Assuming a synergistic or additive effect of Chlamydiaceae in coexistence with other enteropathogenic agents, the viral/bacterial interaction between a cell culture adapted porcine epidemic diarrhea virus (ca-PEDV) and different Chlamydiaceae strains was studied in vitro. Vero cells were dually infected with ca-PEDV and one of the three chlamydial strains Chlamydia trachomatis S45, Chlamydophila abortus S26/3 or Chlamydophila pecorum 1710S. Three experimental protocols were designed varying the inoculation sequence. Cell layers were first inoculated with Chlamydiaceae and 20 h later with ca-PEDV in protocol one. In protocol two, both agents were administered concurrently, whereas in protocol three, ca-PEDV was applied 20 h in advance of the Chlamydiaceae. Immunofluorescence techniques, immunohistochemical (IH) staining and electron microscopy were subsequently employed to investigate the cell layers. Using indirect immunofluorescence (IF) labeling, all mixed infections revealed dually infected cells, however, only incidentally and in low numbers. Characteristically, ca-PEDV syncytia with one or more chlamydial inclusions were detected but dually infected single cells were absent. Some syncytial cells contained enlarged C. abortus or C. pecorum inclusions with abnormally large developmental forms. In comparison with simultaneously conducted monoinfections, larger chlamydial inclusions were observed in dually infected cell layers. Experiments with C. trachomatis showed significantly increased numbers of chlamydial inclusions in dually infected cell layers compared to monoinfected ones. These findings indicate an influence of ca-PEDV on the chlamydial developmental cycle and in the case of C. trachomatis, a positive effect on chlamydial colonization in mixed infections. 相似文献

Cervical swabs and serum samples were taken from Swiss herds of sows with high rates of irregular return to oestrus (group A) and from control herds without reproductive problems (group B. The genital tracts of 21 slaughtered sows of group A were also examined. The swabs and genital tracts were screened for Chlamydiae by a new 16S rRNA PCR and the sera by an ELISA for Chlamydiaceae lipopolysaccharide. Chlamydophila (Cp) abortus was isolated from seven of the 65 swabs taken from group A but from none of the 128 swabs taken from group B. Chlamydia suis was present in swabs from both groups A (1.5 per cent) and B (2.3 per cent). In addition, Cp abortus was detected in 33.3 per cent of the genital tracts. Of the 193 sera tested, 61.7 per cent were positive, with no significant difference between group A (52.3 per cent) and group B (66.4 per cent). Chlamydia-like organisms were detected in 28.2 per cent of the swabs from group A and in 22 per cent of those from group B. 相似文献

Artificial insemination is a valuable method for facilitating genetic exchange between captive colonies of koalas (Phascolarctos cinereus) and for the maintenance of genetically important remnant populations. However, to reduce potential disease transmission, their semen needs to be screened for venereal diseases caused by organisms such as Chlamydia species. Semen samples from 11 koalas, eight of them with clinical signs of cystitis, were examined for the presence of Chlamydia by an optimised PCR assay. Chlamydia was detected in semen from seven of the 11 animals. 相似文献

On a farm housing cattle and goats an abortion storm occurred affecting 50% of the goats during the lambing season 2000/2001. In one of three investigated caprine abortions Chlamydophila abortus could be identified as etiology. During this time a pregnant woman (pregnancy week 19/20) had contact with aborting goats. She developed a severe generalized infection and aborted. The placenta contained Chlamydophila abortus shown by immunohistochemistry and PCR. Aim of the present case report is to alert veterinarians about the potential zoonotic risk of ovine/caprine abortions. 相似文献

The present study gives an overview over the seroprevalence of ovine enzootic abortion in Switzerland. 639 sheep flocks out of eight cantons in Switzerland were examined by a competitive enzyme linked immunosorbent assay (cELISA) for antibodies against Chlamydophila abortus (Chlamydia psittaci serotype 1), the agent causing ovine enzootic abortion. The eight cantons included Aargau, Bern, Zürich, Appenzell-Ausserrhoden, Appenzell-Innerrhoden and Fribourg, the Vallais and the Graubünden. They were representative for 57% of the Swiss sheep flocks and for 60% of Swiss sheep population. In total, almost 19% (118) of the examined flocks were seropositive. Seroprevalence was the highest in Graubünden with 41%; this requires further examination and the evaluation of the need for a monitoring and controlling program. The examination of pooled sera made it possible to test a large number of samples with a reasonable amount of work. Higher sensitivity (92.9%) and specificity (97.6%) than the complement fixation test (CFT) in combination with testing of pooled sera makes the cELISA to be an usuable tool for serological screening on flock level. 相似文献

A short literature review of feline infectious pneumonia, feline Chlamydia and Paramyxoviridae is presented. In a retrospective study (from 1987 to 1996) 245 cases of feline pneumonia or conjunctivitis/rhinitis were investigated: histological diagnoses and aetiologies were compared; all lungs were examined immunohistologically for the occurrence of chlamydia and of canine distemper virus (CDV), but neither pathogen could be demonstrated. The results confirm previous reports indicating that feline chlamydia is not a primarily pulmonary pathogen and that CDV is not a causative agent of pneumonia in cats as it is in large felids.The review provides a summary of the known causes and pathology of infectious pneumonia in cats (in order of frequency), although some remain aetiologically uncertain. It focuses on chlamydia and distemper virus - a recognized and as yet unknown cause of feline pneumonia. The role and especially the frequency of chlamydia as a cause of feline pneumonia are controversial but distemper virus, known to cause pneumonia in dogs and large felids, has not as yet been demonstrated in cats. The aims of the retrospective study were to determine the occurrence of chlamydia in 245 cases of feline pneumonia or conjunctivitis/rhinitis, and to investigate the presence of CDV in these lungs. 相似文献

During a canine distemper epidemic in Switzerland in 1984/85 six cases of spontaneous mixed infections of distemper virus and Toxoplasma spec. were diagnosed. An etiologic diagnosis was made by immunohistological PAP-labelling. Pathological including histopathological findings are reported. The possibilities of histological identification of the organisms (H & E staining) are compared with immunohistology.
