Cation‐exchange–capacity (CEC) results of calcareous soils and clays can be erroneous if the ammonium acetate method is used. In this study, a model is proposed to explain the process for systematic underestimation of the CEC. Seven clayey sediments from Germany with varying calcite and low organic‐C content were studied. After several exchange treatments with concentrated ammonium acetate (NH4Ac) solutions, the exchange population is assumed to be in homoionic ammonium form. Throughout the cation‐exchange experiment, calcite reacts with the NH4Ac exchange solution generating Ca2+ cations. During the necessary washing steps to remove excess salt, calcite dissolution is lower but still occurs. The permanently added Ca2+ ions compete successfully with NH , especially during the washing steps. This leads to a more or less partial biionic exchange population resulting in an underestimation of the CEC which is calculated based on NH concentration of the clay by Kjeldahl analysis. The biionic exchange population was proven using the new silver thiourea technique with presaturation of calcite, AgTU calcite . The clay with 148 g kg–1 calcite had a fraction of 16.4 cmol+ kg–1 exchangeable Ca2+. This is ca. 50% of the CEC of this clay being 31.8 cmol+ kg–1. For clays with similar mineralogical composition, this trend is proportional to the calcite content.  相似文献

Normal intestinal mucosa contains abundant immunoglobulin A (IgA)-secreting cells, which are generated from B cells in gut-associated lymphoid tissues (GALT). We show that dendritic cells (DC) from GALT induce T cell-independent expression of IgA and gut-homing receptors on B cells. GALT-DC-derived retinoic acid (RA) alone conferred gut tropism but could not promote IgA secretion. However, RA potently synergized with GALT-DC-derived interleukin-6 (IL-6) or IL-5 to induce IgA secretion. Consequently, mice deficient in the RA precursor vitamin A lacked IgA-secreting cells in the small intestine. Thus, GALT-DC shape mucosal immunity by modulating B cell migration and effector activity through synergistically acting mediators.  相似文献

Loose smut fungi of barley and wheat (Ustilago nuda and U. tritici, respectively) colonize the plant without causing obvious disease symptoms before heading. The availability of diagnostic methods to detect and follow the growth of these pathogens in the plant would therefore be highly advantageous for both resistance breeding and the development of effective seed treatments. Using seed lots of barley and wheat highly infected with loose smut, we studied the early establishment of the loose smut pathogens in the plant by fluorescence microscopy. In hand-cut sections stained with the fluorochrome Blankophor?, fungal hyphae were observed to invade the shoot apical meristem and leaf primordia during the first days after the onset of germination. At the first node stage the ear and leaf primordia were generally extensively colonized. Hyphae of U. nuda were also regularly observed in high density in the nodes. A protocol was developed for the specific amplification of U. nuda and U. tritici DNA extracted from infected plant tissue. PCR screening of U nuda in seedlings from infected and healthy seed lots was compared to ELISA, microscopy and ultimately head infection of mature plants derived from tillers of the tested seedlings. The results indicated that a prediction of loose smut infection by real-time PCR is possible at the second leaf stage, and that the assay is equally suited for use with spring and winter varieties of barley and wheat.  相似文献

The fine particles serving as cloud condensation nuclei in pristine Amazonian rainforest air consist mostly of secondary organic aerosol. Their origin is enigmatic, however, because new particle formation in the atmosphere is not observed. Here, we show that the growth of organic aerosol particles can be initiated by potassium-salt-rich particles emitted by biota in the rainforest. These particles act as seeds for the condensation of low- or semi-volatile organic compounds from the atmospheric gas phase or multiphase oxidation of isoprene and terpenes. Our findings suggest that the primary emission of biogenic salt particles directly influences the number concentration of cloud condensation nuclei and affects the microphysics of cloud formation and precipitation over the rainforest.  相似文献

