Pinus radiata (D. Don) seedlings were grown for 100 days at day/night temperatures of 10/1, 15/1, 20/1 and 25/1 degrees C, to determine whether temperatures above a threshold of 5 degrees C influence frost hardiness development. Relationships between hardening and relative growth rate, carbohydrate concentration and net carbon balance were also investigated. Seedlings hardened at a nearly constant rate in each treatment, although the rate of hardening was strongly temperature dependent. It increased as the temperature declined, but in a curvilinear fashion. Temperatures below 9.5 degrees C were effective in hardening the seedlings. During the daily temperature cycle, dehardening occurred at temperatures above the threshold, whereas hardening occurred at temperatures below the threshold. The net difference between the two processes determined the development of frost hardiness. The development of frost hardiness was negatively correlated with relative growth rate and positively correlated with the accumulation of starch and sugars. We conclude that frost hardening is a complex process that is causally linked to carbohydrate concentrations.  相似文献

Lectins are plant secondary metabolites (PSM) found in many forages and which may confer anthelmintic properties to gastrointestinal parasites through disrupting the development of parasitic larvae throughout its life cycle. In experiment 1, the ability of the plant lectins jacalin (JAC), concanavalin A (Con A), phytohemagglutinin E2L2 (PHA-E2L2), phytohemagglutinin L4 (PHA-L4), phytohemagglutinin E3L (PHA-E3L), kidney bean albumin (KBA), Robinia pseudoacacia agglutinin (RPA), Maackia amurensis lectin (MAA), Maclura pomifera agglutinin (MAA), Dolichos biflorus agglutinin (DBA), wheat germ agglutinin (WGA) and Galanthus nivalis agglutinin (GNA) to disrupt the feeding of the first stage larvae (L(1)) of the sheep gastro-intestinal nematodes (GIN) Teladorsagia circumcincta, Haemonchus contortus and Trichostrongylus colubriformis was investigated using a larval feeding inhibition test (LFIT). Only PHA-E3L, WGA and Con A had a potent effect on disrupting larval feeding of all of the three species of GIN investigated. The lectin concentration required to inhibit feeding in 50% of L(1) (IC50) was 7.3±1.2, 8.3±1.4 and 4.3±1.7 μg/ml for PHA-E3L; 59.1±32.4, 58.7±11.9 and 8.1±7.0 μg/ml for Con A and 78.9±11.2, 69.4±8.1 and 28.0±14.1 μg/ml for WGA for T. circumcincta, H. contortus and T. colubriformis larvae, respectively (P=0.006). The addition of the lectin inhibitors fetuin, glucose/mannose or N-acetylglucosamine for PHA-E3L, Con A and WGA, respectively, caused an increase in the proportion of larvae that had fed at all concentrations for PHA-E3L only. In experiment 2, the effect of extracts from the tropical plants Azadiractha indica, Trichanthera gigantea, Morus alba, Gliricidia sepium and Leucaena leucocephala on the feeding behaviour of H. contortus L(1,) was examined. A. indica, T. gigantea and M. alba failed to inhibit 50% of larvae from feeding at concentrations up to 10mg plant extract per ml. In contrast, both G. sepium and L. leucocephala demonstrated a dose-dependent effect on larval feeding with respective IC50 estimates (mean±s.e.) of 0.015 mg/ml ±0.001 and 3.465 mg/ml ±0.144, effects which were partly reversed by the inclusion of either the tannin inhibitor polyethylene glycol or the lectin inhibitor Fetuin. These studies demonstrate that plant lectins can have an inhibitory effect on the feeding behaviour of first stage larvae of ovine GIN in vitro. Moreover they also provide novel evidence that lectins may contribute to the anthelmintic properties of some tropical forage plant extracts, such as G. sepium and L. leucocephala.  相似文献

This study was designed to evaluate the cats' acceptance and compliance of the owners and cats towards an extemporaneously prepared palatable compounded atenolol (paste and suspension) formulation in comparison to the commercially obtained tablet, in a randomised, cross-over study design.The three formulations were prescribed twice daily for 6 days to 13 healthy privately-owned cats of 13 different owners, with varying levels of experience in medicating cats. Daily compliance was evaluated via an owner-completed diary, completed after each dose administered. Owner's experience and preference of the formulation was evaluated via questionnaires given prior to, at the end of each treatment protocol, and upon completion of the study. Although compounded suspension was association with fewest missed doses, the majority of cat owners expressed a preference for the divided tablet. Atenolol tablets, compounded paste and suspension acceptance and compliance were comparable. Further work is now required to assess the amount and stability of the active ingredient and the robustness of the paste and suspension formulations prior to any bioavailability comparisons between the formulations.  相似文献

