Since the first successful nuclear transfer (NT) experiments were carried out, various somatic cell types have been used as donor cells for production of cloned animals. In most experiments, fibroblasts are used since they only need to be isolated and cultivated. Recently, some researchers have shown that different cell cultures from different sources possess different capacities to support preimplantation development of NT embryos. The blastocyst rates obtained in our previous studies varied and were as high as 45% in relation to the number of reconstructed embryos. This led us to question whether the origin and culture conditions of the defined male and female fibroblast lines could be responsible for the differences in developmental potency. Taking all our results into consideration, we conclude that different fibroblast lines recovered from the same tissue and cultivated under equal culture conditions could produce dramatically different blastocyst rates. The influence of cell line itself is higher than the influence of passage number. The observed effects of cell cycle stage, chromosomal aberrations, and diminished vitality are important but not sufficient to discriminate well-qualified nuclear donor cells. We speculate that some epigenetically regulated deviations in the gene expression program are responsible for these phenomena. Explanation of the underlying mechanisms should contribute to better understanding of epigenetic reprogramming and may ultimately assist reprogramming in the laboratory.  相似文献

OBJECTIVE: To determine the effects of three anesthetic regimens on serum calcium and parathyroid hormone (PTH) in mice. STUDY DESIGN: Prospective experimental study. ANIMALS: Twenty-seven, 12-week-old, FVB mice. METHODS: Mice were anesthetized with either isoflurane, a combination of ketamine plus xylazine (100 and 10 mg kg(-1), respectively) or ketamine alone (100 mg kg(-1)). Blood was collected before and after the induction of anesthesia. Serum calcium and PTH concentrations were measured and compared. RESULTS: Isoflurane or ketamine did not significantly alter either serum calcium or serum PTH concentrations. A combination of ketamine plus xylazine significantly increased serum PTH, but not serum calcium concentrations. CONCLUSIONS: A combination of ketamine and xylazine should be avoided prior to collection of blood for measurement of serum PTH concentrations. Isoflurane is suitable for this purpose.  相似文献

ABSTRACT

Soil acidity is one of the main limitations for optimal use of land resources for better crop production. And, long-term fertilization experiments found to be helpful in increasing the nutrient supply in these acidic soils. Keeping this in view, a field experiment on rice was carried out by applying biofertilizers and enriched compost in an acidic Inceptisol of Assam over 10 successive years (2006–15) to examine its effects on nutrient availability and soil enzymatic activity. This experiment had five treatments viz. absolute control (T1), 100% recommended doses (RD) of inorganic NPK (T2), 50% RD of inorganic NP + 100% K +biofertilizers (T3), 50% RD of inorganic NP + 100% K +1 tonne enriched compost ha?1 (T4) and 25% RD of inorganic NP + 100% K +2 tonnes enriched compost ha?1 (T5) under randomized block design with four replications. After completing 10 years of experiment, it was observed that integrated use of enriched compost and biofertilizers with reduced doses of inorganic fertilizers enhanced the soil enzymatic activity as well as nutrient availability in rice grown in acidic clay loam soils of Assam. Application of biofertilizers and enriched compost had positive impact on plant accessible nitrogen, phosphorus and potassium in soil as compared to inorganic fertilizers. Also soil organic matter content increased considerably by these treatment. Integrated nutrient management practice in rice had also significantly enhanced the dehydrogenase, fluorescein diacetate and phospho-monoesterase activity in soil.  相似文献

Improvement in litter traits is the key to profitable pig farming that directly enhances the economic standing of the farmers in developing countries. The present study aimed to explore oestrogen receptor (ESR), epidermal growth factor (EGF), follicle-stimulating hormone beta subunit (FSHβ), prolactin receptor (PRLR) and retinol-binding protein 4 (RBP4) genes as possible candidate genetic markers for litter traits in indigenous pigs of India. The breeds included in the study were Ghungroo, Mali, Niang Megha and Tenyi Vo, and the reproductive traits considered were litter size at birth (LSB), number born alive (NBA), litter weight at birth (LWB), litter size at weaning (LSW) and litter weight at weaning (LWW) at their first parity. PCR-RFLP and primer-based mutation detection methods were used to identify polymorphism, and associations between the genotypes and the traits were analysed using a general linear model. The Ghungroo pigs recorded the best litter performances among the breeds (p < .05, LWB p < .01). Different alleles and genotypes of the genes under study were detected. Short interspersed nuclear element (SINE) −/− genotype of FSHβ revealed significantly higher litter traits (p < .05, LSB p < .01). The LWW was also found to be significantly influenced by ESR BB and AB, EGF AB and BB, and PRLR CC genotypes (p < .05). Although we did not find statistically significant and consistently superior litter traits with respect to different genotypes of other studied genes than genotype SINE −/− of the FSHβ, PRLR CC genotype demonstrated superior performances for all the litter traits. Our study revealed the FSHβ as a potential candidate genetic marker for litter traits in indigenous pig breeds of India.  相似文献