Crystal defects form during tectonic deformation and are reactivated by the shear stress associated with passing seismic waves. Although these defects, known as dislocations, potentially contribute to the attenuation of seismic waves in Earth's upper mantle, evidence for dislocation damping from laboratory studies has been circumstantial. We experimentally determined the shear modulus and associated strain-energy dissipation in pre-deformed synthetic olivine aggregates under high pressures and temperatures. Enhanced high-temperature background dissipation occurred in specimens pre-deformed by dislocation creep in either compression or torsion, the enhancement being greater for prior deformation in torsion. These observations suggest the possibility of anisotropic attenuation in relatively coarse-grained rocks where olivine is or was deformed at relatively high stress by dislocation creep in Earth's upper mantle.  相似文献

Knowledge of SGS in plants is vital to understand the ecological and evolutionary dynamics of populations and to plan conservation strategies. Some of the major factors that can affect spatial genetic structure (SGS) in plants are the level of gene flow, spatial arrangement and life stages of individuals within populations. Applying six highly variable microsatellite markers, we investigated the effect of these factors on spatial genetic structure selecting two natural populations of sycamore maple, which is an insect-pollinated, autotetraploid and an indigenous hardwood species in Germany and in other central European countries. The two study populations had different shapes (“compact” and “elongated”) and tree densities. Significant SGS extended to ~180 m in the elongated population and to ~35 m in the compact population. Juvenile plants of the compact population showed significant SGS up to 40 m. Estimate of Sp statistic in high-density population was almost double of that in the population with low density. Gene dispersal distance in the low-density population was about 9 times higher than in the population with high density. The similar level of significant SGS in both adult and juvenile plants suggested minimal or no effect of life stages of individuals on SGS in the sycamore maple population. The data presented in this study can provide guidelines for seed collection and to establish populations for the conservation and management of genetic resources of the species.  相似文献

The objectives were to investigate (1) to which extent water-stable macro- and microaggregates sequester organic matter (OM) in a minimum tillage (MT) system compared to a conventional tillage (CT) system and (2) if the content of biochemically stabilized OM differs between both tillage systems, and (3) to study the temporal dynamics of the distribution of aggregate size classes and of storage of OM within aggregates in the field. Surface soils (0–5 cm) and subsoils (10–20 cm) were sampled after fallow (March 2007) and directly after tillage (November 2007) from a long-term experimental field near Göttingen, Germany. Macroaggregates (>0.25 mm) were in general less abundant after fallow than directly after tillage. In March, only 21% (CT) and 45% (MT) of Corg was stored within macroaggregates in the surface soil, whereas in November, the percentages increased to 58% and 73%, respectively. CT and MT soils of both depths were incubated as bulk soil (CTbulk, MTbulk) and with macroaggregates disrupted (<0.25 mm) (CTmd, MTmd) for 28 days at 22°C and water content of 50% of the maximum water holding capacity. For the MTbulk and MTmd surface soils, C mineralization was significantly higher compared to the CT soils. Incubation of md soils did not generally result in a significantly higher C mineralization compared to the respective bulk soils, except for the MTmd subsoil. Acid hydrolysis showed that the proportion of biochemically stabilized, nonhydrolysable, Corg to total Corg was lower in the MT than in the CT soils. Overall, the data indicate that the effect of physical stabilization of OM stored in the macroaggregates may not be a mechanism protecting very labile C with a turnover time of weeks, but that longer preservation likely occurs after macroaggregate transformation into microaggregates, and the surplus of OM found in the surface soil of MT does not only depend on the biochemically stabilized OM. Finally, our data suggest that the temporal variability of distribution of aggregate size classes in the field is large, but spatial and operator variability also contributed to the observed differences.  相似文献

