A 16.5 liter spherical ion chamber was constructed of conducting muscle-equivalent plastic and filled to 760 mm-Hg with a muscle-equivalent gas. The use of the chamber for measurements of natural environmental backgrounds was made quite feasible with the aid of the recently developed Shonka vibrating quartz fiber electrometer. This instrument is routinely operable at the extremely high sensitivity of better than 5000 divisions per volt. This system, therefore, has made possible reproducible measurements of absorbed dose-rates of fractions of a micro-rad per hour without any need for the usual corrections for wall-effect, stopping power, and so forth.  相似文献

Glycogen depletion patterns in horses performing maximal exercise

Muscle biopsy samples were collected from the left middle gluteal muscle of horses participating in competitive barrier trials. Twelve horses were biopsied the day before and within 30 minutes of completion of an 800 m barrier trial. A further six horses were sampled the day before, and within 30 minutes of, completion of a 1200 m barrier trial. Serial muscle sections were examined histochemically for myosin adenosine triphosphatase activity after acid preincubation, to demonstrate type I, IIA and IIB fibres. The glycogen content in the individual fibres was assessed using the periodic acid Schiff (PAS) reaction. Total glycogen in muscle was measured fluorimetrically after hydrolysis to glucose. Significant decreases in total muscle glycogen levels of 167.1 +/- 22.2 and 158.1 +/- 23.2 mmoles glucose units kg-1 (dry weight) (mean +/- standard error) occurred from pre- to post trial samples for horses in both 800 and 1200 m trials respectively. There were no significant differences between the decreases in total muscle glycogen from horses in either 800 m or 1200 m trials. In both 800 m and 1200 m trials there was a significant decrease in the percentage of type IIA and IIB fibres classified as having high PAS staining intensity, and a significant increase in the percentage of fibres classified as having medium staining intensity when samples taken after the trial were compared to those taken before the trial.  相似文献

Attempts were made to immunise lambs by the intramuscular injection of either lyophilised or concentrated metabolites produced by Ostertagia circumcincta when grown in vitro, or macerated fourth and fifth stage worms. The lyophilised metabolites did not stimulate protection against the establishment of worms in nine-months-old lambs but the macerated worms and concentrated metabolites elicited protection in six-months-old and three-months-old lambs respectively. The worm burdens and worm egg counts of the immunised lambs were less than those of control lambs.  相似文献

A 4.5-year old, male African spur-thighed tortoise (Geochelone sulcata) was presented to the University of Florida Veterinary Teaching Hospital with a 2-week history of lethargy, anorexia, constipation, dyspnea, and coughing up fluid or vomiting. Laboratory results included an inflammatory leukogram and a marked increase in plasma uric acid concentration. Synovial fluid from multiple joints was thick, chalky white, and opaque, with a grainy consistency. Microscopically, the fluid contained numerous brown, needle-like crystals consistent with urates (gout). Gross necropsy findings and histopathology confirmed a diagnosis of systemic gout, with urate deposition, gout tophi, and underlying necrosis in multiple organs, including kidneys, lung, and liver. Dehydration with concurrent renal insufficiency may have impaired urate excretion and led to a build-up of urates in the blood and tissues of this tortoise. A high protein diet also may have contributed to the development of gout. Cytologic evaluation of synovial fluid can be used as a quick and definitive tool to diagnose gout in tortoises.  相似文献
