牛传染性鼻气管炎病毒gD基因的克隆与表达

构建pET32a重组表达载体,转化到BL21(DE3),诱导表达牛传染性鼻气管炎病毒重组gD蛋白。用已发表的IBRV的gD基因序列设计一对特异性引物,通过PCR方法扩增一条766bp的目的基因gD,将gD基因克隆到原核表达载体pet32a,得到重组表达载体pet32-gD,转化到BL21(DE)中,通过IPTG诱导获得融合蛋白。重组表达蛋白经纯化后,免疫印迹分析。结论证明表达的重组蛋白具有良好的免疫原性。  相似文献   

Implantation study on the potential of murine epidermal stem cell differentiation

AIM: To investigate the potential of murine epidermal stem cell (ESC) differentiation after seeded in a biodegradable carrier and implanted subcutaneously into syngeneic recipient mice. METHODS: ES cells were induced in vitro to differentiate into ESCs. After stained with a fluorescent dye Hoechst 33342, these ESCs were seeded into a polyglycolic acid (PGA) net containing collagen gel, functioning as a cell carrier, and implanted subcutaneously into 129/J mice, which were syngeneic to these stem cells. RESULTS: The ESCs kept alive in the implant when observed under a fluorescent microscopy 3 weeks or longer after implantation, and could differentiate into hair follicle-like structure, glandular structure, and gave rise to additional structures displaying features resembling native dermis. No apparent rejection or severe side effects were observed at least 10 weeks post-implantation. CONCLUSION: It is feasible to use these ESCs as seed cells in the study to fabricate dermal equivalent having the potential to develop dermal appendages.  相似文献

通过半轮配双列杂交,分析了甜瓜可溶性固形物含量的遗传特性。研究结果表明,遗传方差组成中,基因的加性效应和显性效应都是显著的,但加性效应占相对主要优势,狭义遗传力(h2N)值为65%。基因的分布是不均衡的,控制该性状的基因W为显性基因,但表现为不完全显性,可溶性固形物含量选择的较好世代在F4代。  相似文献

干法膨化宠物饲料技术参数关系的研究

采用双螺杆膨化机干法工艺对宠物全价粉状饲料进行膨化并制粒,研究所得产品的容重与淀粉糊化度、硬度指标之间的关系,以及设备参数和配方特性对颗粒饲料容重的影响。结果表明:容重与淀粉糊化度、硬度密切相关;设备参数与配方特性均对产品容重有极显著影响;通过容重的测定估测其熟度和硬度具有可行性;可以通过调节喂料速度和膨化玉米比例控制产品容重。  相似文献

Ultrastructural study on scab resistance expressed in epidermal pectin layers of pear leaves

Proliferation and collapse of subcuticular hyphae of Venturia nashicola race 1 were studied ultrastructurally, after inoculation of susceptible Japanese pear cv. Kousui, resistant Japanese pear cv. Kinchaku, resistant Asian pear strain Mamenashi 12 and nonhost European pear cv. Flemish Beauty leaves, to understand the nature of the resistance mechanism. After cuticle penetration by the pathogen, the hyphae were observed at lower frequency in epidermal pectin layers and middle lamellae of leaves of the three resistant plants than in those of susceptible ones. This result suggested that fungal growth was suppressed in the incompatible interaction between pear and V. nashicola race 1. In the pectin layers of all inoculated plants, some hyphae had modifications such as breaks in the plasmalemma with plasmolysis, necrotic cytoplasm and degraded cell walls. More hyphae had collapsed in the leaves of the three resistant plants than in those of the susceptible cv. Kousui. In collapsed hyphae, the polymerized cell walls broke into numerous fibrous and amorphous pieces, showing that the scab resistance might be associated with cell wall-degrading enzymes from pear plants.  相似文献

不同品系及不同生长年限玉竹挥发油含量分析

以吉林省种植的"大玉竹"、"圆叶玉竹"及"吉竹1号"根茎为研究对象,采用水蒸气蒸馏法提取挥发油,应用GC-MS法分析比较,测定挥发油组成,用峰面积归一法测定了各组分成分的相对含量。结果表明:丹皮酚为"大玉竹"挥发油区别另2种挥发油特有成分,六甲基环三硅氧烷为3个品系玉竹挥发油中共有成分,该研究为玉竹品种选育、质量评价及后续产品的开发奠定基础。  相似文献