Two experiments were conducted to study the effects of six processing techniques for barley grain in a 3 x 2 factorial arrangement of grain conditions and roller settings on ruminal degradation of the grain (Exp. 1) and on growth performance by 138 feedlot steers (n = 23 per treatment; Exp. 2). Dry barley (11% moisture, D barley), barley tempered to 20% moisture (M barley), and barley tempered with 60 mL/t of surfactant-based tempering agent (GrainPrep, Agrichem, Inc., Anoka, MN; MS barley), were each rolled at two roller settings selected from preliminary tests. The settings selected for the study were RD, the roller position that had yielded optimally processed D barley, and RMS, the setting that had yielded optimally processed MS barley. Setting RMS was tighter than RD. Barley rolled at the RMS setting was more extensively processed (i.e., had a lower [P < 0.001] processing index, PI), had lighter (P < 0.001) volume weight, thinner (P < 0.001) kernels, and fewer (P < 0.001) whole kernels compared with setting RD. Tempering did not affect (P > 0.05) PI, percentage of whole kernels, or kernel thickness at either roller setting. The processing characteristics of tempered barley were unaffected (P > 0.05) by surfactant. The extent of in situ DM disappearance (ISDMD) was higher (P < 0.01) in grain rolled at setting RMS compared with RD. At both roller settings, tempering reduced (P < 0.05) ISDMD between 4 and 24 h of ruminal incubation. Steers fed RMS-rolled barley had lower (P < 0.001) DMI, slightly lower (P = 0.084) ADG, but increased (P < 0.05) gain:feed (G:F) compared with steers fed RD-rolled barley. Tempering did not affect (P > 0.05) ADG, DMI, or G:F during backgrounding, but improved (P < 0.01) these variables during finishing. Surfactant improved (P < 0.05) G:F but not DMI or ADG. The improvement in G:F was most pronounced when setting RMS was used. The optimal PI values calculated from performance data were numerically greater for the backgrounding diet than for the finishing diet. Steers fed M or MS barley had heavier (P < 0.01) hot carcasses and thicker (P < 0.05) fat cover but lower (P < 0.05) dressing percentages than steers fed D. When the feed barley was rolled at setting RMS, steers fed MS barley produced heavier (P < 0.05) carcasses than those fed M. Tempering with or without surfactant increased performance by feedlot steers compared with not tempering. Diet composition and degree of barley processing mediated this effect.  相似文献

Eichhornia crassipes (water hyacinth), a major aquatic weed in India, is one of the targets in a biological weed control research programme in India. A fungal pathogen, Alternaria alternata, is being evaluated as a biological control agent for this weed. The feasibility of solid substrate for the mass production of A. alternata has been examined. Conidia production and virulence of A. alternata were affected by temperature, light and incubation period. The highest number of conidia were produced on rice seed followed by wheat, sorghum, maize seeds and cornmeal at 20°C when exposed to near‐ultraviolet than on the other substrates, while the least conidia were observed on these substrates under light conditions. At 20°C, large numbers of virulent conidia were produced on rice seeds after 4 weeks of incubation under constant dark conditions. Henceforth, the use of rice seeds as a solid substrate for production of A. alternata could be a feasible method to produce conidia in a village co‐operative scenario in India.  相似文献

In swine, the use of frozen-thawed (FT) sperm for artificial insemination (AI) is limited because of poor sow fertility, possibly associated with a post-thaw capacitation-like status resulting in fewer fully viable sperm. Sow fertility to AI with FT sperm may improve with deeper deposition of sperm within the female tract, insemination very close to ovulation, or reversal of cryocapacitation by seminal plasma (SP). We performed two experiments to examine these suggestions. In experiment 1, 122 multiparous Yorkshire sows received 600 IU equine chorionic gonadotrophin at weaning and 5 mg pLH 80 h later to control time of ovulation. The predicted time of ovulation (PTO) was 38 h after pLH injection. Thereafter, sows were assigned on the basis of parity to a single AI of FT sperm at 2 h before PTO, or at 12 h before PTO, or FT sperm supplemented with 10% SP at 12 h before PTO. Control sows received fresh semen at 12 h before PTO. All semen doses were adjusted to 3 x 10(9) live cells and deposited into the cervix. Experiment 2 employed 99 multiparous crossbred sows and repeated the treatments of experiment 1 except that all FT inseminations were intrauterine. In both experiments, farrowing rates were lower (p < 0.01) following FT inseminations with no effect of time of insemination or of supplemental SP. In experiment 1, litter size was smaller following FT insemination (p < 0.05), but no effect on litter size was evident in experiment 2. Supplemental SP had no effect on litter size in either experiment. The lack of effect of either SP or timing of FT insemination on sow fertility suggests that the non-lethal sperm cryoinjury affecting fertility involves more than just cryocapacitation.  相似文献
