To investigate maturation of spermatia of Botryotinia fuckeliana, we examined micro-structural changes in spermatia in liquid culture. Three to four spherical lipid bodies were seen in the center of the cell at the start of culturing. After 3–7 days, maturing spermatia had developed large mitochondria along with extensively developed granular endoplasmic reticulum. In contrast, the lipid bodies decreased not only in number but also in size over time. The results obtained here suggest that the maturation of spermatia involves active cell metabolism using the energy supply from lipid bodies.  相似文献

An efficient incubation and inoculation system for white rust was established using plantlets of chrysanthemum growing in vitro. The internal conditions of a culture vessel (plant box) were maintained at a humidity of 90‐100% and an optimum temperature of 20‐25°C, which are suitable conditions to germinate teliospores and basidiospores of the pathogen. Telia were maintained continuously on plants in the plant box and were used as an inoculum for infection experiments throughout the year, allowing differences in susceptibility to white rust among chrysanthemum cultivars to be detected. Susceptibility to white rust in the plant‐box evaluation showed a good correlation with the rating of sporulation on plants grown in a greenhouse. The method described here is a simple, space‐saving inoculation system to evaluate the susceptibility of chrysanthemums to white rust.  相似文献

The interaction between fractalkine/CX(3)CL1 and its receptor CX(3)CR1 has been reported to play an important role in various human inflammatory diseases, including inflammatory bowel disease (IBD) mediated by lymphocyte chemoattraction. The objective of this study was to investigate the role of fractalkine and CX(3)CR1 in lymphocyte migration in canine IBD. IBD was diagnosed in 34 dogs, and 19 healthy beagles were used as normal controls. We quantified intestinal mRNA and protein expression of fractalkine and CX(3)CR1 by real-time RT-PCR and ELISA, respectively, and examined the localization of fractalkine in canine intestine by immunohistochemistry. The expression of CX(3)CR1 and surface antigens on peripheral blood mononuclear cells (PBMCs) and intraepithelial lymphocytes (IELs) was analyzed by flow cytometry. Intestinal fractalkine and CX(3)CR1 mRNA was significantly up-regulated in IBD dogs compared with the healthy control dogs. In addition, fractalkine expression on intestinal epithelial cells was significantly increased in the intestinal mucosa of IBD dogs compared with the healthy dogs. CX(3)CR1(+) PBMCs were significantly elevated in IBD dogs and positively correlated with the histopathological severity of IELs infiltration. These CX(3)CR1(+) PBMCs predominantly expressed markers for cytotoxic T cells. Almost all IELs expressed CD3, and the majority of cells expressed CD8 rather than CD4, which was analogous to the CX(3)CR1(+) PBMCs. These results suggest that the fractalkine-CX(3)CR1 interaction may contribute to the pathogenesis of canine IBD through migration of IELs.  相似文献

A heterophilic Hanganutziu-Deicher (HD) antigen is present in many animal sera except human and chicken sera. To visualize the antigenic molecules, nine species animal and human sera were analyzed by SDS-PAGE, followed by Western blotting and immunostaining with avain anti-N-glycolylneuraminyl-lactosyl ceramide antibody which recognizes the terminal N-glycolylneuraminic acid moiety of glycoconjugates as an epitope of the HD antigen. Several HD antigen-active glycoprotein bands were detected in the sera of fetal calf, calf, horse, goat, monkey, rabbit, guinea pig, rat and mouse, except for human serum. The HD antigenic proteins showed heterogeneities in their molecular weights and were not identical with any major band visualized with silver-staining, indicating that they are minor components of serum proteins in each animal. Neuraminidase treatment destroyed the antigenicity of all proteins, confirming that N-glycolylneuraminic acid (NeuGc) at the non-reducing terminal of carbohydrate chains is the antigenic epitope in serum glycoprotein molecules as already confirmed in glycosphingolipid (GSL) antigens. The finding of HD-antigenic glycoproteins in animal sera suggests that they also stimulate HD antibody production in patients who received animal antiserum for therapeutic aim.  相似文献

This study aimed to reveal the effects of direct exposure to cold weather under grazing in winter (GW) on the health of Japanese Black beef cattle, as assessed using physiological, immunological, and behavioral parameters. Ten Japanese Black beef cattle (328 ± 45 kg, 7.6 ± 3.4 years of age) were used in this experiment. In winter, five of the 10 cattle grazed for 2 months in a 1.8 ha pasture (GW), and the remaining cattle were fed under confined conditions with tethering (control [CT]). The two groups were fed similar feed during the experiment, except for the grazing forage. Blood samples were collected approximately every 2 weeks. The numbers of neutrophils and monocytes and antioxidant enzyme activity were higher in the GW group than in the CT group (p < 0.05). The proportions of CD4‐single‐positive cells were lower in GW cattle than in CT cattle (p = 0.06). This study showed that direct exposure of beef cattle to cold weather under GW enhanced the levels of circulating neutrophils and monocytes and contributed to the kinetic homeostasis of lymphocytes but also activated antioxidant enzymes due to an increase in oxidative stress.  相似文献

Metabolism experiments with rats showed that significant isotope effects (kHkD = 2.4 to 3.5) were associated with the in vivo formation of dichloro and trichlorophenylmercapturic acids from a 1:1 mixture of normal and hexadeuterated lindane. This is evidence that rate-determining dehydrogenation and dehydrochlorination, both of which proceed with significant isotope effects, are essential in the pathway of dichloro- and trichlorophenylmercapturic acid formation from lindane. No significant primary isotope effects were associated (kHkD = 1.31 ± 0.17) with the formation of monochlorophenylmercapturic acid. This suggests that the 1,2-dechlorination to tetrachlorocyclohexene followed by glutathione conjugation is the probable pathway that produces this metabolite from lindane.  相似文献

Fatty acid composition is an important indicator of beef quality. The objective of this study was to search the potential candidate region for fatty acid composition. We performed pool‐based genome‐wide association studies (GWAS) for oleic acid percentage (C18:1) in a Japanese Black cattle population from the Hyogo prefecture. GWAS analysis revealed two novel candidate regions on BTA9 and BTA14. The most significant single nucleotide polymorphisms (SNPs) in each region were genotyped in a population (n = 899) to verify their effect on C18:1. Statistical analysis revealed that both SNPs were significantly associated with C18:1 (p = .0080 and .0003), validating the quantitative trait loci (QTLs) detected in GWAS. We subsequently selected VNN1 and LYPLA1 genes as candidate genes from each region on BTA9 and BTA14, respectively. We sequenced full‐length coding sequence (CDS) of these genes in eight individuals and identified a nonsynonymous SNP T66M on VNN1 gene as a putative candidate polymorphism. The polymorphism was also significantly associated with C18:1, but the p value (p = .0162) was higher than the most significant SNP on BTA9, suggesting that it would not be responsible for the QTL. Although further investigation will be needed to determine the responsible gene and polymorphism, our findings would contribute to development of selective markers for fatty acid composition in the Japanese Black cattle of Hyogo.  相似文献
