2-cyanoacrylate) (PECA) nanoparticles were prepared using interfacial polymerisation, and characterised for size, zeta potential, and efficiency of encapsulation of insulin. In-vitro release of insulin-loaded PECA nanoparticles was quantified using reverse-phase highpressure liquid chromatography (HPLC). The in-vivo pharmacokinetics of insulin in PECA nanoparticles was investigated following I/V administration, and when injected directly into the caecum alone or in conjunction with the permeation enhancer EDTA. Blood samples were collected at intervals from ?5 to 180 minutes, and the concentration of insulin in plasma was quantified using a radioimmunoassay (RIA) validated for possum plasma. RESULTS: Poly(ethyl 2-cyanoacrylate) nanoparticles were produced with a uniform particle size of 200–300 nm, and the mean entrapment of insulin was 78%. In-vitro release of insulin from the PECA nanoparticles was controlled, although incomplete, and approximately 30% of the insulin remained entrapped. The bioavailability of insulin when administered in a PECA nanoparticulate formulation injected directly into the caecum was <1%, and was not increased by addition of the permeation enhancer. CONCLUSIONS: The nanoparticulate formulations investigated as part of this study resulted in low bioavailability of the peptide insulin in the brushtail possum. 相似文献

Studies were carried out to investigate the effect of crude extracts from resin, leaves, stem barks and root barks of Commiphora swynnertonii against Newcastle disease virus (NDV) using an in ovo assay. Nine-day-old embryonated chicken eggs were divided into seven groups (n?=?6) and received various treatments. Six groups were inoculated with velogenic NDV strain; five groups out of these were treated with different concentrations of the four extracts or a diluent, dimethylsulphoxide. The uninoculated and inoculated groups were left as negative and positive controls, respectively. Embryo survival was observed daily and embryo weights were measured day?5 post-inoculation; a few eggs from selected groups were left to hatch. Allantoic fluid from treated eggs and serum from hatched chicks were collected for hemagglutination and hemagglutination inhibition (HI) tests to detect NDV in the eggs and antibodies against NDV in the hatched chicks respectively. Results showed that embryo survival and mean embryo weight were significantly higher (p?<?0.001) in those groups which were treated with the crude extracts from C. swynnertonii than the positive control group. Also the extracts significantly (p?<?0.001) reduced virus titres, whereas no viruses were detected in the allantoic fluids of the resin-treated group at the highest concentration of 500???g/mL. Furthermore, the HI test results showed very low levels of antibodies against NDV in chicks hatched from resin and root bark extract-treated eggs suggesting that these plant materials were capable of destroying the NDV before stimulating the developing chick??s immunity. The current findings have clearly demonstrated that crude extracts especially that of resin from C. swynnertonii have strong antiviral activity against NDV in ovo. In vivo trials are needed to validate the use of resin from the tree in controlling Newcastle disease in chickens. 相似文献

A cross-sectional study was conducted between January 2007 and February 2008 to estimate seroprevalence of brucellosis and
identify risk factors associated with Brucella infections in commercial cattle in three districts of Lusaka province (Chongwe, Luangwa, and Kafue; n = 849) and in one rural district from the Central province (n = 48). A total of 897 serum samples were randomly collected from 55 farms along with animal-level data such as sex, age,
and parity. Sera were screened for presence of anti-Brucella antibodies using the Rose Bengal test, and positive samples were confirmed using competitive enzyme-linked immunosorbent
assay. At the animal level, seroprevalence was estimated at 7.9% (95% CI = 4.4–11.4%) in the Lusaka province and 18.7% (95%
CI = 7.5–29.9%) for Chibombo district. Brucellosis seroprevalence varied according to district, with Chongwe district recording
the highest compared to other districts. Seroprevalence also varied according to sex with bulls (n = 96) having higher seroprevalence (12.5%; 95% CI = 3.8–21.1%) compared to females (8.1%; 95% CI = 4.6–11.6). Similarly,
seroprevalence varied according to age groups, with the age category 1–4 years recording the highest (10.7%). The study recorded
relatively low Brucella seroprevalence in commercial farms in Lusaka, compared to the traditional small-scale farms. We suggest that testing and
stamping out of infected animals is likely to improve the situation and significantly reduce the public health risk associated

The relay cropping of sesbania (Sesbania sesban) — a N2-fixing legume — with maize (Zea mays) has been proposed as a strategy to increase soil fertility and food production in densely populated areas in southern Africa. We determined the production of relay-cropped maize and sesbania at three landscape positions under researcher-designed and farmer-managed conditions in southern Malawi. Three landscape positions (dambo valley or bottomland, dambo margin with < 12% slope, and steep slopes with > 12% slope) were examined in factorial combination with N sources for maize (no added N, relay-cropped sesbania, and calcium ammonium nitrate fertilizer). Relay cropping of sesbania with maize increased maize grain yield, as compared to unfertilized sole maize in two of three years. Split application of 96 kg N ha–1 as N fertilizer, however, was more effective than sesbania in increasing maize yields. Survival of sesbania seedlings and biomass production of sesbania were greater in the dambo valley and dambo margin than on steep slopes. Maize yields tended to be lower on steep slopes than in the dambo valley and dambo margin areas. Biomass production of sesbania and hence the potential benefits of intercropping sesbania with maize appear greater in the dambo valleys and dambo margins than on steep slopes.This revised version was published online in November 2005 with corrections to the Cover Date. 相似文献

The Mars Global Surveyor (MGS) z-axis accelerometer has obtained over 200 vertical structures of thermospheric density, temperature, and pressure, ranging from 110 to 170 kilometers, compared to only three previous such vertical structures. In November 1997, a regional dust storm in the Southern Hemisphere triggered an unexpectedly large thermospheric response at mid-northern latitudes, increasing the altitude of thermospheric pressure surfaces there by as much as 8 kilometers and indicating a strong global thermospheric response to a regional dust storm. Throughout the MGS mission, thermospheric density bulges have been detected on opposite sides of the planet near 90 degreesE and 90 degreesW, in the vicinity of maximum terrain heights. The study was undertaken to evaluate form factors for Pinus kesiya growing in the three Zambia Forest and Forestry Industries Corporation (ZAFFICO) plantations – the Ndola, Ichimpe and Chati groups – under different rainfall (900–1 500?mm) and soil conditions (pH 4.0–5.5). Trees were sampled from the three plantation groups and 10 sample plots were established in which 10 trees were randomly sampled for cutting. Height and diameter at breast height (dbh) were the main variables that were measured and constituted the collected data. The trees were then felled and cut into segments of 0.5?m from the bottom to the top of the tree and segment diameters over bark were measured at both ends. The form factor for each tree was then calculated by dividing the sum of the volume of the tree segments by the estimated cylindrical volume of the standing tree. The results showed significant differences (p-value<0.05) in form factors among the three plantation groups. The study further showed a significant difference in form factor when the three plantation groups were compared with a fixed form factor (0.4) used by ZAFFICO plantations, indicating that the form factors are site specific. Therefore, for the benefit of customers and plantation owners, it is important that area-specific form factors should be used for each plantation group, as these groups are affected by different factors such as soils characteristics and rainfall, which have a direct effect on the form factor. 相似文献
