To explore the value of Mb1230 protein of Mycobacterium bovis in the diagnosis of bovine tuberculosis,we obtained the Mb1230 gene by PCR and constructed recombinant plasmid pET-22b-Mb1230.Recombinant Mb1230 protein was obtained by IPTG induction and purified by affinity chromatography.The activity of the recombinant protein was evaluated by TST test,IGRA test and indirect ELISA.The size of the recombinant protein matched with the theoretical value proved by SDS-PAGE;Western blotting result showed that the recombinant protein could react with mouse anti-His antibody,and had specific band;The results of TST test,IGRA test and indirect ELISA test also showed the recombinant protein had antigenic activity.The results indicated the recombinant protein Mb1230 had good B cell and T cell activity,so,it had the potential application in the diagnosis of bovine tuberculosis. 相似文献

In order to establish an indirect ELISA method for detection of equine herpesviruses type 4 （EHV4） antibody, the glycoprotein G （gG） protein with specific epitope worked as a detection antigen. After optimizing conditions, the indirect ELISA method was developed successfully，specificity and repeatability tests were determined. The result was that the EHV4 gG only reacted with antibodies against EHV4;but not with antibodies against EHV1; both the intro-batch and inter-batch variation coefficiencies were lower than 10%.Concordance of the indirect ELISA relative to commercial EHV1/4 antibody kit was above 90%.The results indicated that the indirect ELISA method could be used for the detection and epidemiological surveys of EHV4 infection. 相似文献

为明确在覆膜滴灌条件下玉米品系(种)主要农艺性状与产量的关系,应用DPS软件对15个玉米杂交品系(种)的主要农艺性状和产量性状进行相关性、灰色关联度等分析,结果表明:籽粒重、穗重、产量的变异系数较大;玉米产量与生育期、籽粒水分含量、穗重、籽粒重呈显著正相关关系;聚类分析可将参试的15个品系(种)划分为3类,第Ⅲ类表现良好,值得进行推广种植。灰色关联度分析表明,玉米籽粒重、穗重、生育期、籽粒水分含量等性状是影响玉米产量的主导因素,在玉米高产育种中,应以其为主攻目标,提高玉米后代的有效选择,加速高产育种进程。 相似文献

为了探索一条能大批量处理无籽西瓜种子,有效提高种子活力的途径,应用水势0、-0.2、-0.6和-1.2MPa的聚乙二醇(PEG-6000)溶液对三倍体无籽西瓜种子进行3、6、9、12h引发处理,并与常规萌发条件下的种子萌芽情况进行比较分析。结果表明:(1)各种水势的PEG-6000溶液引发对无籽西瓜种子发芽各项指标均没有显著影响。(2)3h引发处理相对于其他引发时间处理,能最大程度地提高无籽西瓜种子发芽能力,在发芽率、发芽势、发芽指数和活力指数的影响上均存在明显差异,这与常规生产上无籽西瓜浸种时间结果一致。 相似文献

本试验采用不同比例的木耳菌糠替代部分木屑袋栽滑菇,以培养料（木屑89%、麦麸10%、石膏1%）为对照,探讨木耳菌糠栽培滑菇的可行性。结果表明,木耳菌糠添加量25%～45%时,可缩短发菌天数,其中菌糠添加量为45%时,满袋天数仅需52 d,并且长势较好;但菌糠添加量为55%～65%时,则延长了发菌天数,而且菌丝稀疏,长势也弱。对照（CK）与木耳菌糠添加量是25%～45%时,子实体生长良好,并且出菇整齐;菌糠添加量为55%～65%,则出菇不整齐。CK的生物学效率最高,但配方4（菌糠45%、木屑34%、麦麸10%、石膏1%）的生物学效率与CK差异不大,从成本和生态效益考虑,配方4栽培滑菇具有可行性,并已在广灵县进行推广。 相似文献
