In order to establish the mammalian parasitic cycle of the nematode Neostrongylus linearis 18 lambs were infected with 5000 third-stage larvae obtained from the snail Cernuella (Xeromagna) cespitum arigonis. In the subsequent days, larvae were found in the walls of the caecum and colon, in the liver and the lungs, suggesting that the bloodstream is the main migratory route. The presence of low numbers of migrating larvae in the mesenteric lymph nodes indicates that this route is probably secondary. The first still sexually undifferentiated, fourth-stage larvae were found in the lungs on the 8th day post-infection (p.i.) and the first sexually differentiated fifth-stage larvae, on the 10th day. Also described are the macroscopic changes of the parasitized organs.  相似文献

Systemic ciliatosis caused by histophagous ciliates constitutes a serious disease of cultured turbot. Six ciliate isolates were obtained from parasitized turbot during six epizootics at four different farms located in Spain, France and Portugal. Axenic cultures of the six isolates were obtained by periodical subculturing in ATCC 1651MA or supplemented L-15 media. In basal media or seawater, the parasites could survive starving for long periods with no apparent proliferation. In adequate media, growth kinetics was found to be very similar for isolates A and B, with a clear influence of temperature. Morphological studies demonstrated that all isolates share common features that allows their assignment to either Philasterides Kahl, 1931 or Miamiensis Thompson et Moewus, 1964. However, statistically significant differences were evident in pairwise comparisons of the isolates from the four farm sites in 16 taxonomically relevant morphometric features. This could allow the discrimination of different species or strains. Virulence of isolates A and B for healthy turbot was tested in several experiments. Differences in the virulence were especially evident after long-term in vitro culturing, isolate A being clearly attenuated after 35-42 passages, whereas isolate B became more virulent after 20-42 passages. The need of further studies to confirm such virulence variability and its implications in pathogenesis and prevention of turbot scuticociliatoses is stressed.  相似文献

A Geographic Information System (VetEpiGIS) was used to analyze the ADV (Aujeszky's disease virus) sero-status in large-scale pig units regarding certain geographical features in a county of southern Hungary. The ADV sero-statuses were collected from all swine units in Csongrád county in 1998-2000. The units' coordinates were combined with a vector graphical digital map of the county, with a resolution of 1:100,000. Logistic regression tested the associations between sero-status of large-scale units and presence of topographical features, other units and villages in the neighborhood. "Neighborhood" was defined by circular zones with radius 1-10km around the unit (in 1km increments; one logistic regression for each radius). The following topographical features showed significant positive association with the ADV seropositivity: lake (3km OR: 5.7; 5km OR: 7.5; 6km OR: 6.1; 10km OR: 5.4) and highway (5km OR: 4.2; 6km OR: 5.3). Other features had negative association with ADV seropositivity: forest (3km OR: 0.13; 4km OR: 0.15; 5km OR: 0.15; 6km OR: 0.10; 7km OR: 0.10; 8km OR: 0.23) and uninfected large-scale unit (4km OR: 0.07; 5km OR: 0.27; 6km OR: 0.32; 7km OR: 0.31).  相似文献

Embryonic mortality is a significant problem plaguing the hatching success. Its early forms are especially hardly distinguishable from true infertility. Propidium iodide (PI) staining of the germinal disc combined with outer perivitelline layer (OPVL) sperm counting was used for the determination of 'true' fertility of duck eggs in two different experiments: fertility investigation on fresh, unincubated eggs of Hungarian ducks and on incubated eggs of a crossbred, selected as 'infertile' at the 7th day of incubation. Examination of the relationship between OPVL sperm count and fertility seems to be an adequate tool for checking the effectiveness of insemination programmes and the fertilising capacity of poultry spermatozoa. The proportion of fertile eggs was around 50% when the number of OPVL sperm was between 0.1 and 0.2 spermatozoa/mm2. Ninety-nine percent of the eggs containing > 0.3 OPVL sperm/mm2 were fertile and all of the eggs containing < 0.05 sperm/mm2 were infertile. To assure the accuracy of fertility prediction by OPVL sperm counting, PI staining of the germinal disc was used to determine fertility in uncertain cases. Identification of very early embryonic mortality, i.e. that occurring before oviposition, is very difficult. The use of a dissecting microscope for the assessment of real fertility is suitable in most of the cases, while PI staining of the germinal discs proved to be more reliable for detecting very early embryonic death. The combination of the two methods proved to be a useful tool for detecting the 'true' fertility of duck eggs of different breeds.  相似文献