The increasing demand for biomass for food, animal feed, fibre and bioenergy requires optimization of soil productivity, while at the same time, protecting other soil functions such as nutrient cycling and buffering, carbon storage, habitat for biological activity and water filter and storage. Therefore, one of the main challenges for sustainable agriculture is to produce high yields while maintaining all the other soil functions. Mechanistic simulation models are an essential tool to fully understand and predict the complex interactions between physical, biological and chemical processes of soils that generate those functions. We developed a soil model to simulate the impact of various agricultural management options and climate change on soil functions by integrating the relevant processes mechanistically and in a systemic way. As a special feature, we include the dynamics of soil structure induced by tillage and biological activity, which is especially relevant in arable soils. The model operates on a 1D soil profile consisting of a number of discrete layers with dynamic thickness. We demonstrate the model performance by simulating crop growth, root growth, nutrient and water uptake, nitrogen cycling, soil organic matter turnover, microbial activity, water distribution and soil structure dynamics in a long-term field experiment including different crops and different types and levels of fertilization. The model is able to capture essential features that are measured regularly including crop yield, soil organic carbon, and soil nitrogen. In this way, the plausibility of the implemented processes and their interactions is confirmed. Furthermore, we present the results of explorative simulations comparing scenarios with and without tillage events to analyse the effect of soil structure on soil functions. Since the model is process-based, we are confident that the model can also be used to predict quantities that have not been measured or to estimate the effect of management measures and climate states not yet been observed. The model thus has the potential to predict the site-specific impact of management decisions on soil functions, which is of great importance for the development of a sustainable agriculture that is currently also on the agenda of the ‘Green Deal’ at the European level.  相似文献

Background

Fertilization with organic waste compost can close the nutrient cycles between urban and rural environments. However, its effect on yield and soil fertility must be investigated.

Aim

This study investigated the long-term effect of compost on soil nutrient and potentially toxic elements (PTEs) concentration, nutrient budgets, and nitrogen (N) mineralization and efficiency.

Methods

After 21 years of annual compost application (100/400 kg N ha–1 year–1 [100BC/400BC]) alone and combined with mineral fertilization, soil was analyzed for pH, organic carbon (SOC), nutrient (total N and P, Nmin, extractable CAL-P, CAL-K, and Mg), and PTE (Cu, Ni, Zn) concentrations. Yields were recorded and nutrient/PTE budgets and apparent net mineralization (ANM, only 2019) were calculated.

Results

N efficiency was the highest in maize and for mineral fertilization. Compost application led to lower N efficiencies, but increased ANM, SOC, pH, and soil N, and surpluses of N, P, and all PTEs. Higher PTE concentrations were only found in 400BC for Cu. Nutrient budgets correlated with soil nutrient concentration. A surplus of 16.1 kg P ha–1 year–1 and 19.5 kg K ha–1 year–1 resulted in 1 mg kg–1 increase in CAL-P and CAL-K over 21 years.

Conclusion

Compost application supplies nutrients to crops with a minor risk of soil-accumulation of PTEs. However, the nutrient stoichiometry provided by compost does not match crop offtakes causing imbalances. Synchronization of compost N mineralization and plant N demand does not match and limits the yield effect. In winter wheat only 65–70% of N mineralization occurred during the growth period.  相似文献

Background

There is a great need for simple and inexpensive methods to quantify ammonia emissions in multi-plot field trials. However, methods that meet these criteria have to be thoroughly validated. In the calibrated passive sampling approach, acid traps placed in the center of quadratic plots absorb ammonia, enabling relative comparisons between plots. To quantify ammonia emissions, these acid trap samplings are scaled by means of a transfer coefficient (TC) obtained from simultaneous measurements with the dynamic tube method (DTM). However, dynamic tube measurements are also comparatively costly and time-consuming.

Aims

Our objective was to assess the best practice for using calibrated passive sampling in multi-plot field trials. One particular challenge in such experiments is to evaluate the influence of ammonia drift between plots.

Methods

In a series of eight multi-plot field trials, acid traps and DTM were used simultaneously on all plots to measure ammonia emissions caused by different slurry application techniques. Data obtained by both methods were correlated, and the influence of the ubiquitous ammonia background on both methods was evaluated by comparing net values, including the subtraction of the background with gross values (no background subtraction). Finally, we provide recommendations for calculating a TC for calibrating relative differences between plots, based on simultaneous acid trap and dynamic tube measurements on selected plots.

Results

Treatment mean values obtained by both methods correlated well. For most field trials, R2 values between 0.6 and 0.8 were obtained. Ammonia background concentrations affected both methods. Drift between plots contributed to the background for the acid traps, whereas the contamination of the chamber system might have caused the background for the DTM. Treatments with low emissions were comparatively more affected by that background.

Conclusion

For a robust application of calibrated passive sampling, we recommend calculating the TC based on a treatment with high ammonia emissions, reducing the relative influence of the ubiquitous ammonia background.  相似文献
