Banana and plantain are among the most important food crops in developing countries but production is threatened by increasing virulent forms of Fusarium oxysporum f. sp. cubense. Chemical control is not economically effective and,therefore, breeding programs are necessary. Traditional field studies of new genotype resistance to this disease are time-consuming and destructive. Therefore,we developed a rapid and non-destructive procedure to differentiate field-grown banana resistant from susceptible clones. This procedure implicates application of culture filtrates of Fusarium oxysporum f. sp. cubense race 1 onto banana leaves. The relationship between duration of the fungal in vitro incubation, and the fungal culture fresh mass, the culture filtrate absorbency, and the Gross Michel (susceptible cultivar)leaf lesion area (after application of the culture filtrate) were similar and at 24day-incubation the highest values of the recorded indicators were observed. A comparison between Gross Michel and FHIA-01(resistant) was also performed. The most relevant differences between cultivars were observed at 48 hours after application of the culture filtrate, and in the middle-aged leaves. The position of the culture filtrate application in the leaf limb (distal, middle, proximal) was not determinant. A wider comparison among banana cultivars confirmed previous results informed by other researchers using different systems to study this plant-fungus interaction. Such a confirmation validates the effectiveness of the procedure described here to select rapid and non-destructively banana resistance to this disease at field level. Among the main causes of poor yield in common beans are fungal, viral and bacterial diseases. Common bacterial blight, caused by Xanthomonas axonopodis pv. phaseoli (Xap), is one of the major bacterial diseases leading to significant losses in Brazil. Chemical control is ineffective, therefore, the use of resistant varieties becomes an interesting alternative. The objective of the present work was to evaluate disease resistance under natural infection of the pathogen in 109 recombinant inbred lines (F7) of P. vulgaris originated from the cross HAB-52 (susceptible — snapbean) × BAC-6 (resistant — common bean) in two different environments, as well as to calculate genetic parameters to assist in the selection of promising materials to be used in the CBB resistance breeding program. The data of the genetic parameters were compared to those calculated for the F3 generation originated from the same cross. The heritability results for DI (disease index) and VI (variation index) in F3 were 26.85% and 0.26, respectively, whereas in F7 they were 91.77% and 1.36, respectively. These results demonstrate a potential to be explored for this advanced population, that in the future, along with other pathogen variability studies and tests in other environments, may provide more information regarding a more precise evaluation of promising genotypes to be used in common bean breeding programs aiming to obtain CBB resistant varieties. We have previously obtained several lines of tobacco transformed with a trehalose-6-phosphate synthase gene of plant origin (Arabidopsis thaliana), involved in the first step of the biosynthesis of trehalose, a known osmoprotectant. Two showed distinct intensity of expression: high (B5H) and low (B1F). Such lines were analyzed for trehalose-6-phosphate content and the obtained results demonstrated to be in accordance with the expression results. In order to study the responses of photosynthesis to water deficit of transgenic lines in comparison to wild type (WT), three experiments were performed under different conditions: (1) Relative water (2) Leaf gas exchange (3) Modulated Chlorophyll a Fluorescence. Different responses in RWC of plant lines to water withdrawal were detected, with transgenic line B5H indicating less water loss after the water withdrawal period. Similar responses to water deficit regarding the leaf gas exchanges were recorded for the three lines. When subjected to water deficit stress situations, higher F v/F m, ΦPSII and qP were detected for the transgenic lines. Under a SWC of 20% where higher values for such parameters were detected with special relevance for the B5H line, indicating a possible higher ability to withstand severe drought stress and to resist to prolonged periods without water than the B1F and WT lines.  相似文献