Fifteen dairy farms in seven states on the east coast of the US were each visited on two consecutive years to determinate the prevalence of Cryptosporidium species in pre-weaned (5 days to 2 months) and post-weaned calves (3-11 months), respectively. After each of 971 fecal specimens collected directly from each calf was sieved and subjected to density gradient centrifugation to remove debris and concentrate oocysts, specimens were examined by immunofluorescence microscopy, and polymerase chain reaction (PCR). For all PCR-positive specimens the 18S rRNA gene of Cryptosporidium was sequenced. Cryptosporidium was identified from all farms. Types of housing appeared to have no influence with regard to prevalence of infection. Of 971 calves, 345 were infected with Cryptosporidium (35.5%), but more pre-weaned calves (253 of 503; 50.3%) than post-weaned calves (92 of 468; 19.7%) were found to be infected. A total of 278 PCR-positive specimens characterized by gene sequencing revealed Cryptosporidium parvum, Cryptosporidium andersoni, and two unnamed Cryptosporidium genotypes Bovine B (AY120911) and deer-like genotype (AY120910). The prevalence of these Cryptosporidium species and genotypes appeared to be age related between pre- and post-weaned calves. C. parvum, the only zoonotic species/genotype, constituted 85% of the Cryptosporidium infections in pre-weaned calves but only 1% of the Cryptosporidium infections in post-weaned calves. These findings clearly demonstrate that earlier reports on the presence and prevalence of C. parvum in post-weaned cattle that were based solely on oocyst morphology must be reassessed using molecular methods to validate species and genotype. This finding also indicates that persons handling or otherwise exposed to calves under 2 months of age are at greater risk of zoonotic infection from Cryptosporidium than the risk of infection from exposure to older calves.  相似文献

The prevalence of anti-Toxoplasma gondii antibodies was evaluated by the indirect immunofluorescent-antibody test in serum of 57 wild canids from three different species: Lycalopex gymnocercus, Cerdocyon thous and Dusicyon vetulus from the northeast, southeast and southern regions of Brazil. The prevalence was 35.1%, with 20 of the 57 canids demonstrating antibodies anti-T. gondii at dilutions of 1:16 in 2, 1:32 in 4, 1:64 in 2, 1:128 in 2, 1:256 in 6, 1:512 in 2 and 1:2048 in 2 animals. None of the D. vetulus were positive. Among the L. gymnocercus 11 (91.7%) of the 12 samples were positive and among C. thous 9 (60%) of the 15 had antibodies anti-T. gondii.  相似文献

Prevalence of Giardia duodenalis genotypes in pre-weaned dairy calves   总被引:1,自引:0,他引:1  
To determine the prevalence of Giardia genotypes in pre-weaned dairy calves, fecal samples were collected from a minimum of 18, 1-7-week-old dairy calves per farm on two farms each in the states of Vermont, New York, Pennsylvania, Maryland, Virginia, North Carolina, and Florida. Samples cleaned of fecal debris and concentrated using CsCl density gradient centrifugation were stained and examined by immunofluorescence microscopy and also subjected to PCR and gene sequence analysis. Prevalence by PCR ranged from 9% on a farm in Pennsylvania to 93% on a farm in Vermont, with an average prevalence for 407 calves on 14 farms of 40%. Gene sequence analysis of the TPI, beta-giardin and 16S rRNA genes revealed 85% of the positive samples to be Assemblage E, while 15% were Assemblage A, although the percentages of these genotypes varied greatly from farm to farm. Some farms had no Assemblage A Giardia. Thus, while a majority of the calves were infected with a genotype that is not known to be infectious for humans, calves on 7 of 14 farms did harbor Assemblage A Giardia. Calves should be considered as a potential source of human infectious cysts in the environment.  相似文献

Organisms in the genus Anaplasma are obligate intracellular pathogens that multiply in both vertebrate and invertebrate hosts. The type species, Anaplasma marginale, causes bovine anaplasmosis and infects erythrocytes of the vertebrate host and undergoes a complex developmental cycle in ticks which serve as biological vectors. Infected cattle, wild ruminants and ticks can all serve as reservoirs of A. marginale. In this study, hunter killed Iberian red deer (Cervus elaphus hispanicus) from the region of Castilla-La Mancha in southwestern Spain were tested for Anaplasma infection. We found that 10% of the deer examined were seropositive for Anaplasma. Three A. marginale strains were subsequently obtained from salivary glands of Hyalomma marginatum that were removed from these deer, and the sequence of the major surface protein (msp)4 gene was determined for each strain and used for phylogenetic studies. Maximum parsimony analyses of msp4 sequences from H. marginatum ticks in comparison with New World cattle and bison isolates reported previously, suggested different origins for these Spanish A. marginale strains. The results of this study demonstrated that Iberian red deer are naturally infected with Anaplasma, and may therefore serve as a wildlife reservoir of the pathogen. Although the link between deer infection and the strains of A. marginale identified in ticks was not established, H. marginatum and Rhipicephalus bursa were identified as potential biological vectors for A. marginale in this region and may effect transmission of A. marginale between deer and cattle populations.  相似文献
