A blocking enzyme-linked immunosorbent assay (ELISA) has been adapted to detect specific antibodies in bovine sera to respiratory syncytial virus using a horseradish peroxidase-labeled monoclonal antibody to the fusion protein of the virus. This assay plus an indirect blocking ELISA and indirect ELISA were used to detect antibodies to the bovine respiratory syncytial virus (BRSV) in 159 field-origin bovine sera. Results of these assays were compared with serum antibody titers measured by the serum neutralization (SN) test. Over a 56-day period, the mean neutralization titers and the mean delta absorbance values for the blocking ELISA, on the same sera, showed similar declines. However, the calculated correlation coefficients between mean SN titer and mean absorbance value for the blocking ELISA of the individual sera ranged from -0.2 to -0.5 depending on the source of sera. Similar values were obtained whether using crude or purified viral antigen in the assays. Corresponding calculated correlation coefficients were generally higher for the indirect blocking ELISA or indirect ELISA than for the blocking ELISA. The blocking ELISA was between 70 and 64% as sensitive as the serum neutralization test with a specificity of 100 or 90% using the crude and purified viral antigen, respectively. The indirect blocking ELISA and indirect ELISA had similar calculated sensitivities and specificities. The blocking ELISA was faster to run than either of the other ELISA's or the neutralization test. Further, nonspecific background absorbance was obviated because the blocking ELISA detects antibodies to 1 specific viral protein, the fusion protein. Acute posterior paralysis in two weaner pigs was attributed to ischemic infarction of the lumbar spinal cord due to fibrocartilaginous emboli. These arterial and venous emboli were assumed to have originated from the nucleus pulposus of one or more intervertebral discs. There was no indication of an initiating factor in either instance, although some form of trauma may have been involved. Lesions were not grossly evident and were limited to one or two lumbar segments. Although it appears to be a rare occurrence, fibrocartilaginous embolism of the spinal cord should be considered in cases of acute paralysis in pigs particularly when gross lesions are absent. 相似文献

A total of five data sets obtained from four separate sheep flocks were used to evaluate the effects of subclinical maedi-visna virus infection (as determined by serological procedures) on productivity in ewes. In general, infection with maedi-visna virus had a detrimental effect on productivity. Infection resulted in a reduction in conception rates, with the odds of an infected ewe conceiving being only approximately 0.67 times those of a non-infected ewe. For ewes which did conceive, there was no detrimental effect of infection on the number of lambs born. The birth weights of lambs born to middle-aged (3–4 years) infected ewes were 3–6% lower than the weights of lambs born to non-infected ewes of the same age. There was very little apparent effect of infection in younger (1–2 years) and older (5 + years) ewes: some factors which may have biased these results are discussed. Lamb weights at 25–50 days of age appeared to be reduced by maedi-visna infection in the ewe but the results were not statistically significant. 相似文献

Ninety percent of foxes fed commercial ERA vaccine in a specially designed bait developed rabies serum neutralizing antibodies. The vaccine bait did not cause clinical signs of rabies when consumed by foxes, raccoons, skunks, dogs, cats, cattle and monkeys. When presented, in the laboratory, to wild rodents of the species Microtus, Mus musculus and Peromyscus, the vaccine baits caused vaccine-induced rabies only in Mus musculus. Laboratory mice of the CD-1 and CLL strain were susceptible to vaccine-induced rabies; however, studies showed that transmission of virus to other animals did not occur. These studies suggest that the vaccine bait described could be useful in a rabies control program in areas where foxes and wild dogs are the principal vectors. 相似文献

Air quality, respiratory disease, and growth rate were followed in four different farrowing and nursery systems. Ammonia levels varied with ambient air temperature, but were within normally accepted levels (25 ppm). These levels of ammonia did not appear to affect the health or performance of the pigs raised in these units. Hydrogen sulfide levels were consistently low. Counts of bacterial colony forming particles (BCFP) varied and the organisms identified were predominantly micrococci. Bordetella bronchiseptica was isolated from nasal cavities of pigs from 3 out of 4 farms. Three of the farms did not have evidence of atrophic rhinitis; pigs farrowed in the last quarter of the test year on one farm from which B. bronchiseptica was isolated developed lesions of atrophic rhinitis. The B. bronchiseptica isolates from the 3 farms were virulent for gnotobiotic piglets. Groups of pigs for slaughter inspection from one farm had lungs with 11–28% pneumonic lesions; these lesions were not typical of mycoplasmal pneumonia. 相似文献

OBJECTIVE: To determine whether a combination viral vaccine containing modified-live bovine herpesvirus-1 (BHV-1) would protect calves from infection with a recent field isolate of BHV-1. DESIGN: Randomized controlled trial. ANIMALS: Sixty 4- to 6-month-old beef calves. PROCEDURE: Calves were inoculated with a placebo 42 and 20 days prior to challenge (group 1; n = 10) or with the combination vaccine 42 and 20 days prior to challenge (group 2; 10), 146 and 126 days prior to challenge (group 3; 10), 117 and 96 days prior to challenge (group 4; 10), 86 and 65 days prior to challenge (group 5; 10), or 126 days prior to challenge (group 6; 10). All calves were challenged with BHV-1 via aerosol. Clinical signs, immune responses, and nasal shedding of virus were monitored for 14 days after challenge. RESULTS: Vaccination elicited increases in BHV-1-specific IgG antibody titers. Challenge with BHV-1 resulted in mild respiratory tract disease in all groups, but vaccinated calves had less severe signs of clinical disease. Extent and duration of nasal BHV-1 shedding following challenge was significantly lower in vaccinated calves than in control calves. In calves that received 2 doses of the vaccine, the degree of protection varied with the interval between the last vaccination and challenge, as evidenced by increases in risk of clinical signs and extent and duration of viral shedding. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that this combination vaccine provided protection from infection with virulent BHV-1 and significantly reduced nasal shedding of the virus for at least 126 days after vaccination. 相似文献

Irradiation of standard broiler carcasses within 24 hr. of slaughter with a dose of 0.15 Mrad of gamma radiation was sufficient to give a shelf life of more than six days at 5° C. followed by one day at 15° C. The carcasses were treated and stored in air and remained unchanged in appearance and odour. After cooking, treated and stored carcasses were indistinguishable, when tasted in blind comparison tests, from un‐irradiated material held at —20° C. for the same time. The storage conditions chosen were taken to represent the upper limits of commercial and domestic handling during distribution of unfrozen broiler chicken.
