This study investigated the sources of inoculum of Botyosphaeriaceae species in three commercial grapevine nurseries in New Zealand. Botryosphaeriaceae propagules were detected on the surfaces of 33 to 100?% of canes and 33?% of dead grapevine materials using microscopy, plating assays and PCR with Botryosphaeriaceae multi-species primers (Bot100F/Bot472R). Isolations also showed that 15 to 68?% of the canes were internally infected by Botryosphaeriaceae species, with Neofusicoccum luteum and N. parvum being the most predominant species. Incidence varied between nurseries, with Nursery 5 having highest N. luteum incidence (53?%) and Nursery 3 having highest N. parvum incidence (88?%). Botryosphaeriaceae DNA was detected in rain-water run-off from the mother-vine canopy, but not in soil samples collected around infected mother-vines. However, samples from the nursery propagation stages had no visible or viable Botryosphaeriaceae propagules although PCR using multi-species primers detected Botryosphaeriaceae DNA from wash and hydration tanks, on grafting tools and in callusing media from the different nurseries. The single-stranded conformational polymorphism (SSCP) was able to identify N. parvum, N. luteum and Diplodia mutila as the most common species present in the nursery system. Overall results indicated that the canes grown in mother-vine blocks were the most likely source of inoculum for infection of young grafted plants by Botryosphaeriaceae species.  相似文献

Unique bands were identified in single isolates of Neofusicoccum parvum and Neofusicoccum luteum using universally primed polymerase chain reaction (UP‐PCR) analysis of isolates obtained from grapevines and non‐grapevine hosts in New Zealand, Australia, South Africa and the USA. Primers were designed to amplify a 1550 bp portion of the 1573 bp marker band from N. parvum isolate B2141 and a 510 bp portion of the 524 bp marker band from N. luteum isolate G51a2. A PCR‐RFLP assay was developed to distinguish the N. parvum isolate B2141 from other N. parvum isolates, based on a polymorphism found in the marker band using the TaqI restriction endonuclease. For N. luteum isolate G51a2, the designed primers were specific at an annealing temperature of 63°C in the PCR. The sensitivity threshold of the N. parvum and N. luteum isolate‐specific markers was 50 pg and 5 pg, respectively, when used in standard PCR with purified genomic DNA. The sensitivity of the N. parvum isolate‐specific marker was increased to 0·5 pg by nested PCR. The specificity test of both isolate‐specific markers with six other Botryosphaeriaceae spp. showed that they were specific to their respective species and isolates. Both markers were able to detect the conidia of N. parvum and N. luteum marker isolates in rainwater samples collected at different distances from an inoculation point in the vineyard. The results showed that rain splash could disperse the conidia of both of these species up to 2 m from the inoculum point in a single rainfall event.  相似文献

Disseminated histoplasmosis and disseminated intravascular coagulopathy were diagnosed in a 7 mo old, female spayed mixed-breed dog. The dog improved transiently with supportive care, but deteriorated shortly after initiation of antifungal therapy. The dog was subsequently euthanized. At necropsy, marked granulomatous vasculitis was identified in all affected organs. The tunicae and laminae of the arteries and arterioles were obscured by epithelioid macrophages and multinucleated giant cells admixed with necrotic material. Intracytoplasmic yeast were present within some of these macrophages. To the authors' knowledge, this is the first reported case of granulomatous vasculitis associated with Histoplasma capsulatum in a dog.  相似文献

Interest in magnetic resonance (MR) imaging as a major diagnostic tool in veterinary clinical medicine is increasing. Most MR studies are performed with the use of contrast enhancement via intravenous injection of paramagnetic gadolinium-containing contrast media. A vast number of publications are available regarding the use of contrast media in humans. The purpose of this paper is to assist practicing veterinary radiologists in understanding mechanisms of MR contrast enhancement. This paper reviews certain aspects of MR contrast enhancement, including physical, chemical and biologic characteristics of most common MR contrast media targeted primarily at the central nervous system (CNS). Authors also describe processes that explain changes in signal intensity on the MR images.  相似文献

The stifle joints of eleven military working dogs were evaluated using conventional magnetic resonance (MR) imaging and MR arthrography. A protocol optimizing MR imaging of the canine stifle joint is discussed, as well as potential uses for administration of intra-articular gadolinium. The technique for performing MR arthrography is described, and post-contrast image findings are reviewed. MR arthrography was performed by using an intra-articular injection of diluted gadolinium. Consistently good quality images were obtained, and no complications were clinically detected following MR arthrography. Cranial cruciate ligament abnormalities were seen in six dogs, meniscal abnormalities were visualized in nine menisci, and synovitis and medial ligament strain were seen in eight dogs. Surgical and post-mortem confirmation of these findings is discussed in seven dogs. Although MR arthrography adds an invasive procedure to conventional MR imaging, it can provide useful information on pathologic changes in the canine stifle joint.  相似文献

The magnetic resonance imaging appearance of the feline middle ear is described in three healthy cats and in five cats with middle ear disease. Owing to the good spatial resolution, multiplanar slice orientation as well as display high contrast resolution of soft tissue, in particular fluids, MR imaging was helpful prior to surgery. It is superior to radiography which failed to allow identification of the abnormality in two of our five cats. MR imaging for middle ear disease should include dorsal and transverse plane images using T1- and T2-weighted sequences. In the presence of a mass within the bulla or the external ear canal application of contrast medium is helpful.  相似文献

A porous-coated modular total hip system was developed for uncemented implantation in dogs. The operative technique was developed in cadaver bones and live animals. One hundred uncemented total hip arthroplasties were performed in 92 dogs. Results were successful in 98% of the joints during follow-ups of 3 months or more. Complications included three luxations of the prosthetic joint, two fissure fractures of the femoral cortex, and one complete displacement of the acetabular component from its bony bed. Four complications were resolved successfully.  相似文献

Volumetrically correct, three-dimensional color images of the sea otter, ( Enhydra lutris ), were generated from computed tomographic (CT) data using a Sun 3/160 workstation and a Pixar Imaging Computer. These images could be displayed as static images (1024×768) or rotated about any axis in real-time by displaying sequential rotation sequences generated by the Pixar Imaging Computer. The images provided realistic depth of field through overshadowing and differential opacification. This was obtained through an opacification algorithm which traced rays from a selected viewing plane through colored gel volumes, and calculated absorbance percentages as a function of the transparency of the gels penetrated. Reconstructions of this type serve as alternative anatomy tutorials for species otherwise unavailable for routine dissection. They also permit topographical studies without interfering with populations of endangered animals.  相似文献