Correct knowledge of disturbance ecology is essential for understanding the characteristic behavior of forest ecosystems and for guiding appropriate management strategies. However, the role of natural disturbances in shaping European mountain forest ecosystems has not been adequately studied, possibly because of the perception that the development of most European forests is primarily shaped by human influences and/or fine-scale gap-phase dynamics.In the present study, we investigate the long-term disturbance history of old protected forest dominated by Norway spruce in the Parangalitsa Reserve, Bulgaria. We used aerial photo interpretation and dendroecological methods to reconstruct the history of wind, insect, and fire disturbances across a topographically complex landscape. Over the past 150 years wind has been the most important disturbance agent in this ecosystem and at least 18% of the forested area shows evidence of high-severity blowdowns. Windthrow patches ranged in size from <1 ha to >10 ha (minimum 0.11 ha, mean 0.16 ha, maximum 10 ha). Although small disturbances were much more frequent, few larger blowdowns accounted for most of the disturbed area. Pure coniferous and single-cohort coniferous forest patches were more affected by blowdowns than mixed coniferous-deciduous and multi-cohort coniferous forest patches. Although bark beetle (Ips typhographus) populations were large enough to cause mortality of some live trees, the populations did not grow to epidemic proportions during recent decades. Fire disturbance was of limited importance in the last 200 years and only two patches (4% of the study area) showed evidence of fire.The present research indicates that wind disturbances have been characteristic of these ecosystems at least over the past decades to centuries. Thus, blowdowns appear integral to the normal function and structure of the Picea-dominated mountain forests in the region and such events, in and of themselves, do not represent unhealthy forest conditions or environmental emergencies. Management strategies that aim to maintain these ecosystems within a natural range of variation should incorporate wind disturbances into the management strategy. The frequency and magnitude of future wind disturbances may be considered within the historical framework described in the current study to assess potential effects of climate change on altered disturbance regimes.  相似文献

Coupling media are necessary to ensure that transducers bond to wood specimens to minimize coupling losses and improve the accuracy of ultrasonic measurements. There are several types of coupling media available, and the optimal choice is not known. In this work, we analyzed the results of ultrasonic wave attenuation for 0.1-MHz longitudinal and transverse transducers with six different materials as coupling media in nine species of Brazilian wood with densities in the range 700–1170 kg/m3. Tests were performed using constant pressure on the transducer and with wave propagation in the longitudinal direction. For transverse transducers, the polarization was in the radial and tangential directions. The results were analyzed statistically and showed that, for attenuation in both longitudinal and transverse waves, the material used for coupling had significant effects, whereas the wood species had no effect. For longitudinal waves, the statistical evaluation showed that the coupling material performance was strongly dependent on the species of wood, but it was not possible to observe any tendency of behavior associated with specific anatomical properties.  相似文献

Thirty Portuguese and eight foreign olive (Olea europaea L.) cultivars were screened using Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR) markers. Twenty RAPD primers amplified 301 reproducible bands of which 262 were polymorphic; and 17 ISSR primers amplified 204 bands of which 180 were polymorphic. The percentage of polymorphic bands detected by ISSR and RAPD was similar (88 and 87%, respectively). The genetic variability observed was similar in the Portuguese and foreign olive cultivars. Seven ISSR and 12 RAPD primers were able to distinguish individually all 38 olive cultivars. Twenty specific molecular markers are now available to be converted into Sequence Characterised Amplified Region (SCAR) markers. Relationships among Portuguese and foreign cultivars is discussed.  相似文献

The genetic structure of Azospirillum amazonense populations isolated from the rhizosphere soil and washed and surface-sterilised roots of rice, maize and sorghum plants, cropped simultaneously in two different soils (clay loam and sandy loam) was characterised. Genetic diversity was measured by restriction fragment length polymorphism of the amplified 16S–23S rDNA intergenic spacer region (RISA-RFLP) and cluster analysis. Four genetically distinct clusters of isolates were observed with 78% similarity, suggesting that the A. amazonense population was heterogeneous at the strain level regardless of the soil type or host plant. Analysis of molecular variance (AMOVA) demonstrated that the host plant had a highly significant selective effect on the genetic structure of this species, especially on those isolates intimately associated with them, but also to a lesser extent on isolates from the rhizosphere and washed roots. The soil type also had a highly significant selective effect on A. amazonense genetic diversity, especially for those isolates from the rhizosphere soil. The selective effect of the soil type combined with that of the host plant suggests that environmental factors, such as soil texture and composition of exudates provided by C3 or C4 plants, play major roles in the overall genetic structure of A. amazonense populations associated with these cereals.  相似文献