Soil samples were collected from Panchamarhi dry deciduous forest in Satpuda Biosphere Reserve, India to determine the effect of hill slopes and altitude on the population size of methanotrophic bacteria. Population size, in range of 4×105-3.6×107 g−1 dry soil, was negatively correlated with altitude and increased exponentially (r2=0.97, P<0.001) at steep slope (60°) while logarithmically (r2=0.97, P<0.001) at low slope (45°). Soil organic C, total N, and soil moisture increased while C/N ratio and temperature decreased down the hill slope. The results indicated that nutritional status of the soil across the slopes determines the methanotrophic bacterial population size.  相似文献

Outbreak of the emerging infectious disease sudden oak death continues to threaten California and Oregon forests following introduction of the exotic plant pathogen Phytophthora ramorum. Identifying areas at risk and forecasting changes in forest carbon following disease outbreak requires an understanding of the geographical distribution of host populations, which is unknown. In this study, we quantify and map the population density and carbon contents of five key host species for P. ramorum in California and Oregon, including four hosts killed by the pathogen (Notholithocarpus densiflorus, Quercus agrifolia, Quercus kelloggii and Quercus chrysolepis) and the foliar host Umbellularia californica which supports high sporulation rates. We integrate multiple sources of vegetation data, assembled from sparsely distributed (regional-scale) forest inventory and analysis (FIA) plots and more densely distributed (landscape-scale) plots for monitoring sudden oak death, and develop spatial prediction models based on correlation with environmental variables and spatial dependencies in host abundance. We estimate that 1.8 billion N. densiflorus trees (68 Tg C) and 2.6 billion Quercus host trees (227 Tg C) occur across 3.9 and 17.7 million ha of their respective habitat. A total of 436 million U.californica trees (14 Tg C) occur across 4.2 million ha which frequently overlap with Quercus and N. densiflorus host populations. Combination of landscape-scale data with FIA data resulted in more accurate estimation of host populations and their carbon contents. Forests of northern California and southwest Oregon have the highest concentration of the most susceptible hosts along with climatic conditions that favor pathogen spread. This study represents the first spatially-explicit estimate of P. ramorum host populations and their carbon contents which exceed previously published estimates. Our results will inform landscape- to regional-scale models of disease dynamics and guide management decisions regarding ecosystem impacts including risk of C release following widespread tree mortality.  相似文献

The outcome of different extraction procedures (microwave, ultrasound, Soxhlet, and maceration) on the antioxidant activity of seeds, leaves, pulp, and fruits of Hippophae rhamnoides (sea buckthorn or SBT) was investigated by two different bioassays: 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays. The SBT extracts were found to possess strong antioxidant activity measured in terms of TEAC (2.03-182.13 and 6.97-282.75 mg/g) with ABTS and DPPH assays, respectively. In general, the antioxidant capacity of microwave-assisted extracts was found to be significantly higher than those obtained by ultrasound-assisted extraction (UAE) and maceration while being slightly higher than Soxhlet extracts. Further, microwave extracts of seeds were found to possess maximum antioxidant capacity followed by leaves, fruits, and pulp. Also, the chemical composition of extracts, studied in terms of the total phenolic content, was found to be in the range of 1.9-23.5 mg/g Gallic acid equivalent (GAE), which indicates a strong correlation between antioxidant activity and phenolic content present in the SBT. In addition, some of its bioactive phenolic constituents, such as rutin ( 1), quercetin-3- O-galactoside ( 2), quercetin ( 3), myricetin ( 4), kaempferol ( 5), and isorhamnetin ( 6), were also quantified in different extracts by reverse-phase high-performance liquid chromatography (RP-HPLC).  相似文献

Recent findings on the effect of aluminium (Al) on the functioning of legumes and their associated microsymbionts are reviewed here. Al represents 7% of solid matter in the Earth’s crust and is an important abiotic factor that alters microbial and plant functioning at very early stages. The trivalent Al (Al3+) dominates at pH <?5 in soils and becomes a constraint to legume productivity through its lethal effect on rhizobia, the host plant and their interaction. Al3+ has lethal effects on many aspects of the rhizobia/legume symbiosis, which include a decrease in root elongation and root hair formation, lowered soil rhizobial population, and suppression of nitrogen metabolism involving nitrate reduction, nitrite reduction, nitrogenase activity and the functioning of uptake of hydrogenases (Hup), ultimately impairing the N2 fixation process. At the molecular level, Al is known to suppress the expression of nodulation genes in symbiotic rhizobia, as well as the induction of genes for the formation of hexokinase, phosphodiesterase, phosphooxidase and acid/alkaline phosphatase. Al toxicity can also induce the accumulation of reactive oxygen species and callose, in addition to lipoperoxidation in the legume root elongation zone. Al tolerance in plants can be achieved through over-expression of citrate synthase gene in roots and/or the synthesis and release of organic acids that reverse Al-induced changes in proteins, as well as metabolic regulation by plant-secreted microRNAs. In contrast, Al tolerance in symbiotic rhizobia is attained via the production of exopolysaccharides, the synthesis of siderophores that reduce Al uptake, induction of efflux pumps resistant to heavy metals and the expression of metal-inducible (dmeRF) gene clusters in symbiotic Rhizobiaceae. In soils, Al toxicity is usually ameliorated through liming, organic matter supply and use of Al-tolerant species. Our current understanding of crop productivity in high Al soils suggests that a much greater future accumulation of Al is likely to occur in agricultural soils globally if crop irrigation is increased under a changing climate.  相似文献
